With a growing Muslim population, many European countries need to integrate Muslims into their societies. One aspect that can hinder successful integration are substantial differences in human values. This is because such values are consequential for attitudes as well as behavior. We compare basic human values between Muslim immigrants and non-Muslim natives in four European countries with distinct immigration histories and integration politics: Belgium, France, Germany, and Sweden. For most insightful comparisons, we contrast values of Muslim immigrants with those of Christian natives as well as those of non-religious natives. We employ data of more than 50,000 individuals based on the first eight waves of the European Social Survey. Our findings reveal significant differences in value priorities between Muslims, Christians and non-religious individuals in all four countries. Amongst other things, Muslim immigrants score particularly high in conservation values (security and tradition/conformity). At the same time, they also score higher in self-transcendence values (benevolence as well as universalism). While many of these findings are in line with theory and previous research, the higher score in universalism is unexpected. A potential explanation is the combination of religious traditionalism and discrimination experiences. In other words, religious traditions are associated with more conservative views, but being subject to marginalization can still result in an appreciation of equal opportunities. We find only limited support for differences in hedonism. Religiosity correlates with values of tradition/conformity for Muslim immigrants as well as for Christian natives. Thus, accounting for religiosity renders differences in these values between Muslims and other groups statistically insignificant. While most of these findings hold in all countries, differences are most pronounced in Sweden and lower in the other three countries, which is also true after accounting for differences in socio-economic status and religiosity between the three groups. This suggests that a combination of a country's history of diversity and national integration policies either encourages the convergence of values or leads to a solidification of value differences between groups. We discuss these political and social implications of our findings.

A question of striking the right balance : how do digital media influence how we think and act?

What influence do digital technologies have on human perception, thinking and action? Do computer games harm the development of young brains? And is there really such a thing as »digital dementia«, an increasing forgetfulness caused by the use of modern technologies? For some of these questions, answers are available that are empirically corroborated.
Some anaerobic bacteria use biotin-dependent Na+-translocating decarboxylases (Bdc) of β-keto acids or their thioester analogs as key enzymes in their energy metabolism. Glutaconyl-CoA decarboxylase (Gcd), a member of this protein family, drives the endergonic translocation of Na+ across the membrane with the exergonic decarboxylation of glutaconyl-CoA (ΔG0’ ≈−30 kJ/mol) to crotonyl-CoA. Here, we report on the molecular characterization of Gcd from Clostridium symbiosum based on native PAGE, size exclusion chromatography (SEC) and laser-induced liquid bead ion desorption mass spectrometry (LILBID-MS). The obtained molecular mass of ca. 400 kDa fits to the DNA sequence-derived mass of 379 kDa with a subunit composition of 4 GcdA (65 kDa), 2 GcdB (35 kDa), GcdC1 (15 kDa), GcdC2 (14 kDa), and 2 GcdD (10 kDa). Low-resolution structural information was achieved from preliminary electron microscopic (EM) measurements, which resulted in a 3D reconstruction model based on negative-stained particles. The Gcd structure is built up of a membrane-spanning base primarily composed of the GcdB dimer and a solvent-exposed head with the GcdA tetramer as major component. Both globular parts are bridged by a linker presumably built up of segments of GcdC1, GcdC2 and the 2 GcdDs. The structure of the highly mobile Gcd complex represents a template for the global architecture of the Bdc family.
Background and Aim: Several studies observed alterations in the gut microbiota in patients with non‐alcoholic fatty liver disease (NAFLD). However, analyzed patient populations and methods strongly differ among these studies. The aim of this study was to prove the reproducibility of published results and to provide a detailed overview of all findings in our NAFLD cohort using next generation sequencing methods.
Methods: The individual taxonomic microbiota composition of fecal samples from 90 NAFLD patients and 21 healthy controls was analyzed using 16S rRNA gene sequencing. Study participants were grouped according to their disease stage and compared regarding their gut microbiota composition. Studies were identified from PubMed listed publications, and the results were compared with the findings in our cohort.
Results: Results from 13 identified studies were compared with our data. A decreased abundance of the Bacteroidetes and Ruminococcaceae as well as an increased abundance of Lactobacillaceae and Veillonellaceae and Dorea were the most frequently reported changes among NAFLD patients in 4/13, 5/13, 4/13, 2/13, and 3/13 studies, respectively. Even though these alterations in the gut microbiota composition were also observed in our patient cohort, the majority of published differences could not be reproduced, neither in our own nor in other NAFLD cohort studies.
Conclusion: Despite repeatedly reproduced abundance patterns of specific bacteria, the heterogeneous study results did not reveal a consistent disease specific gut microbiota signature. Further prospective studies with homogenous patient cohorts and standardized methods are necessary to phenotype NAFLD by the gut microbiota.
Overconsumption of carbohydrates and lipids are well known to cause nonalcoholic fatty liver disease (NAFLD), while the role of nutritional protein intake is less clear. In Western diet, meat and other animal products are the main protein source, with varying concentrations of specific amino acids. Whether the amount or composition of protein intake is associated with a higher risk for disease severity has not yet been examined. In this study, we investigated associations of dietary components with histological disease activity by analyzing detailed 14‐day food records in a cohort of 61 patients with biopsy‐proven NAFLD. Furthermore, we used 16S ribosomal RNA gene sequencing to detect associations with different abundances of the gut microbiota with dietary patterns. Patients with definite nonalcoholic steatohepatitis (NAFLD activity score of 5‐8 on liver biopsy) had a significantly higher daily relative intake of protein compared with patients with a NAFLD activity score of 0‐4 (18.0% vs. 15.8% of daily protein‐based calories, P = 0.018). After adjustment for several potentially confounding factors, a higher protein intake (≥17.3% of daily protein‐based calories) remained associated with definite nonalcoholic steatohepatitis, with an odds ratio of 5.09 (95% confidence interval 1.22‐21.25, P = 0.026). This association was driven primarily by serine, glycine, arginine, proline, phenylalanine, and methionine. A higher protein intake correlated with a lower Bacteroides abundance and an altered abundance of several other bacterial taxa. Conclusion: A high protein intake was independently associated with more active and severe histological disease activity in patients with NAFLD. Further studies are needed to investigate the potential harmful role of dietary amino acids on NAFLD, with special attention to meat as their major source.
