Mycoplasma pneumoniae is a human pathogen causing atypical community-acquired pneumonia. It is a model for a minimal cell, known for its non-canonical use of surface proteins for host-cell adhesion through ectodomain shedding and antigenic variation to evade the host cell immune response. Mpn444 is an essential mycoplasma surface protein implicated in both processes. It is one of 46 lipoproteins of M. pneumoniae, none of which have been structurally or functionally characterized. Here, we report the structure of Mpn444 at 3.04 Å as well as the molecular architecture of the trimeric Mpn444 complex. Our experimental structure displays striking similarity to structure predictions of several other essential lipoproteins in M. pneumoniae and other related Mycoplasma species, suggesting it to have a specialized and conserved function. The essentiality and involvement of Mpn444 in host immune evasion makes our structure a target for the development of new treatment strategies against mycoplasma infections.

BMPs control postnatal dendrite growth and complexity in sympathetic neurons / von Afsaneh Majdazari

The vertebrate nervous system is a complex network of billions of neurons connected by dendrites and axons, integrated to functional circuits and areas/organs in the central and peripheral nervous system. The cells of the nervous system origin from common progenitors, which take on different cell fates based on intrinsic and extrinsic factors. These factors determine general neuronal traits, but also the morphology and the type of connections made to other cells. Mechanisms underlying axonal and dendritic growth are well described in contrast to the initiation of neurite growth, which remains to be fully elucidated, especially concerning dendrite formation. Recently BMPs have been identified as candidate dendrite inducing factors in sympathetic, cortical and hippocampal neurons. Here we focus on the in vivo role of BMPs on dendrite growth in sympathetic neurons as their development and differentiation processes have been analyzed in detail.
Seit Erlass des Schuldverschreibungsgesetzes von 2009 (SchVG) zeichnet sich ab, dass die Emissionspraxis der deutschen Staatsanleihen im Hinblick auf kollektive Gläubigermechanismen („Collective Action Clauses“ – CACs), wie sie das SchVG erstmals rechtssicher ermöglicht, substantiellen Änderungen unterliegen wird. Während die Haltung inländischer öffentliche Emittenten noch bei Verabschiedung des SchVG reserviert war, hat sich die Situation seit Mai 2010 grundlegend geändert: Verschiedene Mitgliedstaaten der Eurozone sind mit Verbindlichkeiten in ihrer Währung in Schwierigkeiten geraten und mussten um internationale Hilfe nachsuchen. Vor dem Hintergrund der politischen Forderung nach einer Beteiligung der bestehenden Gläubigerschaft („private sector involvement“, „PSI“) musste schließlich Griechenland im Frühjahr 2012 eine Umschuldung (mithilfe entsprechender Klauseln) bemühen. Politische und rechtliche Festlegungen der Eurogruppe über die künftige Dokumentation der (verbrieften) Staatenschuld im Hinblick auf eine potentiell erforderlich werdende Umschuldung folgten. Bei den nationalen Umsetzungsarbeiten sind auch gewisse „Unzulänglichkeiten“ des durch das SchVG geschaffenen gesetzlichen Gestaltungsrahmens aufgegriffen worden.
Die nachfolgende Abhandlung zeichnet im Einzelnen die verschiedenen Entwicklungslinien nach, beginnend (i) mit dem SchVG und der darin zum Ausdruck gekommenen traditionellen Haltung der öffentlichen Hand zu CACs, leitet über (ii) zur GriechenlandUmschuldung („PSI“) und deren rechtlichen Besonderheiten und Ergebnisse unter dem (neuen) Blickwinkel einer Beteiligung privater Gläubiger, soll (iii) die von der Eurogruppe gezogenen Konsequenzen in Gestalt der Empfehlungen des Wirtschafts- und Finanzausschusses für „Umschuldungsklauseln“ der Mitgliedsstaaten der Eurozone im Hinblick auf Art. 12 Abs. 3 des ESM-Vertrags erläutern und schließlich (iv) einen Überblick über die gesetzgeberischen Schritte in Deutschland zur Umsetzung der Empfehlungen liefern.
Device-to-device (D2D) communication is an innovative solution for improving wireless network performance to efficiently handle the ever-increasing mobile data traffic. Communication takes place directly between two devices that are in each other’s transmission range. So far, research has focused on the technical challenges of implementing this technology and assumes a user’s general willingness to participate as forwarder in this technology. However, this simplifying assumption is not realistic, as willingness to participate in D2D communication can vary depending on the user. In this work, we consider the scenario that a user can act as a forwarder for a receiver who is not directly or insufficiently reached by the base station and accordingly has no or poor Internet connection. We take a user-centric approach and investigate the willingness to provide an Internet connection as a forwarder. We are the first to investigate user preferences for D2D communication using a choice-based conjoint analysis. Our results, based on a representative sample of potential users (N=181), show that the social relationship between the potential forwarder and the receiver has the greatest impact on the potential forwarder’s decision to provide an Internet connection to the receiver, accepting sacrifices in terms of additional battery consumption and reduced own service performance. In a detailed segment analysis, we observe significant preference differences depending on smartphone usage behavior and user age. Taking the corresponding preferences into account when matching forwarders and receivers can further increase technology adoption.
Sample-based longitudinal discrete choice experiments: preferences for electric vehicles over time

Discrete choice experiments have emerged as the state-of-the-art method for measuring preferences, but they are mostly used in cross-sectional studies. In seeking to make them applicable for longitudinal studies, our study addresses two common challenges: working with different respondents and handling altering attributes. We propose a sample-based longitudinal discrete choice experiment in combination with a covariate-extended hierarchical Bayes logit estimator that allows one to test the statistical significance of changes. We showcase this method’s use in studies about preferences for electric vehicles over six years and empirically observe that preferences develop in an unpredictable, non-monotonous way. We also find that inspecting only the absolute differences in preferences between samples may result in misleading inferences. Moreover, surveying a new sample produced similar results as asking the same sample of respondents over time. Finally, we experimentally test how adding or removing an attribute affects preferences for the other attributes.
Seit ca. 1973 werden die schriftlichen Staatsexamina in Deutschland in Anlehnung an das amerikanische Prüfungssystem mit Einfachauswahlfragen (auch als MC-Fragen bezeichnet) durchgeführt. Dieser Fragentypus hat sich auch für universitäre Prüfungen durchgesetzt, trotz ungeklärter Validität und Eignung dieses Fragentyps. Das Ziel dieser Arbeit bestand darin, den Einfluss von Formatunterschieden bei inhaltlich identischen Fragen auf das Prüfungsergebnis aufzuzeigen Hierzu wurden klassische MC-Fragen mit Freitextfragen sowie mit Mehrfachauswahlfragen verglichen. Es wurden fachspezifische Klausuren während der Übergangszeit von der 8. ÄAppO von 1989 auf die jetzt gültige 9. ÄAppO von 2002 so konstruiert, dass inhaltsidentische Fragen in einer Version als MC-Fragen, in der anderen Version in anderem Format gestellt wurden; jeweils eine Hälfte der teilnehmenden Studierenden diente als Kontrollgruppe für die Fragestellung in anderem Format. Ausgewertet wurden 11 Klausuren mit jeweils 15 MC-Fragen und 15 alternativen Fragen im Zeitraum SS 2004 bis WS 2004/2005 in den Fächern Augenheilkunde, Dermatologie, Orthopädie, Psychiatrie, Rechtsmedizin und Urologie, mit insgesamt 1219 teilnehmenden Studenten. MC-Fragen (55%) wurden im Mittel häufiger korrekt beantwortet als AW-Fragen (25%) und FT-Fragen (33%). Wenn bei Auswahlfragen Studierende mit vier von fünf zutreffenden Entscheidungen ebenfalls einbezogen werden, kommen etwa 52% der Studierenden auf „richtige“ Antworten. Insbesondere bei Freitextfragen gab es große fragenspezifische Unterschiede, in einigen Fällen wurden Freitextfragen häufiger zutreffend beantwortet als inhaltsidentische MC-Fragen. Diese Unterschiede legen nahe, dass aktives Wissen mit MC-Fragen nicht prüfbar ist. Teilwissen konnte mit Fragen in den klassischen MC-Formaten nicht ermittelt werden. Wie exemplarisch in dieser Arbeit gezeigt wird, werden bei Fragen im MCFormat für die Entscheidung richtig/falsch nicht alle Aussagen berücksichtigt. Ergebnisse aus MC-basierten Prüfungen ergeben einen deutlich höheren Anteil richtiger Antworten, als dem aktuellen Wissen entspricht; häufig scheint die Entscheidung zwischen zwei Alternativen geraten zu werden. Bei einer Umformulierung in Freitextfragen ergeben sich häufig größere fragenspezifische Differenzen, die suggerieren, dass mit MC-Fragen aktives Wissen nicht prüfbar ist. Die vorliegenden Untersuchungen begründen Zweifel an der Eignung von ausschließlich MC-basierten Prüfungen für die Prüfung höherer Wissensebenen, eine Validität MC-basierter Prüfungen für klinisches Alltags- und Basiswissen scheint nicht gegeben zu sein.
Bei den meisten erwachsenen Säugetieren führt ein Herzinfarkt zu Fibrose und Verlust von funktionellem Herzgewebe. Einige Wirbeltiere, wie der Zebrabärbling, besitzen jedoch die bemerkenswerte Fähigkeit, nach einer Schädigung ihres Herzgewebes verlorenes Gewebe zu regenerieren und so schädliche Folgen zu verhindern. Die lokale Immunantwort auf eine Verletzung wird zunehmend als eine wichtige Determinante für das regenerative Potential eines Gewebes gesehen. Das Komplementsystem ist Teil des humoralen Immunsystems. Historisch ist es als eine Sammlung von Protein bekannt, den Komplementkomponenten, die in der Leber synthetisiert werden und im Blutkreislauf zirkulieren. Bei Exposition gegenüber einem Auslöser, wie z. B. einem Pathogen, wird eine Komplementkomponentproteinspaltungskaskade initiiert, die dazu führen kann, dass Immunzellen rekrutiert werden, und, dass die Phagozytose erleichtert, ggf. die Zielzelle lysiert wird. Studien legen nahe, dass das Komplementsystem an zellulären Prozessen beteiligt sei, die für Entwicklungs- und Krankheitsprozesse entscheidend sind, wie etwa Proliferation und Dedifferenzierung. Es gibt Hinweise, dass das Komplementsystem eine Rolle bei Krebserkrankungen und bei regenerativen Prozessen spielen könnte. In verschiedenen Arten wurde eine lokale verletzungsinduzierte Expression von komplementkomponentkodierenden Genen in regenerierendem Gewebe beobachtet.
Einzelne Studien legen nahe, dass Funktionsverlust einzelner Komplementkomponenten regenerative Prozesse beeinträchtigt.
Offene Fragen bleiben jedoch: Ist die lokale Expression von mehreren komplementkomponentkodierenden Genen ein Merkmal von regenerierendem Gewebe, das sie von Geweben unterscheidet, welchem die Fähigkeit zur Regeneration fehlt? Und welche Rolle könnte das Komplementsystem und seine Komponenten während des regenerativen Prozesses spielen? Um diesen Fragen nachzugehen, wurde eine Expressionsanalyse von Zebrabärblingsgewebe nach Verletzung mittels RT-qPCR und in situ Hybridisierung durchgeführt: kardiale Kryoverletzung, Larvenrumpfamputation und Schwanzflossenamputation. Ich beobachtete, dass mehrere komplementkomponentkodierende Gene in diesen Geweben nach Verletzung induziert wurden. Die Interpretation veröffentlichter single cell RNAseq Datensätze legt nahe, dass diese komplementkomponentenkodierenden Gene von verschiedenen Zelltypen exprimiert werden, darunter Immunzellen, Epikardzellen und Fibroblasten. Um transkriptionelle Unterschiede zwischen regenerierendem und nicht regenerierendem Gewebe zu identifizieren, verwendete ich ein nicht regeneratives Zebrabärblingmodell, die il11ra- Mutante. Dieser Mutante fehlt die Fähigkeit, verschiedene Organe zu regenerieren, das ist der Fall beim Herzen, dem larvalen Rumpf, und der Schwanzflosse. Ich stellte fest, dass die Mehrheit der verletzungsinduzierten komplementkomponentkodierenden Gene il11ra nachgeschaltet war. Darüber hinaus zeigten Experimente unter Verwendung chemischer Inhibitoren, dass speziell die Expression der komplementkomponentkodierenden Gene c3a.1,
c4b und c7a im Larvenrumpfamputationsmodell durch den Il11-Stat3-Signalweg moduliert wird.
Zur Klärung der Frage, ob das Komplementsystem und/ oder seine Komponenten eine Rolle während der Regeneration spielen, wurden verschiede Funktionsverlustmodelle generiert und im larvalen Rumpfamputationsmodell auf mögliche Aberrationen getestet. Zum einen generierte ich Überexpressionslinien von endogenen Inhibitoren der Komplementproteinspaltungskaskade. Überexpression eines etablierten Komplementsysteminhibitors rca2.1/ tecrem führte zu einer im Vergleich zu Wildtyp- Geschwistern verringerten Regeneration des larvalen Rumpfs. Zum anderen generierte ich Funktionsverlustmutanten von individuellen Komplementkomponenten durch CRISPR/Cas9 vermittelter Mutagenese, und zwar für masp1, masp2, cfd, c1s, c4b, c5 und c9. Die larvale Rumpfregeneration war in diesen Mutanten unauffällig. Allerdings zeigten c4b Mutanten eine verringerte Kardiomyozytenproliferation und eine differenzielle Expression von einigen Markergenen, einschließlich einer erhöhten Expression von inflammatorischen Zytokinen.
Meine Studien führten zu neuen Einblicken in das Komplementsystem im Kontext der Regeneration. Ich fand heraus, dass mehrere komplementkomponentenkodierenden Gene in regenerierendem Zebrabärblinggewebe exprimiert werden, und zwar im Herzgewebe, im larvalen Rumpf und in der adulten Flosse. Darüber hinaus zeige ich, dass die verletzungsinduzierte Expression von komplementkodierenden Genen in regenerierendem Gewebe dem Regenerationsmasterregulator il11ra nachgeschaltet ist. By the fabrication of periodically arranged nanomagnetic systems it is possible to engineer novel physical properties by realizing artificial lattice geometries that are not accessible via natural crystallization or chemical synthesis. This has been accomplished with great success in two dimensions in the fields of artificial spin ice and magnetic logic devices, to name just two. Although first proposals have been made to advance into three dimensions (3D), established nanofabrication pathways based on electron beam lithography have not been adapted to obtain free-form 3D nanostructures. Here we demonstrate the direct-write fabrication of freestanding ferromagnetic 3D nano-architectures. By employing micro-Hall sensing, we have determined the magnetic stray field generated by our free-form structures in an externally applied magnetic field and we have performed micromagnetic and macro-spin simulations to deduce the spatial magnetization profiles in the structures and analyze their switching behavior. Furthermore we show that the magnetic 3D elements can be combined with other 3D elements of different chemical composition and intrinsic material properties.
Fabrication of three-dimensional (3D) nanoarchitectures by focused electron beam induced deposition (FEBID) has matured to a level that highly complex and functional deposits are becoming available for nanomagnetics and plasmonics. However, the generation of suitable pattern files that control the electron beam’s movement, and thereby reliably map the desired target 3D structure from a purely geometrical description to a shape-conforming 3D deposit, is nontrivial. To address this issue we developed several writing strategies and associated algorithms implemented in C++. Our pattern file generator handles different proximity effects and corrects for height-dependent precursor coverage. Several examples of successful 3D nanoarchitectures using different precursors are presented that validate the effectiveness of the implementation.
In times of a growing world population and the associated demand for high crop yield, the understanding and improvement of plant reproduction is of central importance. One key step of plant reproduction is the development of the male gametophyte, which is better known as pollen. In addition, the development of pollen was shown to be very sensitive to abiotic stresses, such as heat, which can cause crop damage and yield loss. To obtain new insights in the development and heat stress response of pollen, a combined transcriptome and proteome analysis was performed for three pollen developmental stages of non- and heat-stressed tomato plants.
The analysis of the transcriptomes of non-stressed pollen developmental stages enabled the determination of mRNAs accumulated in certain developmental stages. The functional analysis of these mRNAs led to the identification of protein families and functional processes that are important at different times of pollen development. A subsequent comparison of the transcriptomes of non- and heat-stressed pollen revealed a core set of 49 mRNAs, which are upregulated in all three developmental stages. The encoded proteins include among other things different heat stress transcription factors and heat shock proteins, which are known key players of the plant heat stress response.
Furthermore, 793 potential miRNAs could be identified in the transcriptome of non- and heat-stressed pollen. Interestingly, 38 out of the 793 miRNAs have already been identified in plants. For more than half of these miRNAs potential target mRNAs were identified and the interactions between miRNAs and mRNAs linked to the development and heat stress response of pollen. In total, 207 developmentally relevant interactions could be determined, out of which 34 have an effect on transcriptional-networks. In addition, 24 of the interactions contribute the heat stress response of pollen, whereby this mainly affects post-meiotic pollen.
An initial correlation of the proteome and transcriptome of the developmental stages revealed that transcriptome analyses are not sufficient to draw exact conclusions about the state of the proteome. A closer look on the relationship of the transcriptome and proteome during pollen development revealed two translational modes that are active during the development of pollen. One mode leads to a direct translation of mRNAs, while the second mode leads a delayed translation at a later point in time. Regarding the delayed translation, it could be shown that this is likely due to a short-term storage of mRNAs in so-called EPPs. The comparison of the proteome and transcriptome response to heat stress revealed that the proteome reacts much stronger and that the reaction is mainly independent from the transcriptome. Finally, the comparison of the proteome of non- and heat-stressed pollen provided first indications for changes in the ribosome composition in response to heat stress, as 57 ribosomal proteins are differentially regulated in at least one developmental stage.
Improved risk stratification in prevention by use of a panel of selected circulating microRNAs