Es ist schon merkwürdig: Für keines der großen Konzilien des Spätmittelalters schien in den letzten Jahren eine Gesamtdarstellung so nah wie im Fall des Pisanum. Dieter Girgensohn und Hélenè Millet legten eine Fülle von Einzelstudien vor, doch keiner der beiden goss sie in die Form eines opus magnum, sieht man von einem Aufsatzband der französischen Kollegin ab. Durch Mona Kirsch erfuhr die Synode 2016 eine zwar viele Facetten erfassende Würdigung im Rahmen einer ritualgeschichtlich grundierten Geschichte des allgemeinen Konzils im Spätmittelalter, zu der für sie aber die auf Pisa folgenden und damit eng verbundenen Versammlungen von Konstanz und Basel nicht gehörten. Und nun erklärt Florian Eßer gleich mehrfach, bei seinem hier anzuzeigenden, immerhin 874-seitigen Werk handele es sich keinesfalls um eine Gesamtdarstellung. Was die Frage aufwirft, wer angesichts einer solch zerklüfteten Landschaft partieller Monumente das finale Wagnis überhaupt noch angehen mag.
Am ehesten wohl Florian Eßer selbst, der eine auf umfassender Kenntnis der ungedruckten wie gedruckten Quellen beruhende und die gesamte Forschung zum Thema rezipierende Studie vorgelegt hat, die weniger als Dissertation – die sie ist – denn fast schon als Habilitationsschrift gelten darf. Nur wünscht man sich und ihm, dass er sich künftig in der Kunst des Kürzens und Streichens übt. Denn hier wird viel in der Sache wohlgemerkt Treffendes derart breit bis in die feinsten Einzelheiten und Verästelungen ausgeführt, ja ausgewalzt, dass dem Autor am Ende dabei offensichtlich selbst nicht mehr so recht wohl war, widmet er sein Opus doch Freundin, Eltern und Freunden, "auch wenn (beziehungsweise weil) sie es vermutlich nie lesen werden" (S. 14). ...
Für ausgewählten Patienten mit Aortenklappeninsuffizienz (AI) bietet die Aortenklappenrekonstruktion eine sehr attraktive Alternative zum Klappenersatz, wodurch mögliche prothesenbezogene Komplikationen vermieden werden können.
Die begrenzte Zahl der Langzeitstudien sowie das Fehlen von standardisierten Verfahren machen die klappenerhaltende Aortenklappenchirurgie technisch anspruchsvoll.
Ziel dieser Arbeit ist es, die Langzeitergebnisse nach Aortenklappenrekonstruktion zu erfassen und die Haltbarkeit sowie die klappenbezogenen Komplikationen nach klappenerhaltender Aortenklappenchirurgie zu untersuchen.
Hierzu analysierten wir die klinischen Daten von 560 Patienten, die eine operative Versorgung mittels Aortenklappenrekonstruktion erhielten. Dabei wurden sowohl Segelpathologien (bikuspid sowie trikuspid) als auch Aortenwurzelpathologien berücksichtigt. Bei 56% der Patienten (n=313) wurde eine Reimplantation nach David durchgeführt. Bei 247 Patienten wurde bei isolierter Segelpathologie ohne Aortenwurzelbeteiligung eine Segelrekonstruktion durchgeführt. Pathologien der Aortenwurzel konnten mit subkommissuralen Nähten (bei 62 Patienten) oder mit Rekonstruktion des sinotubulären Übergangs (bei 60 Patienten) behoben werden. Begleitende Aorteneingriffe bei Aneurysma oder Dissektion der thorakalen Aorta wurden ebenfalls durchgeführt. So wurden bei 78 Patienten ein partieller Aortenbogenersatz, bei 12 Patienten ein kompletter Aortenbogenersatz und bei 14 Patienten ein Bogenersatz mit „Elephant Trunk“ Technik durchgeführt.
Die Nachuntersuchungen erfolgten anhand eines standardisierten Fragebogens 5 sowie durch transthorakal echokardiographische Verlaufskontrollen. Die durchschnittliche Nachuntersuchungszeit betrug 6.3 ± 4.6 Jahre, 97% der Patienten konnten dabei erfasst werden.
Die 30-Tage-Mortalität betrug 1,4%. Im Langzeitverlauf wurden 132 Verstorbene beobachtet, wobei 13 Patienten aufgrund eines kardiovaskulären Ereignisses verstarben. Das 10-Jahres-Überleben betrug 70%. Eine Reoperation war bei 39 Patienten notwendig, in 25 Fällen aufgrund von signifikanter Restinsuffizienz der Aortenklappe, in 5 Fällen aufgrund eines kombinierten Aortenklappenvitium. Endokarditis führte in 9 Fällen zur Reoperation (0,2% pro Patientenjahr). Die Freiheit von Reoperation betrug 88% nach 10 Jahren. Die klappenbezogenen Komplikationen ergaben eine kumulative linearisierte Inzidenz von 2% pro Patientenjahr.
Schlussfolgernd kann man sagen, dass die Aortenklappenrekonstruktion für ausgewählte Patienten eine gute Alternative zum Aortenklappenersatz darstellt. Die Haltbarkeit der Klappenfunktion erwies sich in der Langzeitbeobachtung als gut. Die adäquate Wahl der Operationstechnik bezogen auf die Pathologie führen zu guten Ergebnissen der Aortenklappenrekonstruktion mit akzeptabler Mortalität und Morbidität im Langzeitverlauf.
Diplura is a group of entognathous hexapods, often considered a sister group to insects. They play an important role in recycling organic matter in soil and subterranean terrestrial ecosystems. The Campodeidae is the most diverse family, divided into four subfamilies. The subfamily Plusiocampinae has a subterranean life-style with many species distributed in the Euro-Mediterranean area. The incertae sedis tachycampoids ("lignée Tachycampoïde") is a group within the family Campodeidae that share with the Plusiocampinae a strong preference for subterranean habitats and several morphological characters, such as slender body shape, elongated appendages, considerable increment in the number of antennomeres and cercal articles, and complexity of sensorial structures. The present monograph provides a taxonomic revision of the subfamily Plusiocampinae and the genera belonging to the tachycampoid lineage from Europe and the Mediterranean region. It comprises detailed morphological descriptions and illustrations together with data on the habitats and distributions of 87 species, 10 subspecies and 11 affinis forms. Seven new species are described among those, namely: Plusiocampa (Plusiocampa) apollo Sendra, Giachino & Vailati sp. nov., P. (P.) chiosensis Sendra & Gasparo sp. nov., P. (P.) dublanskii Sendra & Turbanov sp. nov., P. (P.) hoffmanni Sendra & Paragamian sp. nov., P. (P.) rhea Sendra sp. nov., P. (P.) ternovensis Sendra & Borko sp. nov. and P. (Venetocampa) ferrani Sendra & Delić sp. nov.