Risk stratification is crucial in prevention. Circulating microRNAs have been proposed as biomarkers in cardiovascular disease. Here a miR panel consisting of miRs related to different cardiovascular pathophysiologies, was evaluated to predict outcome in the context of prevention. MiR-34a, miR-223, miR-378, miR-499 and miR-133 were determined from peripheral blood by qPCR and combined to a risk panel. As derivation cohort, 178 individuals of the DETECT study, and as validation cohort, 129 individuals of the SHIP study were used in a case-control approach. Overall mortality and cardiovascular events were outcome measures. The Framingham Risk Score(FRS) and the SCORE system were applied as risk classification systems. The identified miR panel was significantly associated with mortality given by a hazard ratio(HR) of 3.0 (95% (CI): 1.09–8.43; p = 0.034) and of 2.9 (95% CI: 1.32–6.33; p = 0.008) after adjusting for the FRS in the derivation cohort. In a validation cohort the miR-panel had a HR of 1.31 (95% CI: 1.03–1.66; p = 0.03) and of 1.29 (95% CI: 1.02–1.64; p = 0.03) in a FRS/SCORE adjusted-model. A FRS/SCORE risk model was significantly improved to predict mortality by the miR panel with continuous net reclassification index of 0.42/0.49 (p = 0.014/0.005). The present miR panel of 5 circulating miRs is able to improve risk stratification in prevention with respect to mortality beyond the FRS or SCORE.
Eurozentrismus heißt das Schlagwort, welches die Wissenschaft der Völkerrechtsgeschichte seit einigen Jahren – wieder einmal, muss man sagen, blickt man auf die ersten kritischen Stimmen in den 1960er-Jahren zurück – beschäftigt. Im Zentrum steht dabei die Kritik an der klassischen Historiographie des Völkerrechts und insbesondere der führenden Rolle, die dem sogenannten Westen bzw. Europa darin zugeschrieben wird. Nach der traditionellen Narration ist das moderne Völkerrecht das Ergebnis einer europäischen Fortschrittsgeschichte, die ihren Ausgangspunkt im europäischen Mittealter nahm und sich im Laufe des 19. Jahrhunderts zu einem universell gültigen internationalen Rechtssystem herausbildete. Nicht-europäische Staaten spielen in dieser linearen und eurozentrischen Entwicklungsgeschichte, wenn überhaupt, nur eine passive Rolle. Nicht als Akteure, sondern als Rezipienten werden sie im Laufe des 19. Jahrhunderts Teil dieser europäischen Meistererzählung. "[I]t was in fact Europe and not America, Asia, or Africa that dominated and, in so doing, unified the world, it is not our perspective but the historical record itself that can be called Eurocentric", heißt es 1984 bei Hedley Bull und Adam Watson. Trotz verschiedener kritischer Gegenerzählungen ist diese klassische Narration auch heute noch prägend für das Selbstverständnis der Wissenschaft der Völkerrechtsgeschichte. ...
Background: Elucidating the genomic basis of adaptation and speciation is a major challenge in natural systems with large quantities of environmental and phenotypic data, mostly because of the scarcity of genomic resources for non-model organisms. The Atlantic molly (Poecilia mexicana, Poeciliidae) is a small livebearing fish that has been extensively studied for evolutionary ecology research, particularly because this species has repeatedly colonized extreme environments in the form of caves and toxic hydrogen sulfide containing springs. In such extreme environments, populations show strong patterns of adaptive trait divergence and the emergence of reproductive isolation. Here, we used RNA-sequencing to assemble and annotate the first transcriptome of P. mexicana to facilitate ecological genomics studies in the future and aid the identification of genes underlying adaptation and speciation in the system.
Description: We provide the first annotated reference transcriptome of P. mexicana. Our transcriptome shows high congruence with other published fish transcriptomes, including that of the guppy, medaka, zebrafish, and stickleback. Transcriptome annotation uncovered the presence of candidate genes relevant in the study of adaptation to extreme environments. We describe general and oxidative stress response genes as well as genes involved in pathways induced by hypoxia or involved in sulfide metabolism. To facilitate future comparative analyses, we also conducted quantitative comparisons between P. mexicana from different river drainages. 106,524 single nucleotide polymorphisms were detected in our dataset, including potential markers that are putatively fixed across drainages. Furthermore, specimens from different drainages exhibited some consistent differences in gene regulation.
Conclusions: Our study provides a valuable genomic resource to study the molecular underpinnings of adaptation to extreme environments in replicated sulfide spring and cave environments. In addition, this study adds to the increasing number of genomic resources in the family Poeciliidae, which are widely used in comparative analyses of behavior, ecology, evolution, and medical genetics.
Die ursprünglich über zwei Meter hohe Inschrift von Paros, im Folgenden hier gemäß der gängigen Konvention als Marmor Parium bezeichnet, stellt einen faszinierenden Text dar, der in den Altertumswissenschaften jedoch kaum größere Beachtung erfährt. Containment problems belong to the classical problems of (convex) geometry. In the proper sense, a containment problem is the task to decide the set-theoretic inclusion of two given sets, which is hard from both the theoretical and the practical perspective. In a broader sense, this includes, e.g., radii or packing problems, which are even harder. For some classes of convex sets there has been strong interest in containment problems. This includes containment problems of polyhedra and balls, and containment of polyhedra, which have been studied in the late 20th century because of their inherent relevance in linear programming and combinatorics.
Since then, there has only been limited progress in understanding containment problems of that type. In recent years, containment problems for spectrahedra, which naturally generalize the class of polyhedra, have seen great interest. This interest is particularly driven by the intrinsic relevance of spectrahedra and their projections in polynomial optimization and convex algebraic geometry. Except for the treatment of special classes or situations, there has been no overall treatment of that kind of problems, though.
In this thesis, we provide a comprehensive treatment of containment problems concerning polyhedra, spectrahedra, and their projections from the viewpoint of low-degree semialgebraic problems and study algebraic certificates for containment. This leads to a new and systematic access to studying containment problems of (projections of) polyhedra and spectrahedra, and provides several new and partially unexpected results.
The main idea - which is meanwhile common in polynomial optimization, but whose understanding of the particular potential on low-degree geometric problems is still a major challenge - can be explained as follows. One point of view towards linear programming is as an application of Farkas' Lemma which characterizes the (non-)solvability of a system of linear inequalities. The affine form of Farkas' Lemma characterizes linear polynomials which are nonnegative on a given polyhedron. By omitting the linearity condition, one gets a polynomial nonnegativity question on a semialgebraic set, leading to so-called Positivstellensaetze (or, more precisely Nichtnegativstellensaetze). A Positivstellensatz provides a certificate for the positivity of a polynomial function in terms of a polynomial identity. As in the linear case, these Positivstellensaetze are the foundation of polynomial optimization and relaxation methods. The transition from positivity to nonnegativity is still a major challenge in real algebraic geometry and polynomial optimization.
With this in mind, several principal questions arise in the context of containment problems: Can the particular containment problem be formulated as a polynomial nonnegativity (or, feasibility) problem in a sophisticated way? If so, how are positivity and nonnegativity related to the containment question in the sense of their geometric meaning? Is there a sophisticated Positivstellensatz for the particular situation, yielding certificates for containment? Concerning the degree of the semialgebraic certificates, which degree is necessary, which degree is sufficient to decide containment?
Indeed, (almost) all containment problems studied in this thesis can be formulated as polynomial nonnegativity problems allowing the application of semialgebraic relaxations. Other than this general result, the answer to all the other questions (highly) depends on the specific containment problem, particularly with regard to its underlying geometry. An important point is whether the hierarchies coming from increasing the degree in the polynomial relaxations always decide containment in finitely many steps.
We focus on the containment problem of an H-polytope in a V-polytope and of a spectrahedron in a spectrahedron. Moreover, we address containment problems concerning projections of H-polyhedra and spectrahedra. This selection is justified by the fact that the mentioned containment problems are computationally hard and their geometry is not well understood.

The Arp2/3 complex nucleates and cross-links actin filaments at the leading edge of motile cells, and its activity is stimulated by C-terminal regions of WASP/Scar proteins, called VCA domains. VCA domains contain a verprolin homology sequence (V) that binds monomeric actin and central (C) and acidic sequences (A) that bind the Arp2/3 complex. Here we show that the C domain binds to monomeric actin with higher affinity (K(d) = 10 microm) than to the Arp2/3 complex (K(d) > 200 microm). Nuclear magnetic resonance spectroscopy reveals that actin binds to the N-terminal half of the C domain and that both the V and C domains can bind actin independently and simultaneously, indicating that they interact with different sites. Mutation of conserved hydrophobic residues in the actin-binding interface of the C domain disrupts activation of the Arp2/3 complex but does not alter affinity for the complex. By chemical cross-linking the C domain interacts with the p40 subunit of the Arp2/3 complex and, by fluorescence polarization anisotropy, the binding of actin and the Arp2/3 complex are mutually exclusive. Our results indicate that both actin and Arp2/3 binding are important for C domain function but that the C domain does not form a static bridge between the two. We propose a model for activation of the Arp2/3 complex in which the C domain first primes the complex by inducing a necessary conformational change and then initiates nucleus assembly by bringing an actin monomer into proximity of the primed complex.

Ziel des Autors ist es, die monumentalen Felsfassadengräber Etruriens in einem größeren Kontext darzustellen, was im Untertitel "unter besonderer Berücksichtigung der etruskischen Felsgräbernekropolen" zum Ausdruck kommt. Mit 162 überwiegend sehr qualitätvollen Abbildungen auf 144 Seiten fügt sich der Band in die bekannte Reihe, die archäologische Denkmäler für ein breites, Kultur interessiertes Publikum erschließt. Der Autor gehört zu den Kennern dieser etruskischen Gräber und zieht im Vorwort die Verbindungslinien zu seiner wissenschaftlichen Tätigkeit. In 14 Kapiteln spannt der Verfasser einen weiten Bogen (S. 7-86) mit Schlaglichtern auf Felsgräber und Felsarchitektur außerhalb Südetruriens; letzteren ist ein ausführliches Kapitel gewidmet (S. 87-128). Ein Anhang mit einer nützlichen Zusammenstellung der wichtigsten Informationen zu den südetruskischen Felsgräbernekropolen (S. 131-137) und einem Glossar beschließen den Bildband. Die Bibliographie spiegelt in ihrer Auswahl die Schwerpunktsetzung des Bandes. Disturbances in lipid metabolism are responsible for many chronic disorders, such as type 2 diabetes and atherosclerosis. Regulation of lipid metabolism occurs by activated transcription factors peroxisome proliferator-activated receptor δ (PPARδ) and liver X receptor α (LXRα) mediating transcription of different target genes involved in regulation of fatty acid uptake and oxidation or cellular cholesterol homeostasis. This is especially relevant for the macrophages, since pathways regulated by PPARδ and LXRα affect foam cell formation, a process driving the progression of atherosclerotic lesion. AMP-activated protein kinase (AMPK) plays a central role in energy homeostasis in every type of eukaryotic cell, but its role in human macrophages, particularly with regard to lipid metabolism, is not precisely defined yet. Thus, I investigated the impact of AMPK activity on PPARδ and LXRα and the expression of their target genes involved in fatty acid oxidation (FAO) and cholesterol metabolism.
As PPARδ has been described as a potential target for prevention and treatment of several disorders and AMPK as interesting drug target for diabetes and metabolic syndrome, the aim of the first part of my studies was to investigate their interaction in primary human macrophages. Completing the first challenge successfully, I was able to establish a lentiviral transduction system for constitutively active AMPK (consisting of a truncated catalytic AMPKα1 subunit bearing an activating T198D mutation) in primary human macrophages.
Using genome-wide microarray analysis of gene expression, I demonstrate FAO as the strongest affected pathway during combined AMPKα1 overexpression and PPARδ activation.
The most influenced genes were validated by quantitative PCR as well as by Western analysis. I found that AMPK increases the expression of FAO-associated genes targeted by PPARδ. Corroborating the results obtained using AMPKα1 overexpression, PPARδ target gene expression was increased not only by PPARδ agonist GW501516, but also by pharmacological allosteric AMPK activator A-769662. Additional enhancement of target gene mRNA expression was achieved upon co-activation of PPARδ and AMPK. Silencing PPARδ expression increased basal expression of target genes, confirming the repressive nature of ligand-free PPARδ, abolishing the increased target gene expression upon AMPK or PPARδ activation. Measurements of triglyceride contents of human macrophages incubated with VLDL following PPARδ activation demonstrated a reduction of intracellular triglyceride accumulation in cells, which may reflect the enhancement of fat catabolism.
In the second part of my studies, I concentrated on the regulation of cholesterol transporter ATP-binding cassette transporter A1 (ABCA1) expression by AMPK. ABCA1 facilitates
cholesterol efflux from macrophages thus, preventing atherosclerosis progression. For the first time, AMPK implication in the regulation of the ABCA1 pathway could be presented. Both AMPK overexpression and activation lead to significantly increased ABCA1 expression, whereas AMPKα1 knock-down strongly reduced this effect. Besides, I was able to prove an enhanced activity of ABCA1 during AMPK activation in human THP-1 macrophages by measuring cholesterol efflux into apolipoprotein AI-containing medium.
Previous findings showed regulation of ABCA1 by LXRα. I confirmed these results by silencing experiments indicating an essential role of LXRα in ABCA1 regulation pathway.
Here, ABCA1 mRNA as well as protein expression were positively mediated by LXRα. LXRα activation elevated ABCA1 levels, whereas its silencing down-regulated this effect.
Interestingly, ABCA1 was found to be regulated only by LXRα and not through LXRα. At the same time, knock-down of PPARδ, -γ or -δ, which may be also involved in the regulation of LXR/ABCA1 axis, did not influence the activation of ABCA1 expression by an AMPK activator. To confirm that LXRE on Abca1 promoter is essential for ABCA1 regulation, I performed luciferase reporter assay using constructs based on Abca1 promoter with or without LXRE mutation. Mutation of LXRE abolished reporter activity, whereas AMPK activation increased luciferase activity of wild-type LXRE construct. Furthermore, I demonstrate AMPK-dependent LXRα binding to the LXRE site of Abca1 promoter using the method of chromatin immunoprecipitation. AMPK activation significantly increased, whereas silencing of AMPK significantly attenuated LXRα binding, indicating AMPK as one of the most important regulators of ABCA1 expression.
In summary, I provided an evidence for AMPK involvement into lipid and cholesterol metabolism in human macrophages showing the regulation of PPARδ and LXRα target genes. The understanding of AMPK and PPARδ interaction allows the development of new approaches for treatment of metabolic syndrome and related diseases. Increased FAO during the activation of both proteins may exhibit better therapeutic benefit. On the other hand, I have shown the impact of AMPK activation on ABCA1 via LXRα up-regulation leading to increased cholesterol efflux in human macrophages for the first time. These findings thus may impact future improving of anti-atherosclerosis therapies.
AMP-activated protein kinase (AMPK) maintains energy homeostasis by suppressing cellular ATP-consuming processes and activating catabolic, ATP-producing pathways such as fatty acid oxidation (FAO). The transcription factor peroxisome proliferator-activated receptor δ (PPARδ) also affects fatty acid metabolism, stimulating the expression of genes involved in FAO. To question the interplay of AMPK and PPARδ in human macrophages we transduced primary human macrophages with lentiviral particles encoding for the constitutively active AMPKα1 catalytic subunit, followed by microarray expression analysis after treatment with the PPARδ agonist GW501516. Microarray analysis showed that co-activation of AMPK and PPARδ increased expression of FAO genes, which were validated by quantitative PCR. Induction of these FAO-associated genes was also observed upon infecting macrophages with an adenovirus coding for AMPKγ1 regulatory subunit carrying an activating R70Q mutation. The pharmacological AMPK activator A-769662 increased expression of several FAO genes in a PPARδ- and AMPK-dependent manner. Although GW501516 significantly increased FAO and reduced the triglyceride amount in very low density lipoproteins (VLDL)-loaded foam cells, AMPK activation failed to potentiate this effect, suggesting that increased expression of fatty acid catabolic genes alone may be not sufficient to prevent macrophage lipid overload.

The volume under review contains the published proceedings of a conference held in 2009 with the challenging title, "Merowingische Monetarmünzen und der Beginn des Mittelalters". These Merovingian "Monetarmünzen" are a distinctive group of coins of which less than 10 000 are currently known. Quite suddenly, in the late sixth century, this type of gold coinage appears, with the name of a moneyer ( monetarius ) on the obverse and the place name on the reverse (presumably, but not necessarily in all instances, the mint). Thus, over a thousand moneyers and 722 place names are recorded, many only attested once or twice. In the late 7 th c. these coins slowly give way to a system based on the silver penny/denier, no longer showing names of moneyers. Who were these moneyers? What was their relationship with the court and the kings? To what ends were those coins produced, and how were they used in daily commerce? Why are so many different mints attested? These questions have occupied scholars for several generations now. However, Jarnut and Strothmann have added a new perspective: in how far are these coinages and the associated monetary policy a continuation of late Roman practices, or do they represent something altogether different and can, therefore, be understood as an expression of a fundamentally altered society that could be termed medieval? ...

Mit Blick auf das Ganze, das Allgemeine, wird oft das Typische, das Exemplarische bemüht, das man in Einzel- oder Fallstudien einzufangen glaubt. Hinge die Relevanz lokaler Studien von der Prämisse ab, die große Welt im Kleinen finden zu müssen, hätten sie schlechterdings keinerlei Relevanz. So zumindest urteilt Clifford Geertz über entsprechende Studien in der Ethnologie nach dem Modell "Jonesville-ist-die-USA". Der Versuchung eines solchen Modells ist Francisca Loetz in ihrer Heidelberger Habilitationsschrift über frühneuzeitliche Blasphemie am Beispiel Zürcher Gotteslästerer erlegen. In einer Langzeitstudie vom späten 15. bis zum frühen 18. Jahrhundert untersucht Loetz rund 900 dokumentierte Fälle der Wortsünde des Fluchens, Schwörens und Schmähens bei "Gotz schedel", "Gotz bart" oder "Gotz nasa". Delikte, bezüglich derer man im 16. Jahrhundert empfahl, den Angeklagten ein Schloss für ihre Zungen zu schlagen. Die Ergebnisse dieser Studie erfüllen jedoch nicht den erhobenen Anspruch der Repräsentativität für das Westeuropa der frühen Neuzeit. Der Beitrag analysiert Ungleichheitseffekte des 2007 eingeführten Elterngelds. Wir zeigen, dass die familienpolitische Einführung der Ressource Elterngeld die Einkommensungleichheiten der Produktions- bzw. Erwerbssphäre auf die Reproduktions- bzw. Familiensphäre übertragen hat. Das Elterngeld trägt damit aber zumindest bislang nicht (wie angedacht) zur Aufhebung der asymmetrischen Aufteilung von (entlohnter) Erwerbsarbeit und (nicht-entlohnter) Sorgearbeit zwischen Elternteilen bei. Stattdessen verdeutlicht unsere räumlich orientierte Untersuchung des Elterngeldbezugs ungleiche Muster in den Bewältigungsmöglichkeiten kinderbezogener Sorgearbeiten. Die an der ungleichen Geographie des Elterngelds deutlich werdende Ausdifferenzierung von Bearbeitungschancen von Elternschaft interpretieren wir als Ausdruck von sozialen Spaltungstendenzen auf dem Gebiet der Reproduktion, die von der familienpolitischen Einführung des Elterngelds forciert worden sind.
The central goal of this investigation is to describe the dynamic reaction of a multicellular tumour spheroid to treatment with radiotherapy. A focus will be on the triggered dynamic cell cycle reaction in the spheroid and how it can be employed within fractionated radiation schedules.
An agent-based model for cancer cells is employed which features inherent cell cycle progression and reactions to environmental conditions. Cells are represented spatially by a weighted, dynamic and kinetic Voronoi/Delaunay model which also provides for the identification of cells in contact within the multicellular aggregate. Force-based interaction between cells will lead to rearrangement in response to proliferation and can induce cell quiescence via a mechanism of pressure-induced contact inhibition. The evolution of glucose and oxygen concentration inside the tumour spheroid is tracked in a diffusion solver in correspondence to in vitro or in vivo boundary conditions and a corresponding local nutrient uptake by single cells.
Radiation effects are implemented based on the measured single cell survival in the linear-quadratic model. The survival probability will be affected by the radiosensitivity of the current cycle phase and the local oxygen concentration. Quiescent cells will reduce the effective dose they receive as a consequence of their increased radioresistance. The radiation model includes a fast response to fatal DNA damage through cell apoptosis and a slow response via cell loss due to misrepair during the radiation-induced G2-block.
A simplified model for drug delivery in chemotherapy is implemented.
The model can describe the growth dynamics of spheroids in accordance to experimental data, including total number of cells, histological structure and cell cycle distribution. Investigations of possible mechanisms for growth saturation reveal a critical dependence of tumour growth on the shedding rate of cells from the surface.
In response to a dose of irradiation, a synchronisation of the cell cycle progression within the tumour is observed. This will lead to cyclic changes in the overall radiation sensitivity of the tumour which are quantified using an enhancement measure in comparison to the expected radiosensitivity of he tumour. A transient strong peak in radiosensitivity enhancement is observed after administration of irradiation. Mechanisms which influence the peak timing and development are systematically investigated, revealing quiescence and reactivation of cells to be a central mechanism for the enhancement.
Direct redistribution of cells due to different survival in cell cycle phases, re-activation of quiescent cells in response to radiation-induced cell death and blocking of DNA damaged cells at the G2/M checkpoint are identified as the main mechanisms which contribute to a synchronisation and determine the radiosensitivity increase. A typical time scale for the development of radiosensitivity and the relaxation of tumours to a steady-state after irradiation is identified, which is related to the typical total cell cycle time.
A range of clinical radiotherapy schedules is tested for their performance within the simulation and a systematic comparison with alternative delivery schedules is performed, in order to identify schedules which can most effectively employ the described transient enhancement effects. In response to high-dose schedules, a dissolution of the tumour spheroid into smaller aggregates can be observed which is a result of the loss of integrity in the spheroid that is associated with high cell death via apoptosis. Fractionated irradiation of spheroids with constant dose per time unit but different inter-fraction times clearly reveals optimal time-intervals for radiation, which are directly related to the enhancement response of the tumour.
In order to test the use of triggered enhancement effects in tumours, combinations of trigger- and effector doses are examined for their performance in specific treatment regimens. Furthermore, the automatic identification and triggering in response to high enhancement periods in the tumour is analysed.
While triggered schedules and automatic schedules both yield a higher treatment efficiency in comparison to conventional schedules, treatment optimisation is a revealed to be a global problem, which cannot be sufficiently solved using local optimisation only.
The spatio-temporal dynamics of hypoxia in the tumour are studied in response to irradiation. Microscopic, diffusion-induced reoxygenation dynamics are demonstrated to be on a typical time-scale which is in the order of fractionation intervals. Neoadjuvant chemotherapy with hydroxyurea can yield a drastic improvement of radiosensitivity via cell cycle synchronisation and specific toxicity against radioresistant S-phase cells.
The model makes clear predictions of radiation schedules which are especially effective as a result of triggered cell cycle-based radiosensitivity enhancement. Division of radiation into trigger and effector doses is highly effective and especially suited to be combined with adjuvant chemotherapy in order to limit regrowth of cells.
Tumour hypoxia plays a pivotal role in cancer therapy for most therapeutic approaches from radiotherapy to immunotherapy. The detailed and accurate knowledge of the oxygen distribution in a tumour is necessary in order to determine the right treatment strategy. Still, due to the limited spatial and temporal resolution of imaging methods as well as lacking fundamental understanding of internal oxygenation dynamics in tumours, the precise oxygen distribution map is rarely available for treatment planing. We employ an agent-based in silico tumour spheroid model in order to study the complex, localized and fast oxygen dynamics in tumour micro-regions which are induced by radiotherapy. A lattice-free, 3D, agent-based approach for cell representation is coupled with a high-resolution diffusion solver that includes a tissue density-dependent diffusion coefficient. This allows us to assess the space- and time-resolved reoxygenation response of a small subvolume of tumour tissue in response to radiotherapy. In response to irradiation the tumour nodule exhibits characteristic reoxygenation and re-depletion dynamics which we resolve with high spatio-temporal resolution. The reoxygenation follows specific timings, which should be respected in treatment in order to maximise the use of the oxygen enhancement effects. Oxygen dynamics within the tumour create windows of opportunity for the use of adjuvant chemotherapeutica and hypoxia-activated drugs. Overall, we show that by using modelling it is possible to follow the oxygenation dynamics beyond common resolution limits and predict beneficial strategies for therapy and in vitro verification. Models of cell cycle and oxygen dynamics in tumours should in the future be combined with imaging techniques, to allow for a systematic experimental study of possible improved schedules and to ultimately extend the reach of oxygenation monitoring available in clinical treatment.
Tumour cells show a varying susceptibility to radiation damage as a function of the current cell cycle phase. While this sensitivity is averaged out in an unperturbed tumour due to unsynchronised cell cycle progression, external stimuli such as radiation or drug doses can induce a resynchronisation of the cell cycle and consequently induce a collective development of radiosensitivity in tumours. Although this effect has been regularly described in experiments it is currently not exploited in clinical practice and thus a large potential for optimisation is missed. We present an agent-based model for three-dimensional tumour spheroid growth which has been combined with an irradiation damage and kinetics model. We predict the dynamic response of the overall tumour radiosensitivity to delivered radiation doses and describe corresponding time windows of increased or decreased radiation sensitivity. The degree of cell cycle resynchronisation in response to radiation delivery was identified as a main determinant of the transient periods of low and high radiosensitivity enhancement. A range of selected clinical fractionation schemes is examined and new triggered schedules are tested which aim to maximise the effect of the radiation-induced sensitivity enhancement. We find that the cell cycle resynchronisation can yield a strong increase in therapy effectiveness, if employed correctly. While the individual timing of sensitive periods will depend on the exact cell and radiation types, enhancement is a universal effect which is present in every tumour and accordingly should be the target of experimental investigation. Experimental observables which can be assessed non-invasively and with high spatio-temporal resolution have to be connected to the radiosensitivity enhancement in order to allow for a possible tumour-specific design of highly efficient treatment schedules based on induced cell cycle synchronisation.
Author Summary: The sensitivity of a cell to a dose of radiation is largely affected by its current position within the cell cycle. While under normal circumstances progression through the cell cycle will be asynchronous in a tumour mass, external influences such as chemo- or radiotherapy can induce a synchronisation. Such a common progression of the inner clock of the cancer cells results in the critical dependence on the effectiveness of any drug or radiation dose on a suitable timing for its administration. We analyse the exact evolution of the radiosensitivity of a sample tumour spheroid in a computer model, which enables us to predict time windows of decreased or increased radiosensitivity. Fractionated radiotherapy schedules can be tailored in order to avoid periods of high resistance and exploit the induced radiosensitivity for an increase in therapy efficiency. We show that the cell cycle effects can drastically alter the outcome of fractionated irradiation schedules in a spheroid cell system. By using the correct observables and continuous monitoring, the cell cycle sensitivity effects have the potential to be integrated into treatment planing of the future and thus to be employed for a better outcome in clinical cancer therapies.
Lifestyle interventions, including meal replacement, are effective in the prevention and treatment of type-2-diabetes and obesity. Since insulin is the key weight regulator, we hypothesised that the addition of meal replacement to a lifestyle intervention reduces insulin levels more effectively than lifestyle intervention alone. In the international multicentre randomised controlled ACOORH (Almased Concept against Overweight and Obesity and Related Health Risk) trial, overweight or obese persons who meet the criteria for metabolic syndrome (n = 463) were randomised into two groups. Both groups received nutritional advice focusing on carbohydrate restriction and the use of telemonitoring devices. The intervention group substituted all three main meals per day in week 1, two meals per day in weeks 2–4, and one meal per day in weeks 5–26 with a protein-rich, low-glycaemic meal replacement. Data were collected at baseline and after 1, 3, 6 and 12 months. All datasets providing insulin data (n = 446) were included in this predefined subanalysis. Significantly higher reductions in insulin (−3.3 ± 8.7 µU/mL vs. −1.6 ± 9.8 µU/mL), weight (−6.1 ± 5.2 kg vs. −3.2 ± 4.6 kg), and inflammation markers were observed in the intervention group. Insulin reduction correlated with weight reduction and the highest amount of weight loss (−7.6 ± 4.9 kg) was observed in those participants with an insulin decrease > 2 µU/mL. These results underline the potential for meal replacement-based lifestyle interventions in diabetes prevention, and measurement of insulin levels may serve as an indicator for adherence to carbohydrate restriction.