Abstract
Objective: This retrospective study examined patients with probable Lennox-Gastaut syndrome (LGS) identified from German healthcare data.
Methods: This 10-year study (2007–2016) assessed healthcare insurance claims information from the Vilua Healthcare research database. A selection algorithm considering diagnoses and drug prescriptions identified patients with probable LGS. To increase the sensitivity of the identification algorithm, two populations were defined: all patients with probable LGS (broadly defined) and only those with a documented epilepsy diagnosis before 6 years of age (narrowly defined). This specific criterion was used as LGS typically has a peak seizure onset between age 3 and 5 years. Primary analyses were prevalence and demographics; secondary analyses included healthcare costs, hospitalization rate and length of stay (LOS), medication use, and mortality.
Results: In the final year of the study, 545 patients with broadly defined probable LGS (mean [range] age: 31.4 [2–89] years; male: 53%) were identified. Using the narrowly defined probable LGS definition, the number of patients was reduced to 102 (mean [range] age: 7.4 [2–14] years; male: 52%). Prevalence of broadly defined and narrowly defined probable LGS was 39.2 and 6.5 per 100,000 people. During the 10-year study, 208 patients with narrowly defined probable LGS were identified and followed up for 1379 patient-years. The mean annual cost of healthcare was €22,787 per patient-year (PPY); greatest costs were attributable to inpatient care (33%), home nursing care (13%), and medication (10%). Mean annual healthcare costs were significantly greater for those with prescribed rescue medication (45% of patient-years) versus those without (€33,872 vs. €13,785 PPY, p < 0.001). Mean (standard deviation [SD]) annual hospitalization rate was 1.6 (2.0) PPY with mean (SD) annual LOS of 22.7 (46.0) days. Annual hospitalization rate was significantly greater in those who were prescribed rescue medication versus those who were not (2.2 [2.3] vs. 1.1 [1.6] PPY, p < 0.001). The mean (SD) number of different medications prescribed was 11.3 (7.3) PPY and 33.8 (17.0) over the entire observable time per patient (OET); antiepileptic drugs only accounted for 2.1 (1.1) of the medications prescribed PPY and 3.8 (2.0) OET. Over the 10-year study period, mortality in patients with narrowly defined probable LGS was significantly higher than the matched control population (six events [2.88%] vs. one event [0.01%], p < 0.001).
Conclusion: Annual healthcare costs incurred by patients with probable LGS in Germany were substantial, and mostly attributable to inpatient care, home nursing care, and medication. Patients prescribed with rescue medication incurred significantly greater costs than those who were not. Patients with narrowly defined probable LGS had a higher mortality rate versus control populations.
Aim: Left ventricular non-compaction (LVNC) is perceived as a rare high-risk cardiomyopathy characterized by excess left ventricular (LV) trabeculation. However, there is increasing evidence contesting the clinical significance of LV hyper-trabeculation and the existence of LVNC as a distinct cardiomyopathy. The aim of this study is to assess the association of LV trabeculation extent with cardiovascular morbidity and all-cause mortality in patients undergoing clinical cardiac magnetic resonance (CMR) scans across 57 European centers from the EuroCMR registry.
Methods and Results: We studied 822 randomly selected cases from the EuroCMR registry. Image acquisition was according to international guidelines. We manually segmented images for LV chamber quantification and measurement of LV trabeculation (as per Petersen criteria). We report the association between LV trabeculation extent and important cardiovascular morbidities (stroke, atrial fibrillation, heart failure) and all-cause mortality prospectively recorded over 404 ± 82 days of follow-up. Maximal non-compaction to compaction ratio (NC/C) was mean (standard deviation) 1.81 ± 0.67, from these, 17% were above the threshold for hyper-trabeculation (NC/C > 2.3). LV trabeculation extent was not associated with increased risk of the defined outcomes (morbidities, mortality, LV CMR indices) in the whole cohort, or in sub-analyses of individuals without ischaemic heart disease, or those with NC/C > 2.3.
Conclusion: Among 882 patients undergoing clinical CMR, excess LV trabeculation was not associated with a range of important cardiovascular morbidities or all-cause mortality over ~12 months of prospective follow-up. These findings suggest that LV hyper-trabeculation alone is not an indicator for worse cardiovascular prognosis.
Evoked potentials in the amplitude-time spectrum of the electroencephalogram are commonly used to assess the extent of brain responses to stimulation with noxious contact heat. The magnitude of the N- and P-waves are used as a semi-objective measure of the response to the painful stimulus: the higher the magnitude, the more painful the stimulus has been perceived. The strength of the N-P-wave response is also largely dependent on the chosen reference electrode site. The goal of this study was to examine which reference technique excels both in practical and theoretical terms when analyzing noxious contact heat evoked potentials (CHEPS) in the amplitude-time spectrum. We recruited 21 subjects (10 male, 11 female, mean age of 55.79 years). We applied seven noxious contact heat stimuli using two temperatures, 51°C, and 54°C, to each subject. During EEG analysis, we aimed to identify the referencing technique which produces the highest N-wave and P-wave amplitudes with as little artifactual influence as possible. For this purpose, we applied the following six referencing techniques: mathematically linked A1/A2 (earlobes), average reference, REST, AFz, Pz, and mathematically linked PO7/PO8. We evaluated how these techniques impact the N-P amplitudes of CHEPS based on our data from healthy subjects. Considering all factors, we found that mathematically linked earlobes to be the ideal referencing site to use when displaying and evaluating CHEPS in the amplitude-time spectrum.
Recent studies have shown that the speciose Holarctic genera of Leiobunum C.L. Koch, 1839 and Nelima Roewer, 1910 are polyphyletic taxa, and therefore, the traditional diagnostic characters for these European genera of Leiobuninae Banks, 1893 (respectively, the presence or absence of tubercle rows on leg coxae) are unsuitable. We present the description of Leiolima iberica gen. et sp. nov., a new endemic harvestman from the north-western part of the Iberian Peninsula. The newly established genus shows intermediate characters between Leiobunum and Nelima. In addition, the new genus is characterized by shorter legs compared to Leiobunum and the presence of trichomes on all leg femora and pedipalpal patellae, a structure that is absent in all other western Palearctic genera of the subfamily Leiobuninae.