Since leptin is involved in weight regulation, we hypothesised that a meal replacement-based lifestyle intervention would reduce leptin levels more effectively than lifestyle intervention alone. In the international, multicentre, randomised-controlled ACOORH-trial (Almased-Concept-against-Overweight-and-Obesity-and-Related- Health-Risk), overweight or obese participants with metabolic syndrome criteria (n = 463) were randomised into two groups and received telemonitoring devices and nutritional advice. The intervention group additionally used a protein-rich, low-glycaemic meal replacement. Data were collected at baseline, after 1, 3, 6, and 12 months. All datasets providing leptin data (n = 427) were included in this predefined subanalysis. Serum leptin levels significantly correlated with sex, body mass index, weight, and fat mass at baseline (p < 0.0001). Stronger leptin reduction has been observed in the intervention compared to the control group with the lowest levels after 1 month of intervention (estimated treatment difference −3.4 µg/L [1.4; 5.4] for females; −2.2 µg/L [1.2; 3.3] for males; p < 0.001 each) and was predictive for stronger reduction of body weight and fat mass (p < 0.001 each) over 12 months. Strongest weight loss was observed after 6 months (−5.9 ± 5.1 kg in females of the intervention group vs. −2.9 ± 4.9 kg in the control group (p < 0.0001); −6.8 ± 5.3 kg vs. −4.1 ± 4.4 kg (p = 0.003) in males) and in those participants with combined leptin and insulin decrease. A meal replacement-based lifestyle intervention effectively reduces leptin which is predictive for long-term weight loss.
Sowohl die Gifte der Kegel- (Conidae) als auch die der Schraubenschnecken (Terebridae) enthalten eine Vielzahl pharmakologisch aktiver Peptide. Vor allem die Conopeptide bzw. Conotoxine aus den Giften der Kegelschnecken werden aufgrund ihrer Selektivität für Ionenkanäle und Rezeptoren seit langem als Werkzeuge in der neuropharmakologischen Forschung eingesetzt. Hier rücken gerade neuronale nikotinische Acetylcholinrezeptoren immer mehr in den Fokus der medizinischen Forschung, da sie vermutlich an der Entwicklung neurodegenerativer Erkrankungen wie Alzheimer, Parkinson, Demenz, Schizophrenie und Epilepsie beteiligt sind. Ziel dieser Dissertation war es daher, neue Inhibitoren in den Giften Kegelschnecken (Conidae) und der Schraubenschnecken (Terebridae) für nikotinische Acetylcholinrezeptoren, vor allem der neuronalen Subtypen, zu identifizieren. Es erfolgte die:
1. Identifizierung neuer αD-Conotoxine
Aus den Giften von Conus capitaneus und C. mustelinus konnten zwei native αDConotoxine (αD-CAP und αD-MUS) isoliert und charakterisiert werden. Beide Toxine sind Homodimere mit Molekulargewichten von 11 kDa und inhibieren nikotinische ACh-Rezeptoren. Sie blockieren die Subtypen α7>α3β2>α4β2, wobei sich αD-MUS als potenter als αD-CAP erweist (IC50-Werte von αD-MUS: α7 0,12 nM, α3β2 1,08 nM, α4β2 4,5 nM; IC50-Werte von αD-CAP α7 0,25 nM, α3β2 2,8 nM, α4β2 28,6 nM). Hingegen haben die αD-Conotoxine auf die Rezeptorsubtypen α3β4, α4β4 und α1β1γδ keinen hemmenden Einfluss. Zusätzlich konnten drei weitere αD-Conotoxine mit Hilfe der cDNA von C. vexillum und C. betulinus identifiziert werden. Eine Besonderheit hierbei war, dass innerhalb der Familie der αD-Conotoxine zwei unterschiedliche Signalsequenzen vorkommen und somit diese Sequenzen nicht stark konserviert sind.
2. Charakterisierung des α-Conotoxins SI aus dem Gift von C. striatus
Im Gift der Kegelschnecke Conus striatus wurde ein Peptid mit inhibierender Wirkung an α7-Rezeptoren nachgewiesen. Molekulare Masse (1.352,5 Da) und Aminosäuresequenz entsprachen dem α-Conotoxin SI, das als Antagonist muskulärer nACh-Rezeptoren bekannt ist. Da die Ergebnisse mehrere Jahre zurück lagen und bisher keine Analysen im Oozytenexpressions-System durchgeführt wurden, wurdeeine mögliche Aktivität sowohl an neuronalen als auch an muskulären nACh-Rezeptoren vermutet. Voltage Clamp-Messungen bestätigten die spezifische Wirkung am Muskeltyp, wodurch die Aktivität am α7-Rezeptorsubtyp einem anderen Conopeptid, zugewiesen werden muss, das als Beiprodukt isoliert wurde.
3. Identifizierung neuer Conotoxine der A-Superfamilie Mit molekularbiologischen Methoden unter Nutzung von cDNA-Bibliotheken gelang es, 27 Conotoxine (17 neue und 10 bekannte) aus der A-Superfamilie zu identifizieren: drei α- und zwei κA-Conotoxine aus Conus striatus, zwei α-Conotoxine aus C. betulinus, zwei α- und zwei κA-Conotoxine aus C. carinatus, drei α-Conotoxine aus C. catus, drei α- und zwei κA-Conotoxine aus C. circumcisus, ein α-Conotoxin aus. C. geographus, zwei aus C. imperialis, jeweils eines aus C. lividus, C. quercinus, C. sponsalis sowie zwei aus C. terebra Die Vielzahl der identifizierten α-Conotoxine belegt die hohe Diversität dieser Toxine in den Giften der Kegelschecken. Anhand von Vergleichen mit bereits bekannten Toxinen werden die möglichen Wirkungsweisen einiger neuer α-Conotoxine diskutiert. Für einen Teil der α-Conotoxine wurden 3D-Strukturmodelle erstellt, die Einblicke in die Bindung der Toxine an den Rezeptor geben können.
4. Untersuchung der Gifte der Terebridae
Die inhibierende Aktivität einiger Gifte (Terebra consobrina, T. argus, Myurella affinis, Acus felina, A. chlorata, A. maculata und Hastulopsis pertusa) an nACh-Rezeptoren (α7, α3β2, α4β2, α3β4, α4β4 und α1β1γδ) wurde erstmals nachgewiesen. An Kalium- und Natriumkanälen zeigten die Giftextrakte keine Wirkung. Die Giftextrakte von Myurella affinis und Acus maculata waren am potentesten und blockierten alle untersuchten nACh-Rezeptoren. Dies ist besonders ungewöhnlich, da diese Terebriden-Arten nach der Literatur (Puillandre & Holford, 2010) keinen Giftapparat besitzen sollen. Eine weitere Auffälligkeit aller Terebriden-Giftextrakte war neben der Selektivität für a7-Rezeptorsubtypen, eine hohe Aktivität gegenüber α4-enthaltenden Rezeptoren. In den Giften und mit Hilfe von cDNA-Bibliotheken von Kegelschnecken konnte eine Vielzahl neuer Inhibitoren für neuronale nikotinische Acetylcholinrezeptoren identifiziert werden. Sie zeigen ein breites Wirkungsspektrum, das die unterschiedlichsten nAChRSubtypen einschließt, was ihre Verwendung als pharmaklogische Werkzeuge begrenzt. Hingegen zeigen die Gifte der Schraubenschnecken ein Selektivitätsspektrum, das die Analyse ihrer Peptide als vielversprechend erscheinen lässt.
Venomous secretions from marine snails of the Terebridae family target acetylcholine receptors

Venoms from cone snails (Conidae) have been extensively studied during the last decades, but those from other members of the suborder Toxoglossa, such as of Terebridae and Turridae superfamilies attracted less interest so far. Here, we report the effects of venom and gland extracts from three species of the superfamily Terebridae. By 2-electrode voltage-clamp technique the gland extracts were tested on Xenopus oocytes expressing nicotinic acetylcholine receptors (nAChRs) of rat neuronal (α3β2, α3β4, α4β2, α4β4, α7) and muscle subtypes (α1β1γδ), and expressing potassium (Kv1.2 and Kv1.3) and sodium channels (Nav1.2, 1.3, 1.4, 1.6). The extracts were shown to exhibit remarkably high inhibitory activities on almost all nAChRs tested, in particular on the α7 subtype suggesting the presence of peptides of the A-superfamily from the venom of Conus species. In contrast, no effects on the potassium and sodium channels tested were observed. The venoms of terebrid snails may offer an additional source of novel biologically active peptides.
Die wissenschaftliche Vernetzung, Betreuung und Unterstützung in der Promotionsphase steht im Zentrum dieser Forschungsarbeit, für die eine soziale Netzwerkanalyse (SNA) zur Karriereentwicklung in einem Bildungsforschungsprogramm in Deutschland durchgeführt wurde. Als Ausgangsproblem wird die schwierige Situation der unsicheren beruflichen Perspektiven Promovierter aufgegriffen.
Die Arbeit beschäftigt sich mit der Frage, wie Promovierende sowohl eine hohe fachliche Qualifikation erreichen, die für eine wissenschaftliche Karriere erforderlich ist, als auch eine gute Vernetzung in der wissenschaftlichen Gemeinschaft aufbauen können, um sich Chancen für die nächsten Karriereschritte zu eröffnen. Aus der Perspektive der Promovierten werden Chancen und Risiken der Vernetzung behandelt, um Überlegungen zu förderlichen und hinderlichen Aspekten der Betreuung und Unterstützung anzustellen.
Die Ergebnisse dieser Forschungsarbeit zeigen auf, welche Bedeutung soziale Netzwerke in der Berufsbiografie haben können und wie wissenschaftliches und soziales Kapital für die Karriere genutzt werden kann. Dazu richtet sich die erziehungswissenschaftliche Perspektive auf Prozesse des Wissenserwerbs und gemeinschaftlicher Lernprozesse, die zur Qualifizierung dienen und unterschiedliche Modelle von Betreuung umfassen. Im Verlauf der Promotionsphase und der weiteren wissenschaftlichen Karriere beginnen die Promovierenden auf der Wissensgrundlage ihres Studiums als Neulinge ihre erste umfangreiche empirische Forschungsarbeit. Sie werden durch die Mitarbeit im Projekt und ihre Beteiligung an Aktivitäten an die Arbeitsweisen in der Wissenschaft herangeführt. Von einer solchen eher randständigen Position aus erwerben sie Wissen, lernen hinzu, tauschen sich mit fortgeschrittenen WissenschaftlerInnen aus und sammeln eigene Erfahrungen. Nach einiger Zeit weiterer Qualifizierung und größerer Sichtbarkeit in der wissenschaftlichen Gemeinschaft können sie Positionen näher am Zentrum ihres Forschungsgebiets einnehmen, die auf ihren Kenntnissen und ihrer zunehmenden fachlichen und methodischen Expertise beruhen.
Neue Erkenntnisse über Betreuungsmodelle sind hilfreich, denn die Betreuung orientiert sich mal stark an dem klassischen Lehrer-Schüler-Modell (Individualbetreuung) oder an einer gemeinsamen Betreuung in einer Forschungsgruppe (Gruppenbetreuung). Die Untersuchung der Bedeutung von sozialem Kapital und situiertem Lernen in der Qualifikationsphase dient dazu, die Vernetzung von Promovierten in ihrer späteren akademischen Karriere zu analysieren.
Investigators in the cognitive neurosciences have turned to Big Data to address persistent replication and reliability issues by increasing sample sizes, statistical power, and representativeness of data. While there is tremendous potential to advance science through open data sharing, these efforts unveil a host of new questions about how to integrate data arising from distinct sources and instruments. We focus on the most frequently assessed area of cognition - memory testing - and demonstrate a process for reliable data harmonization across three common measures. We aggregated raw data from 53 studies from around the world which measured at least one of three distinct verbal learning tasks, totaling N = 10,505 healthy and brain-injured individuals. A mega analysis was conducted using empirical bayes harmonization to isolate and remove site effects, followed by linear models which adjusted for common covariates. After corrections, a continuous item response theory (IRT) model estimated each individual subject’s latent verbal learning ability while accounting for item difficulties. Harmonization significantly reduced inter-site variance by 37% while preserving covariate effects. The effects of age, sex, and education on scores were found to be highly consistent across memory tests. IRT methods for equating scores across AVLTs agreed with held-out data of dually-administered tests, and these tools are made available for free online. This work demonstrates that large-scale data sharing and harmonization initiatives can offer opportunities to address reproducibility and integration challenges across the behavioral sciences.
Background: Early-life institutional deprivation produces disinhibited social engagement (DSE). Portrayed as a childhood condition, little is known about the persistence of DSE-type behaviours into, presentation during, and their impact on, functioning in adulthood.
Aims: We examine these issues in the young adult follow-up of the English and Romanian Adoptees study.
Method: A total of 122 of the original 165 Romanian adoptees who had spent up to 43 months as children in Ceauşescu's Romanian orphanages and 42 UK adoptees were assessed for DSE behaviours, neurodevelopmental and mental health problems, and impairment between ages 2 and 25 years.
Results: Young adult DSE behaviour was strongly associated with early childhood deprivation, with a sixfold increase for those who spent more than 6 months in institutions. However, although DSE overlapped with autism spectrum disorder and attention-deficit hyperactivity disorder symptoms it was not, in itself, related to broader patterns of mental health problems or impairments in daily functioning in young adulthood.
Conclusions: DSE behaviour remained a prominent, but largely clinically benign, young adult feature of some adoptees who experienced early deprivation.
Reactive oxygen species (ROS) are derivatives of molecular oxygen (O2) involved in various physiological and pathological processes. In immune cells, ROS are mediators of pivotal functions such as phagocytosis, antigen presentation and recognition, cytolysis as well as phenotypical differentiation. Furthermore, ROS exert immunosuppressive effects on T and natural killer (NK) cells which is of particular importance in the so-called “tumor microenvironment” (TME) of solid tumors. This term describes the heterogenous group of non-malignant cells including tumor-associated fibroblasts and immune cells, vascular cells, bacteria etc. by which cancer cells are surrounded and with whom they engage in functional crosstalk. Importantly, pharmacological targeting of the TME and, specifically, tumor-associated immune cells utilizing immune checkpoint inhibitors - monoclonal antibodies that mitigate immunosuppression - turned out to be a major breakthrough in the treatment of malignant tumors. In this review, we aim to give an overview of the role that ROS produced in tumor-associated immune cells play during initiation, progression and metastatic outgrowth of solid cancers. Finally, we summarize findings on how ROS in the TME could be targeted therapeutically to increase the efficacy of cancer immunotherapy and discuss factors determining therapeutic success of redox modulation in tumors.
Viele Studien konnten nachweisen, dass die Produktion von cGMP eine entscheidende Funktion im nozizeptiven System einnimmt. Hierbei wurde vor allem die cGMP-Produktion über lösliche Guanylatzyklasen untersucht. Welche Rolle die partikulären Guanlyatzyklasen bei der Entstehung von Schmerzen haben ist weitgehend ungeklärt. Die vorliegende Arbeit zeigte, dass die partikuläre Guanylatzyklase NPR2 stark in DRG-Neuronen exprimiert wird und dort mit cGKI-alpha sowie CRP4 colokalisiert ist. Aktiviert wird NPR2 über den Peptidliganden CNP. Hervorzuheben ist, dass CNP nicht in primär afferenten Neuronen, dafür jedoch vermehrt im Dorsalhorn des Rückenmarks gebildet wird. Tierexperimentelle Untersuchungen zeigten, dass SNS-Npr2-/--Mäuse ein verringertes Schmerzverhalten bei thermischer Stimulation aufwiesen. Während sie im Capsaicin-Test keinen Phänotyp zeigten, wiesen sie in Phase II des Formalin-Modells ein signifikant reduziertes Leckverhalten auf. Diese Ergebnisse liefern Hinweise für eine Beteiligung des CNP/NPR2/cGKI Signalwegs an der Detektion von Hitzeschmerz und an der TRPA1-vermittelten Schmerzantwort. Dabei scheint NPR2 eine pronozizeptive Funktion zu besitzen. CRP4 als Zielprotein scheint hingegen eine antinozizeptive Wirkung zu haben. Zudem kann die Hypothese aufgestellt werden, dass CNP über einen retrograden Transport aus dem Rückenmark die Aktivierung von NPR2 auslösen könnte. Zusammengefasst zeigen die Daten dieser Arbeit, dass eine cGMP-abhängige Aktivierung durch NPR2 primär für die Detektion thermischer Reize zuständig ist, während die Literatur Hinweise darauf gibt, dass lösliche Guanylatzyklasen vor allem an inflammatorischen und neuropathischen Prozessen beteiligt sind. Daher scheinen partikuläre und lösliche Guanylatzyklasen unterschiedliche Eigenschaften im nozizeptiven System zu besitzen.
Biochemical and functional analysis of the ubiquitin binding properties of the NF-κB regulator NEMO