Recent samplings in cavities of the Iberian Peninsula led to the discovery of a new millipede species of the order Callipodida in one cave in the region of El Bierzo (León, Spain): Lusitanipus xanin sp. nov. In additional samplings (near that cave), more specimens of this new species were discovered in surface habitats. The species has various distinctive characteristics, such as its green colour, but the morphology of the gonopod has several similarities with that of Lusitanipus alternans (Verhoeff, 1893), indicating a close relationship between them. The diagnosis of the genus Lusitanipus Mauriès, 1978 has to be changed to accommodate the new species. The presence of crests in two different sizes is no longer a diagnostic character of the genus Lusitanipus, but of the species L. alternans. Lusitanipus xanin sp. nov. is the fourth species of the order Callipodida for the Iberian Peninsula, and the second species for the genus. Lusitanipus xanin sp. nov. seems to be an epigean but troglophile species.
Consuming dividends

This paper studies why investors buy dividend-paying assets and how they time their consumption accordingly. We combine administrative bank data linking customers’ consumption transactions and income to detailed portfolio data and survey responses on financial behavior. We find that private consumption is excessively sensitive to dividend income. Investors across wealth, income, and age distributions increase spending precisely around days of dividend receipt. Importantly, the consumption response is driven by financially prudent investors who select dividend portfolios, anticipate dividend income, and plan consumption accordingly. Our results contribute to the literature on a dividend clientele and provide evidence of ‘planned’ excess sensitivity.
This article demonstrates the use of guided elastic waves (GEW) for multiple-in and multiple-out (MIMO) data communication in the framework of a structural health monitoring (SHM) system. Therefore, miniaturized low-voltage communication nodes have been developed. They are arranged in a spatially distributed and permanently installed network. Wireless exchange of encoded information across a metallic plate and a stiffened carbon-fiber reinforced plastics (CFRP) structure is investigated. A combination of square-wave excitation sequences and frequency-division multiplexing (FDM) is explored for parallel communication with multiple nodes. Moreover, the impact of the excitation-sequence length on the reliability of information transmission is studied in view of future energy-aware application scenarios. The presented system achieves in both studied structures error-free transmission at a data rate of 0.17 kbps (per carrier frequency) with a power consumption of 224 mW.
The third dataset dedicated to the Open Guided Waves platform aims at carbon fiber composite plates with an additional omega stringer at constant temperature conditions. The two structures used in this work are representative for real aircraft components. Comprehensive measurements were recorded in order to study (I) the impact of the omega stringer on guided wave propagation, and (II) elliptical reference damages of different sizes located at three separate positions on the structure. Measurements were recorded for narrowband excitation (5-cycle toneburst with varying carrier frequencies) and broadband excitation (using chirp waveforms). The paper presents the results of a technical validation including numerical modelling, and enables further research, for example related to probability of detection (POD) analysis.
A central motivation for the development of x-ray free-electron lasers has been the prospect of time-resolved single-molecule imaging with atomic resolution. Here, we show that x-ray photoelectron diffraction—where a photoelectron emitted after x-ray absorption illuminates the molecular structure from within—can be used to image the increase of the internuclear distance during the x-ray-induced fragmentation of an O2 molecule. By measuring the molecular-frame photoelectron emission patterns for a two-photon sequential K-shell ionization in coincidence with the fragment ions, and by sorting the data as a function of the measured kinetic energy release, we can resolve the elongation of the molecular bond by approximately 1.2 a.u. within the duration of the x-ray pulse. The experiment paves the road toward time-resolved pump-probe photoelectron diffraction imaging at high-repetition-rate x-ray free-electron lasers.
The retinoid X receptor (RXR) is a ligand-sensing transcription factor acting mainly as a universal heterodimer partner for other nuclear receptors. Despite presenting as a potential therapeutic target for cancer and neurodegeneration, adverse effects typically observed for RXR agonists, likely due to the lack of isoform selectivity, limit chemotherapeutic application of currently available RXR ligands. The three human RXR isoforms exhibit different expression patterns; however, they share high sequence similarity, presenting a major obstacle toward the development of subtype-selective ligands. Here, we report the discovery of the saturated fatty acid, palmitic acid, as an RXR ligand and disclose a uniform set of crystal structures of all three RXR isoforms in an active conformation induced by palmitic acid. A structural comparison revealed subtle differences among the RXR subtypes. We also observed an ability of palmitic acid as well as myristic acid and stearic acid to induce recruitment of steroid receptor co-activator 1 to the RXR ligand-binding domain with low micromolar potencies. With the high, millimolar endogenous concentrations of these highly abundant lipids, our results suggest their potential involvement in RXR signaling.
A new species of Limnophilinae (Diptera: Limoniidae), Dicranophragma (Brachylimnophila) relictum Mederos sp. nov., is described. This is the third species of this genus recorded from the Iberian Peninsula. This new species was discovered in a cave in the Sant Llorenç del Munt i Serra de l’Obac Natural Park, Barcelona Province (Catalonia) and it is the first record of a species of this genus from a hypogeous environment. The absence of suitable ecological conditions for the survival of this new species, particularly in the vicinity of the cave, suggests that this population of D. relictum Mederos sp. nov. is isolated. Dicranophragma relictum Mederos sp. nov. is characterized by the following features: a general grey-to-greenish-grey coloration on its thorax and a dark-grey abdomen; wings more than four times longer than wide; pale-brown-to-yellowish tinted wings, with brown veins and a well-marked stigma; in the male genitalia the upper part of the aedeagus is acute when viewed laterally and is longer than the lower part. A key is provided for separating the three species of Dicranophragma known to be present in the Iberian Peninsula.
Non-alcoholic steatohepatitis (NASH) - a hepatic manifestation of the metabolic syndrome - is a multifactorial disease with alarming global prevalence. It involves steatosis, inflammation and fibrosis in the liver, thus demanding multiple modes of action for robust therapeutic efficacy. Aiming to fuse complementary validated anti-NASH strategies in a single molecule, we have designed and systematically optimized a scaffold for triple activation of farnesoid X receptor (FXR), peroxisome proliferator-activated receptor (PPAR) α and PPARδ. Pilot profiling of the resulting triple modulator demonstrated target engagement in native cellular settings and in mice, rendering it a suitable tool to probe the triple modulator concept in vivo. In DIO NASH in mice, the triple agonist counteracted hepatic inflammation and reversed hepatic fibrosis highlighting the potential of designed polypharmacology in NASH.
Introduction: Esophageal atresia with or without tracheoesophageal fistula (EA/TEF) occurs approximately 1 in 3.500 live births representing the most common malformation of the upper digestive tract. Only half a century ago, EA/TEF was fatal among affected newborns suggesting that the steady birth prevalence might in parts be due to mutational de novo events in genes involved in foregut development.