Posttranslationale Modifikationen regulieren wesentliche Eigenschaften von Proteinen, wie z. B. Lokalisation, Konformation, Aktivität, Stabilität und Interaktionsfähigkeit. Eine besondere Form der Proteinmodifikation ist die Ubiquitylierung, bei der das kleine Protein Ubiquitin mit seinem C-Terminus kovalent an ein Substratprotein gebunden wird.
Die am besten untersuchte Funktion der Ubiquitylierung ist die Markierung eines Substrates für den Abbau durch das Proteasom. In den letzten Jahren wurde jedoch entdeckt, dass Ubiquitylierung in vielen Bereichen der Zelle eine wichtige Rolle spielt. Dazu gehören der Transport von Vesikeln, die Reparatur von DNA-Schäden und zelluläre Signalübertragung. Ubiquitin kann verschieden-artige Ketten bilden, indem ein Ubiquitin an eines der sieben Lysine (K6, K11, K27, K29, K33, K48, K63) oder den N-Terminus eines anderen gebunden wird. Diese unterschiedlichen Kettentypen regulieren verschiedene Prozesse. Z. B. dienen K48-verknüpfte Ubiquitinketten als Signal für den proteasomalen Abbau, wohingegen über K63 verknüpfte Ketten hauptsächlich eine Rolle bei Signalübertragungen spielen.
Die meisten Funktionen die durch Ubiquitylierung reguliert werden, werden durch Ubiquitinrezeptoren vermittelt, die eine Ubiquitinbindedomäne (UBD) besitzen. Manche UBDs binden selektiv nur einen Ubiquitinkettentyp und sind somit in der Lage gezielt Prozesse regulieren zu können, indem sie nur durch diesen speziellen Kettentyp aktiviert werden.
Das Protein NEMO ist ein Ubiquitinrezeptor, dessen UBD UBAN selektiv bestimmte Ubiquitinketten bindet. NEMO spielt eine zentrale Rolle bei der Aktivierung der Transkriptionsfaktorfamilie NF-κB, indem es den IKK-Kinasekomplex reguliert. Dieser Kinasekomplex sorgt durch die Phosphorylierung des NF-κB-Inhibitors IκBα für dessen proteasomalen Abbau, wodurch schließlich NF-κB aktiviert wird. Die NF-κB-Aktivierung kann u. a. durch den TNF-Rezeptor (TNFR) induziert werden. Am aktivierten TNFR werden viele Proteine durch verschiedene Ubiquitinketten modifiziert. Bisher wurde angenommen, dass die spezifische Bindung von NEMO an K63-verknüpfte Ubiquitinketten ausschlaggebend für die Aktivierung von IKK ist. Jedoch spielen lineare Ubiquitinketten, die über den N-Terminus verknüpft sind, auch eine wichtige Rolle bei der Aktivierung von NF-κB und die UBAN von NEMO hat eine sehr hohe Affinität zu linearen Ubiquitinketten.
Um die genauen Vorgänge zu verstehen, die zur Aktivierung von NF-κB am TNFR führen, ist es nötig, zu analysieren, welche Proteine mit welchen Ubiquitinketten modifiziert werden und welche Ubiquitinrezeptoren daran binden.
In dieser Studie sollte detailliert untersucht werden, mit welchen Ubiquitin-ketten NEMO bevorzugt interagiert. Dazu wurden in vitro-Bindungsstudien mit bakteriell aufgereinigtem NEMO und verschiedenen Ubiquitinketten durchgeführt. Des Weiteren sollte geprüft werden, wie die Bindung von NEMO an bestimmte Ubiquitinketten die Aktivierung von NF-κB reguliert.
Dabei ergab sich, dass sowohl NEMO in voller Länge, als auch die UBAN, bevorzugt mit linearen Ubiquitinketten interagieren, wohingegen die Interaktion von NEMO mit anderen Ubiquitinketten relativ schwach ist. Ausgehend von einer Kristallstruktur eines Komplexes aus der NEMO-UBAN und linearem di-Ubiquitin, wurden NEMO-Mutanten generiert, die seletkiv die Bindung von NEMO an lineare Ubiquitinketten verhindern, während die schwache Bindung von NEMO an längere K63-verknüpfte Ketten erhalten blieb. Um die Relevanz der Interaktion von NEMO mit linearen Ubiquitinketten für die Aktivierung von NF κB zu überprüfen, wurden diese NEMO-Mutanten dann verwendet um Zellen die kein NEMO exprimieren zu rekonstituieren. Nach Stimulation dieser Zellen mit TNFα wurde NF-κB kaum aktiviert, womit gezeigt werden konnte, dass NEMO gezielt an lineare Ubiquitinketten binden muss, um NF-κB zu aktivieren. Zusätzlich zu seiner Rolle bei der Aktivierung von NF-κB ist NEMO ein wichtiger Inhibitor der durch den TNFR induzierten Apoptose. In dieser Studie wurde gezeigt, dass diese Apoptoseinhibierung abhängig von der Bindung von NEMO an lineare Ubiquitinketten ist, da die Zellen die NEMO-Mutanten exprimierten, die keine linearen Ketten binden können, durch Apoptose starben, währen Wildtyp-Zellen überlebten.
Zusammenfassend konnte in dieser Studie gezeigt werden, dass NEMO bevorzugt und mit vergleichsweise hoher Affinität an lineare Ubiquitinketten bindet und dass diese spezifische Bindung wichtig für die Inhibierung von TNFR-induzierter Apoptose sowie für die Aktivierung von NF-κB ist.
This dissertation is concerned with the role of prosody and, specifically, linguistic rhythm for the syntactic processing of written text. My aim is to put forward, provide evidence for, and defend the following claims:
1. While processing written sentences, readers make use of their phonological knowledge and generate a mental prosodic-phonological representation of the printed text.
2. The mental prosodic representation is constructed in accordance with a syntactic description of the written string. Constraints at the interface of syntax and phonology provide for the compatibility of the syntactic analysis and the (mental) prosodic rendition of the sentence.
3. The implicit prosodic structure readers impose on the written string entails phonological phrasing and accentuation, but also lower level prosodic features such as linguistic rhythm which emerges from the pattern of stressed and unstressed syllables.
4. Phonological well-formedness conditions accompany and influence the process of syntactic parsing in reading from the very beginning, i.e. already at the level of recognizing lexical categories. At points of underspecified syntactic structure, syntactic parsing decisions may be made on the basis of phonological constraints alone.
5. In reading, the implicit local lexical-prosodic information may be more readily available to the processing mechanism than higher-level discourse structural representations and consequently may have more immediate influence on sentence processing.
6. The process of sentence comprehension in reading is conditioned by factors that are geared towards sentence production.
7. The interplay of syntactic and phonological processes in reading can be explained with recourse to a performance-compatible competence grammar.
The evidence from three reading experiments supports these points and suggests a model of grammatical competence in which constraints from various domains (syntax, semantics, pragmatics, discourse structure, and phonology) interact in providing the possible structural, i.e. grammatical descriptions.
Does linguistic rhythm matter to syntax, and if so, what kinds of syntactic decisions are susceptible to rhythm? By means of two recall-based sentence production experiments and two corpus studies – one on spoken and one on written language – we investigated whether linguistic rhythm affects the choice between introduced and un-introduced complement clauses in German. Apart from the presence or absence of the complementiser dass (‘that’), these two sentence types differ with respect to the position of the tensed verb (verb-final/verb-second). Against our predictions, that were based on previously reported rhythmic effects on the use of the optional complementiser that in English, the experiments fail to obtain compelling evidence for rhythmic/prosodic influences on the structure of complement clauses in German. An overview of pertinent studies showing rhythmic influences on syntactic encoding suggests these effects to be generally restricted to syntactic domains smaller than a clause. We assume that, in the course of language production, initially, clause level syntactic projections are specified; their specification is in fact the prerequisite for phonological encoding to start. Consequently, prosodic effects may only touch upon the lower level categories that are to be integrated into the clausal projection, but not upon the syntactic makeup of the higher order projection itself.
Reduplicative words like chiffchaff or helter-skelter are part of ordinary language use yet most often found in substandard registers in which attitudinal and expressive meaning components are iconically foregrounded. In a rating experiment using nonwords that either conform to, or deviate from, conventional reduplicative patterns in German, the present study identified affective meaning dimensions, judgments of familiarity and esthetic evaluations of sound qualities associated with such words. In a subsequent recall test, we examined
the respective mnemonic potential of the different types of reduplication. Results suggest that, in the absence of semantic content, reduplicative forms are inherently associated with
several affective meaning associations that are generally considered positive. Two types of reduplicative patterns, namely full reduplication and [i-a]-vowel-alternating reduplication,
boost these positive effects to a particularly pronounced degree, leading to an increase in perceived euphony, funniness, familiarity, appreciation, and positive belittling (cuteness) and, at the same time, a decrease in arousal. These two types also turn out to be particularly memorable when compared both to other types of reduplication and to non-reduplicative structures. This study demonstrates that reduplicative morphology may in and of itself, that is, irrespective of the phonemic and the semantic content, contribute to the affective meaning and esthetic evaluation of words.
Understanding a sentence and integrating it into the discourse depends upon the identification of its focus, which, in spoken German, is marked by accentuation. In the case of written language, which lacks explicit cues to accent, readers have to draw on other kinds of information to determine the focus. We study the joint or interactive effects of two kinds of information that have no direct representation in print but have each been shown to be influential in the reader's text comprehension: (i) the (low-level) rhythmic-prosodic structure that is based on the distribution of lexically stressed syllables, and (ii) the (high-level) discourse context that is grounded in the memory of previous linguistic content. Systematically manipulating these factors, we examine the way readers resolve a syntactic ambiguity involving the scopally ambiguous focus operator auch (engl. “too”) in both oral (Experiment 1) and silent reading (Experiment 2). The results of both experiments attest that discourse context and local linguistic rhythm conspire to guide the syntactic and, concomitantly, the focus-structural analysis of ambiguous sentences. We argue that reading comprehension requires the (implicit) assignment of accents according to the focus structure and that, by establishing a prominence profile, the implicit prosodic rhythm directly affects accent assignment.
In erstem Teil der Arbeit wurde der Begriff Drehbuch in Hinsicht auf die Erstellung von eLearning Kursen erläutert und eLearning Inhalte auf ihre typischen Merkmale untersucht. Dabei war es wichtig zu verstehen, ob und wie die kooperative Arbeit an Inhalten in gängigen Textbearbeitungsprogrammen unterstützt wird und welche Vor- und Nachteile der jeweilige Editor aufweist. Wie sich herausgestellt hat, setzen bestehende Lösungen auf Auszeichnungen von Textbausteinen und der Kursstruktur mit Hilfe von speziellen Makros, so dass beim Transformieren des Kurses diese wiedererkannt werden können. Leider sind diese Lösungen auf einen bestimmten Editor spezialisiert und betrachten die kooperativen Aspekte der Arbeit an Inhalten nicht.
Da das Drehbuch in einem Textbearbeitungsprogramm erstellt wird, ist die Möglichkeit der Wahl von einem Editor für die Autoren vorteilhaft. Deshalb sieht der Lösungsansatz in meiner Arbeit vor, dass die Umsetzung des Drehbuches nicht auf einen bestimmten Editor beschränkt ist und dass für jeden Editor kooperative Bearbeitung ermöglicht wird. Dies wird zum einen durch Verwendung eines Versionskontrollesystems und zum anderen durch die kooperativen Eigenschaften der Editoren erreicht.
Das Transformieren eines Drehbuches zu einem eLearning Kurs wurde einheitlich gestaltet, da HTML als ein gemeinsames Format der Ausgabe von Editoren ausgewählt wurde. In der Implementierung des Lösungsansatzes konnte am Beispiel des Autorensprogramms LernBar die Unterstützung von bereits drei Editoren erreicht werden.
Einige Einschränkungen sind bei der Verwendung dieser Lösung zu beachten. Bei den Veränderungen in der HTML-Ausgabe der Editoren sind möglicherweise auch Anpassungen im Programm vorzunehmen. Die Verwendung von Tabellen impliziert, dass ihre Struktur durch den Autor nicht geändert wird, was durchaus auch versehentlich passieren kann. Bei der Erstellung von Tests ist eine umfassende Validierung empfehlenswert, damit die Tests wie erwartet durchgeführt werden können. Es wurden in der Implementierung nur einige Testtypen umgesetzt und möglicherweise sind einige Tests nicht für die Erfassung im Drehbuch geeignet. In dem Fall müssen sie in den verwendeten Autorenprogramm direkt erstellt werden.
Die rasante Entwicklung von Internettechnologien wird sicherlich neue Möglichkeiten sowohl für die kooperative Arbeit, als auch für Textbearbeitungsfunktionalitäten anbieten. Allein im Jahr 2010 wurden mehrere Neuerungen auf dem Markt von Online-Editoren eingeführt. Microsoft hat eine Online-Version1 von seiner Office-Suite veröffentlicht und damit eine neue Alternative zu Google Docs angeboten. In Google-Docs werden nahezu im monatlichen Takt neue nützliche Funktionen implementiert, die sowohl die Textverarbeitung als auch die Kooperation verbessern.

Research suggests that neural activity spanning prefrontal, parietal, temporal, and visual areas supports the generation of mental images. Exactly how this network controls the strength of visual imagery remains unknown. Here, brain imaging and transcranial magnetic phosphene data show that lower resting activity and excitability levels in early visual cortex (V1-V3) predict stronger sensory imagery. Electrically decreasing visual cortex excitability using tDCS increases imagery strength, demonstrating a causative role of visual cortex excitability in controlling visual imagery. These data suggest a neurophysiological mechanism of cortical excitability involved in controlling the strength of mental images.