Methods: To identify mutational de novo events in EA/TEF patients, we surveyed the exome of 30 case-parent trios. Identified and confirmed de novo variants were prioritized using in silico prediction tools. To investigate the embryonic role of genes harboring prioritized de novo variants we performed targeted analysis of mouse transcriptome data of esophageal tissue obtained at the embryonic day (E) E8.5, E12.5, and postnatal.
Results: In total we prioritized 14 novel de novo variants in 14 different genes (APOL2, EEF1D, CHD7, FANCB, GGT6, KIAA0556, NFX1, NPR2, PIGC, SLC5A2, TANC2, TRPS1, UBA3, and ZFHX3) and eight rare de novo variants in eight additional genes (CELSR1, CLP1, GPR133, HPS3, MTA3, PLEC, STAB1, and PPIP5K2). Through personal communication during the project, we identified an additional EA/TEF case-parent trio with a rare de novo variant in ZFHX3. In silico prediction analysis of the identified variants and comparative analysis of mouse transcriptome data of esophageal tissue obtained at E8.5, E12.5, and postnatal prioritized CHD7, TRPS1, and ZFHX3 as EA/TEF candidate genes. Re-sequencing of ZFHX3 in additional 192 EA/TEF patients did not identify further putative EA/TEF-associated variants.
Conclusion: Our study suggests that rare mutational de novo events in genes involved in foregut development contribute to the development of EA/TEF.
This article studies whether people want to control what information on their own past pro-social behavior is revealed to others. Participants are assigned a color that depends on their past pro-social behavior. They can spend money to manipulate the probability with which their color is revealed to another participant. The data show that participants are more likely to reveal colors with more favorable informational content. This pattern is not found in a control treatment in which colors are randomly assigned, thus revealing nothing about past pro-social behavior. Regression analysis confrms these fndings, also when controlling for past pro-social behavior. These results complement the existing empirical evidence, confrming that people strategically and, therefore, consciously manipulate their social image.
The RHO gene encodes the G-protein-coupled receptor (GPCR) rhodopsin. Numerous mutations associated with impaired visual cycle have been reported; the G90D mutation leads to a constitutively active mutant form of rhodopsin that causes CSNB disease. We report on the structural investigation of the retinal configuration and conformation in the binding pocket in the dark and light-activated state by solution and MAS-NMR spectroscopy. We found two long-lived dark states for the G90D mutant with the 11-cis retinal bound as Schiff base in both populations. The second minor population in the dark state is attributed to a slight shift in conformation of the covalently bound 11-cis retinal caused by the mutation-induced distortion on the salt bridge formation in the binding pocket. Time-resolved UV/Vis spectroscopy was used to monitor the functional dynamics of the G90D mutant rhodopsin for all relevant time scales of the photocycle. The G90D mutant retains its conformational heterogeneity during the photocycle.
Investigation of the linear and mode-coupled flow harmonics in Au+Au collisions at √sNN = 200 GeV

Flow harmonics (vn) of the Fourier expansion for the azimuthal distributions of hadrons are commonly employed to quantify the azimuthal anisotropy of particle production relative to the collision symmetry planes. While lower order Fourier coefficients (v2 and v3) are more directly related to the corresponding eccentricities of the initial state, the higher-order flow harmonics (vn>3) can be induced by a modecoupled response to the lower-order anisotropies, in addition to a linear response to the same-order anisotropies. These higher-order flow harmonics and their linear and mode-coupled contributions can be used to more precisely constrain the initial conditions and the transport properties of the medium in theoretical models. The multiparticle azimuthal cumulant method is used to measure the linear and mode-coupled contributions in the higher-order anisotropic flow, the mode-coupled response coefficients, and the correlations of the event plane angles for charged particles as functions of centrality and transverse momentum in Au+Au collisions at nucleon-nucleon center-of-mass energy √sN N= 200 GeV. The results are compared to similar LHC measurements as well as to several viscous hydrodynamic calculations with varying initial conditions.
Measurement of groomed jet substructure observables in p+p collisions at √s = 200 GeV with STAR

In this letter, measurements of the shared momentum fraction (zg) and the groomed jet radius (Rg), as defined in the SoftDrop algorithm, are reported in p+p collisions at √s = 200 GeV collected by the STAR experiment. These substructure observables are differentially measured for jets of varying resolution parameters from R = 0.2 − 0.6 in the transverse momentum range 15 < pT,jet < 60 GeV/c. These studies show that, in the pT,jet range accessible at √s = 200 GeV and with increasing jet resolution parameter and jet transverse momentum, the zg distribution asymptotically converges to the DGLAP splitting kernel for a quark radiating a gluon. The groomed jet radius measurements reflect a momentum-dependent narrowing of the jet structure for jets of a given resolution parameter, i.e., the larger the pT,jet, the narrower the first splitting. For the first time, these fully corrected measurements are compared to Monte Carlo generators with leading order QCD matrix elements and leading log in the parton shower, and to state-of-the-art theoretical calculations at next-to-leading-log accuracy. We observe that PYTHIA 6 with parameters tuned to reproduce RHIC measurements is able to quantitatively describe data, whereas PYTHIA 8 and HERWIG 7, tuned to reproduce LHC data, are unable to provide a simultaneous description of both zg and Rg, resulting in opportunities for fine parameter tuning of these models for p+p collisions at RHIC energies. We also find that the theoretical calculations without non-perturbative corrections are able to qualitatively describe the trend in data for jets of large resolution parameters at high pT,jet, but fail at small jet resolution parameters and low jet transverse momenta.