Study Design: Survey of 100 worldwide spine surgeons.
Objective: To develop a spine injury score for the AOSpine Thoracolumbar Spine Injury Classification System.
Methods: Each respondent was asked to numerically grade the severity of each variable of the AOSpine Thoracolumbar Spine Injury Classification System. Using the results, as well as limited input from the AOSpine Trauma Knowledge Forum, the Thoracolumbar AOSpine Injury Score was developed.
Results: Beginning with 1 point for A1, groups A, B, and C were consecutively awarded an additional point (A1, 1 point; A2, 2 points; A3, 3 points); however, because of a significant increase in the severity between A3 and A4 and because the severity of A4 and B1 was similar, both A4 and B1 were awarded 5 points. An uneven stepwise increase in severity moving from N0 to N4, with a substantial increase in severity between N2 (nerve root injury with radicular symptoms) and N3 (incomplete spinal cord injury) injuries, was identified. Hence, each grade of neurologic injury was progressively given an additional point starting with 0 points for N0, and the substantial difference in severity between N2 and N3 injuries was recognized by elevating N3 to 4 points. Finally, 1 point was awarded to the M1 modifier (indeterminate posterolateral ligamentous complex injury).
Conclusion: The Thoracolumbar AOSpine Injury Score is an easy-to-use, data-driven metric that will allow for the development of a surgical algorithm to accompany the AOSpine Thoracolumbar Spine Injury Classification System.
Poster presentation at 5th German Conference on Cheminformatics: 23. CIC-Workshop Goslar, Germany. 8-10 November 2009 Protein kinases are important targets for drug development. The almost identical protein folding of kinases and the common co-substrate ATP leads to the problem of inhibitor selectivity. Type II inhibitors, targeting the inactive conformation of kinases, occupy a hydrophobic pocket with less conserved surrounding amino acids. Human polo-like kinase 1 (Plk1) represents a promising target for approaches to identify new therapeutic agents. Plk1 belongs to a family of highly conserved serine/threonine kinases, and is a key player in mitosis, where it modulates the spindle checkpoint at metaphase/anaphase transition. Plk1 is over-expressed in all today analyzed human tumors of different origin and serves as a negative prognostic marker in cancer patients. The newly identified inhibitor, SBE13, a vanillin derivative, targets Plk1 in its inactive conformation. This leads to selectivity within the Plk family and towards Aurora A. This selectivity can be explained by docking studies of SBE13 into the binding pocket of homology models of Plk1, Plk2 and Plk3 in their inactive conformation. SBE13 showed anti-proliferative effects in cancer cell lines of different origins with EC50 values between 5 microM and 39 microM and induced apoptosis. Increasing concentrations of SBE13 result in increasing amounts of cells in G2/M phase 13 hours after double thymidin block of HeLa cells. The kinase activity of Plk1 was inhibited with an IC50 of 200 pM. Taken together, we could show that carefully designed structure-based virtual screening is well-suited to identify selective type II kinase inhibitors targeting Plk1 as potential anti-cancer therapeutics.
Klimawandel, demographische Veränderungen, steigende Energiepreise, politische Rahmensetzungen und rechtliche Zielvorgaben erfordern eine Neuausrichtung der siedlungswasserwirtschaftlichen Leistungserbringung. Ziel ist, die Siedlungswasserwirtschaft nachhaltig und zukunftsfähig zu gestalten.
Das vorliegende Papier skizziert zum Thema „Instandhaltung der Wasserinfrastruktur:
finanzielle und organisatorische Spielräume“ Maßnahmen, die eine nachhaltige und zukunftsfähige Ausrichtung der siedlungswasserwirtschaftlicher Praxis unterstützen.
Die Maßnahmen wurden im Projekt im Rahmen von zwei szenariobasierten Planspielworkshops entwickelt. Ausgehend von den Diskussionsergebnissen legt das Papier dar, wo Handlungsmöglichkeiten ansetzen können und gibt zugleich Hinweise für die Umsetzung und Bewertung der vorgestellten Maßnahmen. Der Katalog ist dabei als Anstoß für eine stärkere Integration von Nachhaltigkeit in das unternehmerische Handeln zu verstehen.
Innovative Wasserinfrastrukturen, wie sie etwa mit den Neuartigen Sanitärsystemen entwickelt worden sind, versprechen Effizienzgewinne. Ihre Anwendung bedeutet nicht nur, den Einsatz neuer Techniken, sondern auch, dass sich die im konventionellen System erprobten Arbeitsteilungen zwischen verschiedenen Akteuren verändern. Ebenso können sich Beweggründe und Motivationen der beteiligten Akteure wandeln. Die Innovations- und Umsetzungsschritte werden dabei komplexer. Die Konstellationen der verschiedenen (heterogenen) Akteure und ihre Zusammenarbeit haben dabei hohe Relevanz für die Umsetzung innovativer Infrastrukturkonzepte.
Das vorliegende Diskussionspapier zeigt – aufbauend auf Ergebnisse aus zwei BMBFForschungsvorhaben – welcher Koordinationsbedarf bei einer Umsetzung auf der Quartiersebene zu erwarten ist. Zudem werden Hinweise gegeben, wie sich die Koordination zwischen den beteiligten Akteuren optimieren lässt.
Arzneimittelrückstände lassen sich heute in nahezu allen Gewässern nachweisen. Die Konzentrationen sind sehr gering; dennoch werden Risiken für die Umwelt und die Menschen vermutet. Daher ist zu klären, ob und wie es möglich ist, in Verfolgung des Vorsorgegedankens ihren Eintrag in die aquatische Umwelt möglichst zu verhindern oder zumindest zu reduzieren. Hierzu bieten sich umwelttechnische und gesundheitspolitische Maßnahmen sowie innovative Ansätze in der Arzneimittelentwicklung an.
Im Rahmen des BMBF-Projekts „SAUBER+ Innovative Konzepte und Technologien für die separate Behandlung von Abwasser aus Einrichtungen des Gesundheitswesens“ sind im Austausch mit Praxisakteuren zwei Zukunftsszenarien entstanden. Diese beschreiben von heute bis zum Jahr 2030 wie Maßnahmen in den genannten Handlungsfeldern umgesetzt und miteinander kombiniert werden können. Das erste Szenario ist eine Fortführung der bisherigen Entwicklungen. Im zweiten wird hingegen davon ausgegangen, dass sich die Bundesregierung einer sektor-übergreifenden Nachhaltigkeitspolitik verpflichtet und sich auf diese Weise die Einträge in die aquatische Umwelt auf ein Minimum reduzieren lassen.
Partizipative Szenarioverfahren werden vielfach in der Nachhaltigkeitsforschung angewendet. Zu den Vorteilen zählen: Praxiswissen macht die Zukunftsbilder robuster, strategisches Denken wird gefördert, ein kreativer Diskussionsrahmen entsteht. Nur wie werden narrative Szenarien im transdisziplinären Kontext entwickelt?
Die vorliegende Arbeit versteht sich als differenzierte methodische Beschreibung. Der Erfahrungsbericht legt am Beispiel des BMBF-Forschungsprojektes „SAUBER+: Innovative Konzepte und Technologien für die separate Behandlung von Abwasser aus Einrichtungen des Gesundheitswesens“ die einzelnen Arbeitsschritte der Szenarioentwicklung offen und diskutiert ihren Stellenwert. Die Vorgehensweise im Projekt wird kritisch reflektiert; daraus werden Hinweise für die Verwendung partizipativer Szenarien in anderen Forschungsprojekten abgeleitet.
Cross-sectional findings suggest that volumes of specific hippocampal subfields increase in middle childhood and early adolescence. In contrast, a small number of available longitudinal studies reported decreased volumes in most subfields over this age range. Further, it remains unknown whether structural changes in development are associated with corresponding gains in children’s memory. Here we report cross-sectional age differences in children’s hippocampal subfield volumes together with longitudinal developmental trajectories and their relationships with memory performance. In two waves, 109 participants aged 6–10 years (wave 1: MAge=7.25, wave 2: MAge=9.27) underwent high-resolution magnetic resonance imaging to assess hippocampal subfield volumes (imaging data available at both waves for 65 participants) and completed tasks assessing hippocampus dependent memory processes. We found that cross-sectional age-associations and longitudinal developmental trends in hippocampal subfield volumes were discrepant, both by subfields and in direction. Further, volumetric changes were largely unrelated to changes in memory, with the exception that increase in subiculum volume was associated with gains in spatial memory. Longitudinal and cross-sectional patterns of brain-cognition couplings were also discrepant. We discuss potential sources of these discrepancies. This study underscores that children’s structural brain development and its relationship to cognition cannot be inferred from cross-sectional age comparisons.

Mit dem Ziel der kardiovaskulären Primärprävention wurden insgesamt 185 Schülerinnen und Schüler 7. Klassen einer Gesamtschule mit Gymnasial-, Real- und Hauptschulzweig für einen Tag in eine kardiologische Reha-Klinik eingeladen. Dabei wurde ihnen Wissenswertes über Herz-/Kreislauferkrankungen in Theorie und Praxis vorgestellt. Dadurch konnte bei rund einem Drittel der Schüler anhaltende Lebensstiländerungen bezüglich Ernährung, Aktivität und Tabakkonsum erzielt werden. Der Ansatz einer umfassenden Intervention bei 11- bis 13-jährigen Schülern außerhalb der Schule erwies sich als praktikabel und mit 10€ pro Schüler als erschwinglich.
In der Neurobiologie nimmt die Untersuchung der Großhirnrinde (Neokortex) eine gewisse Sonderstellung ein, weil das Verständnis dieser hierarchisch übergeordneten Region für die Analyse bzw. Rekonstruktion der Hirnfunktionen insgesamt von entscheidender Bedeutung ist.
Dabei macht Folgendes die bedeutungsvolle Stellung des Neokortex aus: seine späte stammesgeschichtliche und ontogenetische Entwicklung, welche bei mehreren Säugetiergruppen mit einer ungewöhnlichen Massenzunahme und Plastizität verbunden ist und letzten Endes auch Raum für Individualität und Intelligenz schafft. Dabei kommt es speziell bei Primaten inklusive des Menschen zu einer zunehmenden Diversifizierung in Areale, welche primär den Sinnessystemen (Sehen, Hören, Tastsinn) sowie der Motorik zugeordnet sind. Mit steigender "Evolutionshöhe" der Säugetiere treten aber auch hierarchisch übergeordnete sekundäre, tertiäre und weitere Areale auf, welche zunehmend der Assoziation bzw. Integration von Sinnessystemen gewidmet sind. All diese Areale steuern die Interaktion des Individuums mit seiner Umwelt, d.h. sie formulieren anhand des aus der Peripherie eingehenden afferenten Inputs eine biologisch sinnvolle (motorische) Reiz-Antwort und ermöglichen in ihrer Gesamtheit (vor allem beim Menschen) auch kognitive Prozesse, so z.B. multisensorisches und assoziatives Denken, aber auch Antrieb, Planung, Erinnerung und ein hochkompliziertes Sozialverhalten.
Die Zielsetzung der vorliegenden Arbeit besteht darin, bei verschiedenen, teilweise extrem unterschiedlichen Säugetiergruppen über die vergleichende Morphologie der primären Neokortex-Areale zu einem besseren Verständnis grundsätzlicher neokortikaler Funktionsprinzipien (Input, intrinsische Verschaltung, Output) beizutragen. Die Einbeziehung phylogenetischer Aspekte kann dabei helfen, die kortikalen Spezifika der jeweiligen Säugetiere auf ihren Anpassungswert hin kritisch zu überprüfen.
Im Detail werden die vier primären Rindenfelder des auditorischen [A1], motorischen [M1], somatosensiblen [S1] und visuellen Systems [V1]) bei Spezies aus unterschiedlichen Ordnungen wie den Primaten (Mensch, Gorilla), Raubtieren (Hund), Paarhufer (Artiodactyla: Schwein, Schaf) sowie der Wale und Delphine (Zahnwale oder Odontoceti; u.a. Großer Tümmler, Schweinswal) anhand einer ganzen Palette von qualitativen und quantitativen Methoden konsequent miteinander
verglichen. Als eine solide Basis dient hier die allgemeine Zytoarchitektonik (Nissl- und teilweise Golgi-Färbung), welche durch immunhistochemische Marker (Calbindin, Calretinin, Parvalbumin und Neurofilament) um eine funktionell-neurobiologische Ebene erweitert wird. Die neben den Primaten im Fokus stehenden Delphine, welche sich durch eine erstaunliche Uniformität ihrer Großhirnrinde auszeichnen, werden mittels der “design-basierten“ Stereologie zusätzlich auf die Neuronendichte der kortikalen Areale bzw. ihrer Rindenschichten hin untersucht. Dabei wurden anhand phylogenetischer und evolutionsbiologischer Überlegungen jeweils die Rindenschichten III-V als "Schlüsselregion" ausgewählt, um über die Berechnung von Neuronendichten innerhalb dieser Schichten III und V mehr über die funktionellen Implikationen dieses eigentümlichen Neokortex herauszufinden.
Insgesamt zeigt sich, dass der Neokortex im Laufe der Evolution wohl gerade bei den landlebenden Primaten besonders stark diversifiziert worden ist: ihre vier primären Rindenfelder unterscheiden sich im Vergleich mit anderen Säugetieren besonders deutlich: hinsichtlich der Rindengliederung, der Ausstattung mit Neuronentypen sowie der intrinsischen Verschaltung erreicht dieser Kortex ein Höchstmaß an Komplexität. Besonders deutlich wird dies in den granulären Arealen, welche besonders viele Körnerzellen aufweisen (vor allem in der inneren Körnerzellschicht, Lamina IV).
Demgegenüber finden sich bei den holaquatischen Delphinen stark abweichende Verhältnisse. Ihre Großhirnrinde erscheint nicht nur allgemein recht einheitlich bzw. monoton, sondern auch in ihrer intrinsischen Funktionsweise stark abgeleitet: hier zeigt sich ein genereller Trend zur Entwicklung einer uniformen Rinde auf gänzlich pyramidal-agranulärer Basis, welche durch das Fehlen einer deutlich erkennbaren Schicht IV gekennzeichnet ist.
Bei einem Vergleich der bearbeiteten Säugetiergruppen ergibt sich ein neokortikales Kontinuum zwischen der granulären und der pyramidalen Bauweise zweier herausgehobener “Modellsäuger“, welche einander gewissermaßen als morphologische und funktionelle Extreme gegenüberstehen: mit den Primaten als dem einen (granulären) Endpunkt und den Delphinen als dem anderen (pyramidalen) Endpunkt sowie dazwischen vermittelnden Übergangsformen.
Die Hirnrinde des terrestrischen Karnivoren (Hund) zeigt dabei zuweilen Charakteristika, welche ihn eher in die Nähe der Primaten rücken. Bei den
terrestrischen Paarhufern (Schaf, Schwein) finden sich dagegen manche Kortex-Merkmale, welche an die Situation bei Delphinen erinnern.
Von besonderer Bedeutung ist hier nun die Feststellung, dass wohl gerade die sekundäre Anpassung der Zahnwale (Delphine) an eine ausschließlich aquatische Lebensweise für die Ausbildung ihres ureigenen Typus von Kortex entscheidend gewesen sein dürfte. In diesem Zusammenhang werden die physikalischen Eigenschaften von Wasser die Rahmenbedingungen für evolutionäre Abwandlungen innerhalb des Gehirns als Ganzem vorgegeben haben. Interessanterweise werden neben den höchst- enzephalisierten Vertretern unter den Primaten (Hominidae; Mensch und Menschenaffen) auch den Zahnwalen (Delphinen) von manchen Neurobiologen herausragende kognitive und intellektuelle Fähigkeiten zugeschrieben - trotz der diametral unterschiedlichen Organisation ihres Kortex. Ob und inwieweit dies zutrifft, bleibt weiteren Untersuchungen vorbehalten.
Die Ergebnisse der vorliegenden Arbeit verlangen für die Zukunft nach weiteren gleichartigen Untersuchungen der nicht-primären, also hierarchisch übergeordneten neokortikalen Assoziationsareale im Hinblick auf funktionelle und evolutions-biologische Implikationen.
Die überwältigende Expansion des Neokortex und die daran gekoppelte starke Zunahme der neurobiologischen Kapazität des Gehirns insgesamt dürfte nicht nur bei den Primaten, sondern (in bescheidenerem Umfang) auch bei Delphinen zur Entstehung von kortikalen "Freiräumen" geführt haben. Beim Menschen handelt es sich bei diesem Zuwachs an Hirnmasse und Hirnkapazität vermutlich um das Substrat für die Entstehung der phylogenetisch jüngsten und kognitiv höchstabgeleiteten Merkmale, nämlich Sprache, Persönlichkeit und Vernunft.

Während sich staatliche Sicherheitspolitik fortwährend verändert, scheint die Kritik an ihr verhältnismäßig unflexibel. Vielmehr wird der ständige Prozess der Ausdifferenzierung und Transformation der staatlichen Sicherheitsapparate von ihren Kritiker_innen seit Jahrzehnten so vehement mit Warnungen vor der Herausbildung eines totalitären Überwachungsstaates begleitet, dass diese zunehmend zu Unkenrufen verkommen. Mit dem Begriff des Sicherheitsregimes wird im Folgenden gegen einen Alarmismus argumentiert, der sich in Hinweisen auf die ständige Eskalation von Sicherheitspolitiken und eine damit einhergehende Beschneidung der Freiheit von Bürger_innen, Prekarisierten, Exkludierten oder sozialen Bewegungen erschöpft. Denn eine Kritik, die bei Skandalisierungen verbleibt, ist einerseits blind für qualitative Veränderungen. Andererseits schreibt sie selbst tradierte Herrschaftsverhältnisse fest, indem sie Vergangenes romantisiert und so an der Erzählung von der grundsätzlichen Alternativlosigkeit einer herrschaftsförmig organisierten Gesellschaft mitwirkt.
Dagegen wird eine materialistische Analyseperspektive eingenommen, die die Transformationen von Politiken der Sicherheit im Kontext der historischen Entwicklung der kapitalistischen Produktionsweise begreift. Anhand von Fallbeispielen zum Polizieren der Felder häusliche Gewalt und gewaltaffine Fußballfans in Frankfurt am Main wird die neoliberale Konzeption des umfassenden Zusammenwirkens öffentlicher, privatwirtschaftlicher und zivilgesellschaftlicher Akteure empirisch skizziert und das sich ausdifferenzierende Akteursfeld in diesem Bereich dargestellt. So können auch soziale Konfliktlinien identifiziert werden, die über die herkömmliche Gegenüberstellung von Freiheit und Sicherheit hinausweisen.
Macrophages are highly versatile cells, which acquire, depending on their microenvironment, pro- (M1-like), or antiinflammatory (M2-like) phenotypes. Here, we studied the role of the G-protein coupled receptor G2A (GPR132), in chemotactic migration and polarization of macrophages, using the zymosan-model of acute inflammation. G2A-deficient mice showed a reduced zymosan-induced thermal hyperalgesia, which was reversed after macrophage depletion. Fittingly, the number of M1-like macrophages was reduced in the inflamed tissue in G2A-deficient mice. However, G2A activation was not sufficient to promote M1-polarization in bone marrow-derived macrophages. While the number of monocyte-derived macrophages in the inflamed paw was not altered, G2A-deficient mice had less macrophages in the direct vicinity of the origin of inflammation, an area marked by the presence of zymosan, neutrophil accumulation and proinflammatory cytokines. Fittingly neutrophil efferocytosis was decreased in G2A-deficient mice and several lipids, which are released by neutrophils and promote G2A-mediated chemotaxis, were increased in the inflamed tissue. Taken together, G2A is necessary to position macrophages in the proinflammatory microenvironment surrounding the center of inflammation. In absence of G2A the macrophages are localized in an antiinflammatory microenvironment and macrophage polarization is shifted toward M2-like macrophages.
Existing social stressor concepts disregard the variety of task-related situations at work that require skillful social behavior to maintain good social relationships while achieving certain task goals. In this article, we challenge the view that social stressors at work are solely dysfunctional aspects evoking employee ill health. Drawing from the challenge-hindrance stressor framework, we introduce the concept of social challenge stressors as a job characteristic and examine their relationships with individual well-and ill-being. In study 1, we developed a new scale for the measurement of social challenge stressors and tested the validity of the scale. Results from two independent samples indicated support for a single-factor structure and showed that social challenge stressors are distinct from related stressor concepts. Using two samples, one of which was already used to test the factor structure, we analyzed the unique contribution of social challenge stressors in predicting employee well- and ill-being. As expected, social challenge stressors were simultaneously related to psychological strain and well-being. Using time-lagged data, study 2 investigated mechanisms that may explain how social challenge stressors are linked to well-being and strain. In line with the stress-as-offense-to-self approach, we expected indirect relationships via self-esteem. Additionally, social support was expected to moderate the relationships between social stressors and self-esteem. Whereas the indirect relationships were mostly confirmed, we found no support for the buffering role of social support in the social hindrance stressors-self-esteem link. Although we found a moderation effect for social challenge stressors, results indicated a compensation model that conflicted with expectations.
Hintergrund: Mit dem Masterplan 2020 und den an mehreren Universitäten eingeführten Modellstudiengängen befindet sich das Medizinstudium aktuell im Umbruch. Sowohl im Regel- als auch im Modellstudiengang werden medizinrechtliche Aspekte überwiegend im Rahmen rechtsmedizinischer Ausbildungsabschnitte unterrichtet. Allerdings werden Studierende bereits während Famulaturen oder im praktischen Jahr mit juristischen Fragen konfrontiert.
Ziel der Studie war es herauszufinden, ob und in welchem Umfang Studierende der Humanmedizin insbesondere zur ärztlichen Aufklärung und zu den ärztlichen Informationspflichten bis zum Beginn des 4. bzw. 5. klinischen Semesters auf medizinrechtliche Aspekte vorbereitet wurden, und ob Verbesserungen bei der medizinrechtlichen Lehre gewünscht werden.
Material und Methoden: Zwischen den Sommersemestern 2017 und 2019 wurde zu Beginn des Kurses für Rechtsmedizin eine quantitative, standardisierte Umfrage mit insgesamt 373 Studierenden durchgeführt.
Ergebnisse: Wenngleich 98,8 % der Studierenden angaben, Aufklärungsgespräche bereits (mehrfach) praktisch ausgeübt zu haben, bestanden deutliche Defizite in Bezug auf die juristischen Anforderungen an das ärztliche Aufklärungsgespräch und dessen Delegation. So gaben lediglich 5,1 % der Studierenden an, die rechtlichen Grundlagen der ärztlichen Aufklärung sowie die entsprechende Norm aus dem Zivilrecht zu kennen. Über 80 % der Befragten fühlten sich unzureichend auf die rechtlichen Aspekte des praktischen Jahres vorbereitet. Über 90 % der Studierenden wünschten sich eine bessere medizinrechtliche Ausbildung.
Diskussion: Eine fächerübergreifende Etablierung sowie eine über das gesamte Studium verteilte Lehre von Medizinrecht könnte die Vorbereitung auf das praktische Jahr verbessern und das Verständnis für die rechtlichen Anforderungen an die ärztliche Berufstätigkeit fördern. Nach dem derzeitigen Stand der Umsetzung des Masterplans 2020 soll das Medizinrecht in der Learning Opportunities, Objectives and Outcomes Platform (LOOOP) als verbindlicher Ausbildungs- und Lehrinhalt etabliert werden.
Ein 7 Monate alter weiblicher Säugling wurde mit Kontaktverbrennungen 2. Grades an beiden Beinen von seinen Eltern in der Notaufnahme eines Krankenhauses vorgestellt. Die Eltern berichteten, das Kind sei unbeaufsichtigt und nur mit einem Body bekleidet gegen den Nachtspeicherofen im Kinderzimmer gekrabbelt. Bei der 10 Tage später durchgeführten klinisch-rechtsmedizinischen Untersuchung zeigten sich streifige, teils parallel zueinander gestellte und gelenkübergreifende Verbrennungen an der rechten Oberschenkelaußen- und Unterschenkelrückaußenseite, an beiden Fußrücken und den Zehen sowie ein flächenhaftes Verbrennungsareal an der linken Unterschenkelaußenseite mit abgrenzbaren streifigen Anteilen.
Im Rahmen einer Ortsbegehung der elterlichen Wohnung mit Vermessung und Begutachtung der in der Wohnung befindlichen 3 Nachtspeicheröfen konnte zunächst festgestellt werden, dass sich die Verbrennungsmuster an den Beinen des Kindes mit dem Luftauslassgitter der beiden Nachtspeicheröfen im Wohn- und im Elternschlafzimmer (jeweils identisches Modell), hingegen nicht mit dem des Nachtspeicherofens im Kinderzimmer in Deckung bringen ließen. Für die Begutachtung konnte durch ergänzende Informationen eines technischen Sachverständigen zu den entsprechenden Nachtspeicheröfen und durch eine Literaturrecherche ein möglicher Geschehensablauf rekonstruiert werden.
Dieser Fall verdeutlicht zum einen, welche Gefahr für Säuglinge und Kleinkinder von Nachtspeicheröfen ausgehen kann, wenn diese nicht regelrecht gesichert und die Kinder unbeaufsichtigt sind. Zum anderen wird die Bedeutung einer detaillierten und – wenn nötig – interdisziplinären Rekonstruktion, inklusive einer Ortsbegehung, zur Abgrenzung eines möglichen Unfallhergangs von einer Kindesmisshandlung unterstrichen.
To gain a better understanding of complex mechanisms in biological systems, simultaneous control over multiple processes is key. To this purpose selective photouncaging has been developed. Photo-uncaging is an experimental scheme in which a molecule of interest has been inactivated synthetically and is activated by light. Usually a bond is cleaved and a leaving group is set free. The molecule which inactivates the molecule of interest and sets the leaving group free is called (photo-)cage. In a selective photo-uncaging scheme a number of leaving groups can be released independently, usually by irradiation with light of different wavelengths. This approach is, however, seriously limited in its applicability due to the properties of the involved cages and irradiation schemes. A major drawback is the usually quite broad UV-Vis absorption of the cages. This makes a selective activation by light difficult and limits the maximal number of independent cages severely.
Therefore, the aim of this thesis is to introduce the Vibrationally Promoted Electronic Resonance (VIPER) 2D-IR pulse sequence in a alternative selective uncaging scheme.
The VIPER 2D-IR pulse sequence is a spectroscopic tool which allows to generate 2D-IR signals whose lifetime are independent of the vibrational relaxation lifetime. It has been first used to monitor chemical exchange. It consists of a narrowband infared pump pulse, a subsequent UV-Vis pump pulse and a broadband infrared probe pulse. The UV-Vis pump pulse is off-resonant with regard to the UV-Vis absorption band. Electronic excitation becomes only possible, if the infrared pump pulse modulates the UV-Vis transition of the IR-excited molecule. This modulation brings the UV-Vis transition in resonance with the UV-Vis pump pulse. Thereby, only the molecules which were pre-excited with the infrared pulse can be excited into the electronically excited state. A computational prediction of the modulation was carried out by Jan von Cosel in the Burghardt group.
The narrowband infrared pump pulse can be used to selectively excite a subensemble of molecules in a mixture into an electronically excited state even if the UV-Vis spectra of all molecules are virtually identical. For this the sub-ensemble needs to exhibit an identifiable infrared spectrum. Combined with the introduction of isotope labels, which lead to changes in the infrared absorption spectra, the larger selectivity in the infrared region can be exploited for an alternative selective uncaging approach. In VIPER uncaging the infrared pump pulse selects the species and the subsequent UV-Vis pulse provides the energy needed for electronic excitation upon which the photo cleavage can occur.
After an introduction of the principle idea of uncaging and VIPER spectroscopy, the concept of VIPER uncaging is introduced and its limits and requirements are discussed. Some examples for possible VIPER cages are reviewed.
A coumarin molecule (7-diethylamino coumarin) which can release an azide group was chosen as a first test molecule for VIPER uncaging. Its isotopomers were characterized to determine suitable spectroscopic markers for successful uncaging and to find fitting experimental conditions. The chosen coumarin cage has an UV-Vis absorption band at approximately 380 nm and a steep flank on the high wavelength side of the band. The quantum yield for the azide compound is between 10-20 % depending on the solvent’s water content. The release was found to be on a picosecond timescale which is among the fastest known photo reactions, but the photo reaction mechanism has proven to be not straightforward. For the VIPER experiment on the mixture two isotopomers were chosen with a 13C atom at different positions. In one species a ring mode of the coumarin is changed by the 13C atom. In the other isotopomer the carbonyl stretching mode is influenced. The change in the ring mode region allows to select one species or the other with the infrared pre-excitation. Because of experimental difficulties only isotopomers with the same leaving group could be used. The successful selective electronic excitation of the individual isotopomers in a mixture was monitored by probing the carbonyl region.
As a second VIPER cage, para-hydroxyphenacyl (pHP) was chosen. A thiocyanate group was selected as leaving group. pHP cages have their electronic transition in the UV, with a maximum absorption at 290 nm. The shape of the spectrum is suitable and the quantum yield is very high, with values in the literature of up to 90 %. Also the photo reaction is well studied and the expected byproducts are well characterized. The chosen isotopologues were characterized spectroscopically. The resulting data on the photo reaction were in agreement with the mechanism proposed in the literature. The mixture for the VIPER experiment consisted of two isotopologues, where for one species all the C atoms in the ring were labelled and for the other the C-atom in the thiocyanate leaving group was labelled. Here the release of the different leaving groups, labelled and unlabelled thiocyanate, could be monitored selectively. This shows that it is possible to selectively release a molecule in a mixture of caged molecules by applying the VIPER pulse sequence.
The samples were synthesized by Matiss Reinfelds from the Heckel group and the VIPER experiments were done together with Carsten Neumann and with support
of the Bredenbeck group.
The leaving groups were chosen because of their infrared absorption which allowed to directly monitor the successful cleavage by spectroscopy. This was needed for the proof-of-concept experiment and to allow direct optimization of the experimental parameters but is not necessarily a requirement for VIPER uncaging.
Concerning the selectivity of the VIPER uncaging, the approach is at the moment mainly limited by the infrared pulse energy. The selective VIPER excitation is competing with unselective excitation directly by just the UV-Vis pulse. A more intense infrared pump pulse would increase only the selective VIPER excitation and thereby improve the contrast to the unspecific background.
To address this issue, the first steps towards an alternative infrared light generation are undertaken. In this alternative approach the infrared light for preexcitation is directly generated by difference frequency generation of the laser output, i.e. the high energy 800 nm fundamental, and the output of a non-collinear optical parametric amplifier (NOPA). To achieve a narrowband pump pulse the pulses are chirped before mixing. In the scope of this thesis a NOPA has been installed and the mixing has been tested with available test crystal medium. While infrared wavelength region and power were not in the aspired range with this alternative crystal the feasibility of mixing between a NOPA output and the fundamental could be shown.
Other possibilities to increase the contrast to the unspecific background excitation by the UV-Vis pump pulse are discussed. For most applications of selective VIPER uncaging the detection by fs-laser spectroscopy will not be needed and could be replaced by other methods e.g. chromatography. This will allow the experimental parameters of the VIPER pulse sequence to be changed in a way which reduces unspecific excitation i.e. reducing the UV-Vis-pump energy and result in much better contrast.
In conclusion, the experimental data in this thesis shows the VIPER pulse sequence to be applicable to selective uncaging schemes and indicates measures to arrive at the specificity necessary for uncaging applications. This thesis was focused on uncaging photo reactions with isotopomers and isotopologues, but other types of photo reactions could in principle be controlled in the same way. It should be possible to address different isomers in mixtures or different ground states of proteins selectively. The discussed experiments are a significant step towards control over photo reactions in mixtures.
Bei Kindern mit akuter lymphoblastischer Leukämie ist eine möglichst frühe und genaue Diagnostik der Infiltration des Zentralen Nervensystems für die Festlegung der weiteren Therapie von essenzieller Bedeutung. Ziel dieser Studie war es, die diagnostische Wertigkeit der Schädel-MRT im Vergleich zum Standarddiagnostikum Lumbalpunktion bezüglich einer leukämischen Beteiligung des ZNS zu untersuchen. Außerdem sollte die Häufigkeit relevanter Zufallsbefunde festgestellt werden, um den Nutzen einer zusätzlich zur Lumbalpunktion durchgeführten MRT zu beurteilen. Es erfolgte eine retrospektive Analyse der Daten von 277 Patienten mit Erstdiagnose und 56 Patienten mit Rezidiv einer ALL, die zwischen 1998 und 2016 an der Klinik für Kinder- und Jugendmedizin des Universitätsklinikums Frankfurt am Main behandelt worden waren. Sie hatten im Rahmen der initialen Diagnostik zusätzlich zur Lumbalpunktion eine Schädel-MRT mit Kontrastmittel erhalten.
Der durchschnittliche zeitliche Abstand zwischen Diagnosestellung und MRT betrug 11 Tage (39,5 Tage bei Rezidivpatienten). Die Sensitivitäten und Spezifitäten der beiden diagnostischen Mittel MRT und Liquor wurden berechnet.
Dazu diente als Goldstandard die endgültige Diagnose des ZNS-Status, die entweder durch die Liquordiagnostik, die Bildgebung oder die Klinik (RetinaInfiltration, Fazialisparese) gestellt wurde.
Insgesamt fanden sich bei 14 der 277 Patienten mit Erstdiagnose Leukämie eine Infiltration des ZNS. Davon waren 2 Patienten in der MRT, 11 Patienten in der Lumbalpunktion und 2 Patienten durch eine Retina-Infiltration als positiv diagnostiziert worden. Nur ein Patient, der in der MRT positiv befundet worden war, hatte in der Liquordiagnostik ein negatives Ergebnis. Bei den 56 Patienten mit Rezidiv ergab die MRT 6 positive Befunde und die Liquordiagnostik zeigte 13 positive Befunde. 3 Patienten waren zudem klinisch mit Fazialisparese (n=2) und retinaler Infiltration (n=1) positiv zu werten. Diese 3 hatten jedoch auch in der Lumbalpunktion ein positives Ergebnis. Von den Patienten, die in der MRT positiv befundet wurden, hatte 1 Patient ein negatives Ergebnis in der Lumbalpunktion.
Dieser zeigte allerdings Symptome und hätte somit die Bildgebung ohnehin erhalten. Für die MRT ergibt sich bei den Patienten mit Erstdiagnose eine Sensitivität von 14,3%, bei den Rezidivpatienten eine Sensitivität von 43%. Die Spezifität liegt bei den Patienten mit Erstdiagnose ALL bei 99,6% und bei den Rezidivpatienten bei 100%. Für die Lumbalpunktion errechnet sich bei den Patienten mit Erstdiagnose eine Sensitivität von 78,6% und bei den Rezidivpatienten eine Sensitivität von 92,9%, mit einer Falsch-negativ-Rate von 21,4% und 7,1%. Die Spezifität der Lumbalpunktion liegt in beiden Gruppen bei 100%.
Bezüglich relevanter Zufallsbefunde ist bei den Patienten mit Erstdiagnose eine Sinusvenenthrombose bei einer klinisch unauffälligen Patientin zu nennen.
Weitere Nebenbefunde, die keine Auswirkungen auf die Therapie hatten, waren Schleimhautschwellungen der Nasennebenhöhlen (n=188), Verlegung der Mastoidzellen (n=45), Hirnvolumenminderung (n=27), Blutungen ohne Therapiebedürftigkeit (n=5), Zysten (n=11) und angeborene Fehlbildungen (n=7). Außerdem wurde bei 6 Rezidivpatienten eine chronische therapieassoziierte Leukenzephalopathie diagnostiziert.
Die vorliegende Studie stellt unseres Wissens nach die bisher umfangreichste Schädel-MRT-Studie bei Kindern mit ALL unter dieser Fragestellung dar. Ihre Nachteile ergeben sich durch die retrospektive Betrachtung und dadurch bedingte eingeschränkte Einheitlichkeit.
Aus unseren Ergebnissen lässt sich schlussfolgern, dass die Schädel-MRT keinen zusätzlichen Nutzen zur alleinigen Lumbalpunktion bringt. Nur ein einziger Patient hatte in der MRT ein positives Ergebnis, welches weder durch die Lumbalpunktion noch durch die Klinik erkannt worden war. Auch in Hinblick auf die geringe Rate an relevanten Nebenbefunden bei asymptomatischen Patienten ergibt sich keine grundsätzliche Notwendigkeit zur Durchführung dieser Bildgebung. Die zusätzliche Belastung einer kontrastmittelgestützten MRT, für die bei kleinen Kindern zudem häufig eine Sedierung erforderlich ist, kann klinisch neurologisch unauffälligen Patienten mit ALL also erspart werden.
Introduction: Preclinical CT-guided radiotherapy platforms are increasingly used but the CT images are characterized by poor soft tissue contrast. The aim of this study was to develop a robust and accurate method of MRI-guided radiotherapy (MR-IGRT) delivery to abdominal targets in the mouse.
Methods: A multimodality cradle was developed for providing subject immobilisation and its performance was evaluated. Whilst CT was still used for dose calculations, target identification was based on MRI. Each step of the radiotherapy planning procedure was validated initially in vitro using BANG gel dosimeters. Subsequently, MR-IGRT of normal adrenal glands with a size-matched collimated beam was performed. Additionally, the SK-N-SH neuroblastoma xenograft model and the transgenic KPC model of pancreatic ductal adenocarcinoma were used to demonstrate the applicability of our methods for the accurate delivery of radiation to CT-invisible abdominal tumours.
Results: The BANG gel phantoms demonstrated a targeting efficiency error of 0.56 ± 0.18 mm. The in vivo stability tests of body motion during MR-IGRT and the associated cradle transfer showed that the residual body movements are within this MR-IGRT targeting error. Accurate MR-IGRT of the normal adrenal glands with a size-matched collimated beam was confirmed by γH2AX staining. Regression in tumour volume was observed almost immediately post MR-IGRT in the neuroblastoma model, further demonstrating accuracy of x-ray delivery. Finally, MR-IGRT in the KPC model facilitated precise contouring and comparison of different treatment plans and radiotherapy dose distributions not only to the intra-abdominal tumour but also to the organs at risk.
Conclusion: This is, to our knowledge, the first study to demonstrate preclinical MR-IGRT in intra-abdominal organs. The proposed MR-IGRT method presents a state-of-the-art solution to enabling robust, accurate and efficient targeting of extracranial organs in the mouse and can operate with a sufficiently high throughput to allow fractionated treatments to be given.
Background: In Europe, the number of females exhibiting oppositional defiant disorder (ODD) and conduct disorder (CD) is growing. Many of these females live in youth welfare institutions. Consequently, there is a great need for evidence-based interventions within youth welfare settings. A recently developed approach targeting the specific needs of girls with ODD and CD in residential care is START NOW. The aim of this group-based behavioural skills training programme is to specifically enhance emotional regulation capacities to enable females with CD or ODD to appropriately deal with daily-life demands. It is intended to enhance psychosocial adjustment and well-being as well as reduce oppositional and aggressive behaviour. We present the study protocol (version 4.1; 10 February 2016) of the FemNAT-CD intervention trial titled "Group-Based Treatment of Adolescent Female Conduct Disorders: The Central Role of Emotion Regulation".
Methods/design: The study is a prospective, confirmatory, cluster-randomised, parallel-group, multi-centre, randomised controlled trial with 128 institutionalised female adolescents who fulfil the diagnostic criteria of ODD and/or CD. Institutions/wards will be randomised either to provide the 12-week skills training as an add-on intervention or to provide treatment as usual. Once the first cycle is completed, each institution will run a second cycle with the opposite condition. Primary endpoints are the pre-post change in number of CD/ODD symptoms as assessed by a standardised, semi-structured psychiatric interview (Kiddie Schedule for Affective Disorders and Schizophrenia for School-Age Children–Present and Lifetime, CD/ODD section) between baseline and the end of intervention, as well as between baseline and a 3-month follow-up point. Secondary objectives include pre-post change in CD/ODD-related outcome measures, most notably emotional regulation on a behavioural and neurobiological level after completion of START NOW compared with treatment as usual.
Discussion: To our knowledge, this study is the first to date to systematically investigate the effectiveness of an adapted integrative psychosocial intervention designed for female adolescents with ODD and CD in youth welfare settings.