We report results on the total and elastic cross sections in proton-proton collisions at √s = 200 GeV obtained with the Roman Pot setup of the STAR experiment at the Relativistic Heavy Ion Collider (RHIC). The elastic differential cross section was measured in the squared four-momentum transfer range 0.045 ≤ −t ≤ 0.135 GeV2. The value of the exponential slope parameter B of the elastic differential cross section dσ/dt ∼ e−Bt in the measured −t range was found to be B = 14.32 ± 0.09(stat.)+0.13 −0.28(syst.) GeV−2. The total cross section σtot, obtained from extrapolation of the dσ/dt to the optical point at −t = 0, is σtot = 54.67 ± 0.21(stat.)+1.28 −1.38(syst.) mb. We also present the values of the elastic cross section σel = 10.85 ± 0.03(stat.)+0..49 −0.41(syst.) mb, the elastic cross section integrated within the STAR t-range σ det el = 4.05 ± 0.01(stat.)+0.18−0.17(syst.) mb, and the inelastic cross section σinel = 43.82 ± 0.21(stat.)+1.37−1.44(syst.) mb. The results are compared with the world data
Acinetobacter baumannii is an opportunistic pathogen, which has become a rising threat in healthcare facilities worldwide due to increasing antibiotic resistances and optimal adaptation to clinical environments and the human host. We reported in a former publication on the identification of three phopholipases of the phospholipase D (PLD) superfamily in A. baumannii ATCC 19606T acting in concerted manner as virulence factors in Galleria mellonella infection and lung epithelial cell invasion. This study focussed on the function of the three PLDs. A Δpld1-3 mutant was defect in biosynthesis of the phospholipids cardiolipin (CL) and monolysocardiolipin (MLCL), whereas the deletion of pld2 and pld3 abolished the production of MLCL. Complementation of the Δpld1-3 mutant with pld1 restored CL biosynthesis demonstrating that the PLD1 is implicated in CL biosynthesis. Complementation of the Δpld1-3 mutant with either pld2 or pld3 restored MLCL and CL production leading to the conclusion that PLD2 and PLD3 are implicated in CL and MLCL production. Mutant studies revealed that two catalytic motifs are essential for the PLD3-mediated biosynthesis of CL and MLCL. The Δpld1-3 mutant exhibited a decreased colistin and polymyxin B resistance indicating a role of CL in cationic antimicrobial peptides (CAMPs) resistance.
The ruling of the German Federal Constitutional Court and its call for conducting and communicating proportionality assessments regarding monetary policy have been the subject of some controversy. However, it can also be understood as a way to strengthen the de-facto independence of the European Central Bank. The authors shows how a regular proportionality check could be integrated in the ECB’s strategy that is currently undergoing a systematic review. In particular, they propose to include quantitative benchmarks for policy rates and the central bank balance sheet. Deviations from such benchmarks can have benefits in terms of the intended path for inflation while involving costs in terms of risks and side effects that need to be balanced. Background & Aims: Spontaneous portosystemic shunts (SPSS) frequently develop in liver cirrhosis. Recent data suggested that the presence of a single large SPSS is associated with complications, especially overt hepatic encephalopathy (oHE). However, the presence of >1 SPSS is common. This study evaluates the impact of total cross-sectional SPSS area (TSA) on outcomes in patients with liver cirrhosis.
Methods: In this retrospective international multicentric study, CT scans of 908 cirrhotic patients with SPSS were evaluated for TSA. Clinical and laboratory data were recorded. Each detected SPSS radius was measured and TSA calculated. One-year survival was the primary endpoint and acute decompensation (oHE, variceal bleeding, ascites) was the secondary endpoint.
Results: A total of 301 patients (169 male) were included in the training cohort. Thirty percent of all patients presented with >1 SPSS. A TSA cut-off of 83 mm2 was used to classify patients with small or large TSA (S-/L-TSA). Patients with L-TSA presented with higher model for end-stage liver disease score (11 vs. 14) and more commonly had a history of oHE (12% vs. 21%, p <0.05). During follow-up, patients with L-TSA experienced more oHE episodes (33% vs. 47%, p <0.05) and had lower 1-year survival than those with S-TSA (84% vs. 69%, p <0.001). Multivariate analysis identified L-TSA (hazard ratio 1.66; 95% CI 1.02–2.70, p <0.05) as an independent predictor of mortality. An independent multicentric validation cohort of 607 patients confirmed that patients with L-TSA had lower 1-year survival (77% vs. 64%, p <0.001) and more oHE development (35% vs. 49%, p <0.001) than those with S-TSA.
Conclusion: This study suggests that TSA >83 mm2 increases the risk for oHE and mortality in patients with cirrhosis. Our results support the clinical use of TSA/SPSS for risk stratification and decision-making in the management of patients with cirrhosis.
Lay summary: The prevalence of spontaneous portosystemic shunts (SPSS) is higher in patients with more advanced chronic liver disease. The presence of more than 1 SPSS is common in advanced chronic liver disease and is associated with the development of hepatic encephalopathy. This study shows that total cross-sectional SPSS area (rather than diameter of the single largest SPSS) predicts survival in patients with advanced chronic liver disease. Our results support the clinical use of total cross-sectional SPSS area for risk stratification and decision-making in the management of SPSS.
Long non-coding RNAs (lncRNAs) contribute to cardiac (patho)physiology. Aging is the major risk factor for cardiovascular disease with cardiomyocyte apoptosis as one underlying cause. Here, we report the identification of the aging-regulated lncRNA Sarrah (ENSMUST00000140003) that is anti-apoptotic in cardiomyocytes. Importantly, loss of SARRAH (OXCT1-AS1) in human engineered heart tissue results in impaired contractile force development. SARRAH directly binds to the promoters of genes downregulated after SARRAH silencing via RNA-DNA triple helix formation and cardiomyocytes lacking the triple helix forming domain of Sarrah show an increase in apoptosis. One of the direct SARRAH targets is NRF2, and restoration of NRF2 levels after SARRAH silencing partially rescues the reduction in cell viability. Overexpression of Sarrah in mice shows better recovery of cardiac contractile function after AMI compared to control mice. In summary, we identified the anti-apoptotic evolutionary conserved lncRNA Sarrah, which is downregulated by aging, as a regulator of cardiomyocyte survival.
Fruiting body-forming members of the Basidiomycota maintain their ecological fitness against various antagonists like ascomycetous mycoparasites. To achieve that, they produce myriads of bioactive compounds, some of which are now being used as agrochemicals or pharmaceutical lead structures. Here, we screened ethyl acetate crude extracts from cultures of thirty-five mushroom species for antifungal bioactivity, for their effect on the ascomycete Saccharomyces cerevisiae and the basidiomycete Ustilago maydis. One extract that inhibited the growth of S. cerevisiae much stronger than that of U. maydis was further analyzed. For bioactive compound identification, we performed bioactivity-guided HPLC/MS fractionation. Fractions showing inhibition against S. cerevisiae but reduced activity against U. maydis were further analyzed. NMR-based structure elucidation from one such fraction revealed the polyyne we named feldin, which displays prominent antifungal bioactivity. Future studies with additional mushroom-derived eukaryotic toxic compounds or antifungals will show whether U. maydis could be used as a suitable host to shortcut an otherwise laborious production of such mushroom compounds, as could recently be shown for heterologous sesquiterpene production in U. maydis.
Space optimizations in deterministic and concurrent call-by-need functional programming languages

In this thesis the space consumption and runtime of lazy-evaluating functional programming languages are analyzed.
The typed and extended lambda-calculi LRP and CHF* as core languages for Haskell and Concurrent Haskell are used. For each LRP and CHF* compatible abstract machines are introduced.
Too lower the distortion of space measurement a classical implementable garbage collector is applied after each LRP reduction step. Die size of expressions and the space measure spmax as maximal size of all garbage-free expressions during an LRP-evaluation, are defined.
Program-Transformations are considered as code-to-code transformations. The notions Space Improvement and Space Equivalence as properties of transformations are defined. A Space Improvement does neither change the semantics nor it increases the needed space consumption, for a space equivalence the space consumption is required to remain the same. Several transformations are shown as Space Improvements and Equivalences.
An abstract machine for space measurements is introduced. An implementation of this machine is used for more complex space- and runtime-analyses.
Total Garbage Collection replaces subexpressions by a non-terminating constant with size zero, if the overall termination is not affected. Thereby the notion of improvement is more independent from the used garbage collector.
Analogous to Space Improvements and Equivalences the notions Total Space Improvement and Total Space Equivalence are defined, which use Total Garbage Collection during the space measurement. Several Total Space Improvements and Equivalences are shown.
Space measures for CHF* are defined, that are compatible to the space measure of LRP. An algorithm with sort-complexity is developed, that calculates the required space of independent processes that all start and end together. If a constant amount of synchronization restrictions is added and a constant number of processors is used, the runtime is polynomial, if arbitrary synchronizations are used, then the problem is NP-complete.
Abstract machines for space- and time-analyses in CHF* are developed and implementations of these are used for space and runtime analyses.
We study whether and how time preferences change over the life cycle, exploiting representative long-term panel data. We estimate the age patterns of discount rates from age 25 to 80. In order to identify age effects, we have to disentangle them from cohort and period factors. We address this identification problem by estimating individual fixed effects models, where we substitute period effects with determinants of time preferences that depend on calendar years. We find that discount rates decrease with age and the decline is remarkably linear over the life cycle.
Hintergrund und Ziel der Studie: Steigende Erwartungen der Kataraktpatienten führen zu stetigen Weiterentwicklungen in der Linsenchirurgie. Der Wunsch nach einem perfekten Sehvermögen und nach Brillenfreiheit ist ein häufiges Anliegen insbesondere jüngerer Patienten. Die Einführung von Multifokallinsen hat die Therapie der Katarakt revolutioniert. Mit ihnen kann der Patient sowohl in der Nähe und Ferne als auch im Intermediärbereich scharf sehen. Für die präoperative Berechnung der Linsenstärke sind verschiedene Formeln verfügbar. Bis jetzt wurde keine Studie publiziert, die die Präzision dieser Formeln für die Kalkulation der Stärke von Tri- oder Quadrifokallinsen untersucht hat. Das Ziel dieser Studie war die Evaluation neun moderner Formeln für die Berechnung der Linsenstärke der quadrifokalen AcrySof IQ PanOptix TFNT00.
Methoden:
Die vorliegende Studie ist eine retrospektive konsekutive Fallserie, die an der Augenklinik der Johann Wolfgang Goethe-Universität Frankfurt am Main durchgeführt wurde. Es wurden alle Patienten eingeschlossen, die sich einer Femtosekundenlaser-assistierten Operation mit Implantation der quadrifokalen Intraokularlinse unterzogen. Mit dem IOLMaster 500 wurden die präoperativen Biometrie- und Keratometriemessungen durchgeführt. Das Scheimpflug-System Pentacam wurde zur Bestimmung der zentralen Hornhautdicke genutzt. Drei Monate nach der Operation fand eine subjektive Refraktionsbestimmung statt.
Die folgenden Formeln wurden untersucht: Holladay 1, Sanders-Retzlaff-Kraff/ theoretical (SRK/T), Hoffer Q, T2, Holladay 2, Haigis, Barrett Universal II, Olsen und Hill-radiale Basisfunktion (RBF). Aus den präoperativ gemessenen Parametern und der implantierten Linsenstärke wurde für jedes Auge die postoperative Refraktion berechnet. Der Vorhersagefehler (PE) ist definiert als die Differenz zwischen dem tatsächlich erreichten postoperativen sphärischen Äquivalent und dem vorhergesagten postoperativen sphärischen Äquivalent. Zur Reduktion systematischer Fehler und somit des mittleren PE wurden die Linsenkonstanten optimiert. Primäre Endpunkte waren die Unterschiede der mittleren absoluten Vorhersagefehler (MAE) zwischen den Formeln. Die Standardabweichung sowie mediane und maximale absolute PE wurden ebenso untersucht wie die Prozent-zahlen der Augen, deren PE innerhalb von ± 0,25 Dioptrien (dpt), ± 0,5 dpt, ± 1,0 dpt und ± 2,0 dpt lagen.
Ergebnisse: 75 Augen von 38 Patienten wurden in die vorliegende Studie eingeschlossen. Es gab signifikante Unterschiede zwischen den Formeln bezüglich des MAE (p Wert < 0,001). Die Rangfolge der Formeln entsprechend ihrer MAE lautet wie folgt: Barrett Universal II (0,294 dpt), Hill-RBF (0,332 dpt), Olsen (0,339 dpt), T2 (0,351 dpt), Holladay 1 (0,381 dpt), Haigis (0,382 dpt), SRK/T (0,393 dpt), Holladay 2 (0,399 dpt) und Hoffer Q (0,410 dpt). Den niedrigsten maximalen absoluten PE erreichte die Barrett Universal II. Bei der Verwendung der Haigis, Barrett Universal II, Olsen und Hill-RBF hatten 80% der Augen einen PE innerhalb von ± 0,5 dpt. In der Subgruppe der kurzen Augen wurde der niedrigste PE mit der Hill-RBF erreicht, in der Subgruppe der langen Augen mit der Barrett Universal II und mit der Olsen. Die Unterschiede zwischen den Formeln in den Subgruppenanalysen waren jedoch nicht signifikant.
Schlussfolgerung:Die exaktesten Vorhersagen der postoperativen Refraktion konnten mit der Barrett Universal II erzielt werden. Diese Formel sollte daher künftig für die Berechnung der Linsenstärke der quadrifokalen PanOptix genutzt werden. Die T2, Olsen und Hill-RBF führten ebenfalls zu niedrigen PE. Weitere Studien sollten durchgeführt werden, um insbesondere die neuen Formeln im Zusammenhang mit verschiedenen Multifokallinsen zu untersuchen. Um die Vorhersagegenauigkeit der Formeln in Augen mit extremen Achsenlängen zu bewerten, sind Studien mit größeren Fallzahlen nötig.