Exposure to community violence through witnessing or being directly victimized has been associated with conduct problems in a range of studies. However, the relationship between community violence exposure (CVE) and conduct problems has never been studied separately in healthy individuals and individuals with conduct disorder (CD). Therefore, it is not clear whether the association between CVE and conduct problems is due to confounding factors, because those with high conduct problems also tend to live in more violent neighborhoods, i.e., an ecological fallacy. Hence, the aim of the present study was: (1) to investigate whether the association between recent CVE and current conduct problems holds true for healthy controls as well as adolescents with a diagnosis of CD; (2) to examine whether the association is stable in both groups when including effects of aggression subtypes (proactive/reactive aggression), age, gender, site and socioeconomic status (SES); and (3) to test whether proactive or reactive aggression mediate the link between CVE and conduct problems. Data from 1178 children and adolescents (62% female; 44% CD) aged between 9 years and 18 years from seven European countries were analyzed. Conduct problems were assessed using the Kiddie-Schedule of Affective Disorders and Schizophrenia diagnostic interview. Information about CVE and aggression subtypes was obtained using self-report questionnaires (Social and Health Assessment and Reactive-Proactive aggression Questionnaire (RPQ), respectively). The association between witnessing community violence and conduct problems was significant in both groups (adolescents with CD and healthy controls). The association was also stable after examining the mediating effects of aggression subtypes while including moderating effects of age, gender and SES and controlling for effects of site in both groups. There were no clear differences between the groups in the strength of the association between witnessing violence and conduct problems. However, we found evidence for a ceiling effect, i.e., individuals with very high levels of conduct problems could not show a further increase if exposed to CVE and vice versa. Results indicate that there was no evidence for an ecological fallacy being the primary cause of the association, i.e., CVE must be considered a valid risk factor in the etiology of CD.
Der Name Engelbert Humperdinck wird von den meisten Musikinteressierten unwillkürlich mit seinem berühmtesten Werk "Hänsel und Gretel" oder allgemein mit dem Begriff "Märchenoper" assoziiert. Auch herausragende Musikwissenschaftler - wie z.B. Carl Dahlhaus im "Neuen Handbuch der Musikwissenschaft" - beschränken sich beim Stichwort "Humperdinck" in der Regel auf Erläuterungen zu "Hänsel und Gretel". Humperdinck hat in der Tat eine ganze Reihe von Märchenstoffen vertont: Er begann mit Liedern zu Märchenspielen, die seine Schwester Adelheid Wette für ihre Kinder geschrieben hatte ("Schneewittchen" 1888, "Hänsel und Gretel" 1890 und "Die sieben Geislein" 1895). Nach dem Erfolg der durchkomponierten Oper "Hänsel und Gretel" 1893 versuchte er sich an der Gattung Melodram ("Königskinder" 1895/97 nach einem Text von Elsa Bernstein), einer Mischform aus Singspiel und Melodram ("Dornröschen" 1902 nach einem Text von Elisabeth Filhés-Ebeling) und schließlich wieder an der durchkomponierten Märchenoper ("Königskinder" 1910). Außerdem schrieb er ein Vorspiel und zwei Musiknummern zu Maurice Maeterlincks "Der blaue Vogel" (1910-12).
Jedoch war dies nur ein Bereich der Musik für die Bühne, in dem er sich betätigte. Eine bedeutende Phase war für Humperdinck die Zusammenarbeit mit Max Reinhardt am Deutschen Theater und Alfred Halm am "Deutschen Schauspielhaus" in Berlin (von 1905 bis 1907), in der u.a. Bühnenmusik zu vier Shakespeareschen Werken entstand ("Der Kaufmann von Venedig", "Der Sturm", "Das Wintermärchen" und "Was ihr wollt"). In seiner letzten Schaffensperiode konnte Humperdinck sich verstärkt der Gattung widmen, die ihm schon vor Hänsel und Gretel als Ausweg aus der Wagner-Nachfolge-Lethargie erschienen war, der komischen Oper ("Heirat wider Willen" 1905, "Die Marketenderin" 1914, "Gaudeamus" 1919).
In diesem Beitrag soll "Dornröschen" genauer betrachtet werden - ein Märchenspiel, das im Unterschied zu den beiden Märchenopern Humperdincks wenig Erfolg hatte und in der Literatur bisher so gut wie gar nicht behandelt wurde. Wie ist das Werk entstanden, was ist das Besondere an ihm, und welche Gründe gibt es für sein Scheitern auf der Bühne? Zunächst soll je doch ein kurzer Überblick über Inhalt, Besetzung und Form in das Werk ein führen...
„Zu viel Musik“ lautet der Titel eines Aufsatzes des Pianisten, Komponisten, Dirigenten und Musikschriftstellers Ferdinand Hiller (1811-1885) über die Musikbelästigung eines musikalischen Bürgers durch unterschiedlichste Quellen in seinem Umfeld im Laufe eines Tages, sicherlich auf eigenen Erfahrungen fußend.1 Er wird als Titel für diesen Beitrag im übertragenen Sinne zitiert, weil der Nachlass Hiller in der Universitätsbibliothek Frankfurt am Main beim ersten Sichten bisher sehr unübersichtlich erschien. Der Nachlass umfasst 74 Bände Musikhandschriften, davon 21 Sammelhandschriften, dazu 9 Mappen mit Stimmen, 59 Bände Musikdrucke eigener Werke, davon 29 Sammelbände, dazu eine dicke Mappe mit Stimmen. Weitere Handschriften wurden nachträglich erworben. In verschiedenen Sammlungen sind darüber hinaus Briefe und Bildnisse Hillers überliefert.
Dieser Beitrag hat das Ziel, eine strukturierte Übersicht über die enthaltenen Werke zu geben: Der Inhalt der im Druck überlieferten Sammelbände wird anhand einer Tabelle dargestellt. Den Hauptteil dieses Beitrages bildet eine nach musikalischen Gattungen gegliederte Liste der in Frankfurt überlieferten Musikautographe, die als Register zu dem mittler weile digital verfügbaren Nachlassverzeichnis von Christine Ihl genutzt werden kann.
Zwei große Theaterzettelsammlungen mit über 100.000 Stück sind seit dem ersten Drittel des 20. Jahrhunderts in der damaligen Stadt- und heutigen Universitätsbibliothek Frankfurt am Main beheimatet und machen sie in dieser Hinsicht zu einer bedeutenden theaterhistorischen Institution im deutschsprachigen Raum: die Sammlung der Städtischen Bühnen Frankfurt am Main und ihrer Vorläufer sowie die Sammlung des Manskopfschen Musik- und Theaterhistorischen Museums...
Epoxides and diols of polyunsaturated fatty acids (PUFAs) are bioactive and can influence processes such as tumor cell proliferation and angiogenesis. Studies with inhibitors of the soluble epoxide hydrolase (sEH) in animals overexpressing cytochrome P450 enzymes or following the systemic administration of specific epoxides revealed a markedly increased incidence of tumor metastases. To determine whether PUFA epoxides increased metastases in a model of spontaneous breast cancer, sEH-/- mice were crossed onto the polyoma middle T oncogene (PyMT) background. We found that the deletion of the sEH accelerated the growth of primary tumors and increased both the tumor macrophage count and angiogenesis. There were small differences in the epoxide/diol content of tumors, particularly in epoxyoctadecamonoenic acid versus dihydroxyoctadecenoic acid, and marked changes in the expression of proteins linked with cell proliferation and metabolism. However, there was no consequence of sEH inhibition on the formation of metastases in the lymph node or lung. Taken together, our results confirm previous reports of increased tumor growth in animals lacking sEH but fail to substantiate reports of enhanced lymph node or pulmonary metastases.
We present results for calculating fusion cross-sections using a new microscopic approach based on a time-dependent density-constrained DFT calculations. The theory is implemented by using densities and other information obtained from TDDFT time-evolution of the nuclear system as a constraint on the density for DFT calculations.
Sulphuric acid, ammonia, amines, and oxidised organics play a crucial role in nanoparticle formation in the atmosphere. In this study, we investigate the composition of nucleated nanoparticles formed from these compounds in the CLOUD (Cosmics Leaving Outdoor Droplets) chamber experiments at CERN (Centre européen pour la recherche nucléaire). The investigation was carried out via analysis of the particle hygroscopicity, ethanol affinity, oxidation state, and ion composition. Hygroscopicity was studied by a hygroscopic tandem differential mobility analyser and a cloud condensation nuclei counter, ethanol affinity by an organic differential mobility analyser and particle oxidation level by a high-resolution time-of-flight aerosol mass spectrometer. The ion composition was studied by an atmospheric pressure interface time-of-flight mass spectrometer. The volume fraction of the organics in the particles during their growth from sizes of a few nanometers to tens of nanometers was derived from measured hygroscopicity assuming the Zdanovskii–Stokes–Robinson relationship, and compared to values gained from the spectrometers. The ZSR-relationship was also applied to obtain the measured ethanol affinities during the particle growth, which were used to derive the volume fractions of sulphuric acid and the other inorganics (e.g. ammonium salts). In the presence of sulphuric acid and ammonia, particles with a mobility diameter of 150 nm were chemically neutralised to ammonium sulphate. In the presence of oxidation products of pinanediol, the organic volume fraction of freshly nucleated particles increased from 0.4 to ~0.9, with an increase in diameter from 2 to 63 nm. Conversely, the sulphuric acid volume fraction decreased from 0.6 to 0.1 when the particle diameter increased from 2 to 50 nm. The results provide information on the composition of nucleated aerosol particles during their growth in the presence of various combinations of sulphuric acid, ammonia, dimethylamine and organic oxidation products.