The branchial parasitic isopod Pleurocryptella altalis sp. nov. (Bopyridae: Pseudioninae) is described from the squat lobster host Munidopsis petalorhyncha Baba, 2005. The new species is morphologically similar to Pleurocryptella formosa Bonnier, 1900 and P. wolffi Bourdon, 1972b but can be distinguished based on male characters (differences in head, pleon and uropods) and female characters (differences in barbula, pleopods and pleotelson). The parasite specimens (a female and male pair) were collected with the squat lobster host at a depth of 5060–5130 m from the Kuril-Kamchatka Trench, representing the deepest record for any of the 850+ described bopyrid isopod species and for any record of an infested host. Dichotomous identification keys to females and males of Pleurocryptella species and subspecies are provided.
CD4+ T cell lymphopenia predicts mortality from Pneumocystis pneumonia in kidney transplant patients

Background: Pneumocystis jirovecii pneumonia (PcP) remains a life-threatening opportunistic infection after solid organ transplantation, even in the era of Pneumocystis prophylaxis. The association between risk of developing PcP and low CD4+ T cell counts has been well established. However, it is unknown whether lymphopenia in the context of post-renal transplant PcP increases the risk of mortality. Methods: We carried out a retrospective analysis of a cohort of kidney transplant patients with PcP (n = 49) to determine the risk factors for mortality associated with PcP. We correlated clinical and demographic data with the outcome of the disease. For CD4+ T cell counts, we used the Wilcoxon rank sum test for in-hospital mortality and a Cox proportional-hazards regression model for 60-day mortality. Results: In univariate analyses, high CRP, high neutrophils, CD4+ T cell lymphopenia, mechanical ventilation, and high acute kidney injury network stage were associated with in-hospital mortality following presentation with PcP. In a receiver-operator characteristic (ROC) analysis, an optimum cutoff of ≤200 CD4+ T cells/µL predicted in-hospital mortality, CD4+ T cell lymphopenia remained a risk factor in a Cox regression model. Conclusions: Low CD4+ T cell count in kidney transplant recipients is a biomarker for disease severity and a risk factor for in-hospital mortality following presentation with PcP.
The miRNA biogenesis is tightly regulated to avoid dysfunction and consequent disease development. Here, we describe modulation of miRNA processing as a novel noncanonical function of the 5-lipoxygenase (5-LO) enzyme in monocytic cells. In differentiated Mono Mac 6 (MM6) cells, we found an in situ interaction of 5-LO with Dicer, a key enzyme in miRNA biogenesis. RNA sequencing of small noncoding RNAs revealed a functional impact, knockout of 5-LO altered the expression profile of several miRNAs. Effects of 5-LO could be observed at two levels. qPCR analyses thus indicated that (a) 5-LO promotes the transcription of the evolutionarily conserved miR-99b/let-7e/miR-125a cluster and (b) the 5-LO-Dicer interaction downregulates the processing of pre-let-7e, resulting in an increase in miR-125a and miR-99b levels by 5-LO without concomitant changes in let-7e levels in differentiated MM6 cells. Our observations suggest that 5-LO regulates the miRNA profile by modulating the Dicer-mediated processing of distinct pre-miRNAs. 5-LO inhibits the formation of let-7e which is a well-known inducer of cell differentiation, but promotes the generation of miR-99b and miR-125a known to induce cell proliferation and the maintenance of leukemic stem cell functions.
An integrative redescription of Hypsibius pallidoides Pilato, Kiosya, Lisi, Inshina & Biserov, 2011 was undertaken following a reexamination of the type material and new material using highquality light microscopy, scanning electron microscopy and methods of molecular taxonomy. Detailed morphological investigations revealed a unique complex of characters that precluded the attribution of this species to the genus Hypsibius Ehrenberg, 1848. Furthermore, phylogenetic analyses indicated the affi nity of this species within the subfamily Pilatobiinae (Hypsibiidae). Notahypsibius gen. nov. is erected for H. pallidoides and two putatively related species: H. scaber Maucci, 1987 and Ramazzottius arcticus (Murray, 1907). An emended diagnosis for the genus Pilatobius is given, while the subfamily Pilatobiinae lacks a cohesive morphological diagnosis despite representing, at the same time, a wellsupported molecular clade. Obvious controversy between the results of the morphological and molecular analyses of the phylogeny of Hypsibioidea is discussed. The distribution of morphological characters such as the claw type, organization of the bucco-pharyngeal apparatus, and egg shell sculpture type within Eutardigrada is analyzed and their phylogenetic signifi cance discussed.
In this paper we describe Macrobiotus engbergi sp. nov. and Tenuibiotus zandrae sp. nov. from Greenland. Our study has involved both classical taxonomic methods, which include morphological and morphometric analyses conducted with the use of light and scanning electron microscopy, and genetic analysis based on four molecular markers (three nuclear: 18S rRNA, 28S rRNA, ITS-2, and one mitochondrial: COI). Moreover, we re-examined the type series of Tenuibiotus voronkovi (Tumanov, 2007) as well as the original sample where the species was found and we provide new morphological data from light and scanning electron microscopy which enabled us to amend its description. Finally, we also analysed slides with animals and egg of two populations from Nordaustlandet and Edgeøya (archipelago of Svalbard, Norway) designated as T. voronkovi within its recent redescription. The results and comparisons presented in our study question the validity of this designation.
The widespread Ophioderma hendleri sp. nov., from the Eastern Tropical Pacific (Mexico to Colombia) is distinguished from its congeners by having radial shields covered by granules, naked adoral shields, up to 11 arm spines, and by its brown and beige coloration. Ophioderma hendleri sp. nov. belongs to the group of species with naked adoral shields (i.e., O. pentacanthum H.L. Clark, 1917, O. variegatum Lütken, 1856), and it has frequently been misidentified as O. panamense Lütken, 1859 or O. variegatum. Therefore, the main aim of the present work was to describe Ophioderma hendleri sp. nov. and differentiate it from its congeners. The original description of O. panamense was incomplete; thus, we provide a redescription. Due to the confusion in previous designations of its type material, we designate a lectotype and paralectotype of O. variegatum. Finally, we expand the distribution range of O. pentacanthum to Cocos Island, Costa Rica. With this work, the total number of valid species of Ophioderma Müller & Troschel, 1840 in the world increases to 33 and in the Eastern Pacific to nine species.
A new species of the genus Mesobiotus is described from the Republic of South Africa using a traditional morphological approach (light and scanning electron microscopy) combined with molecular analysis (18S rRNA, 28S rRNA, ITS-2 and COI markers). Mesobiotus anastasiae sp. nov. differs from all known Mesobiotus species by having a unique combination of characters of the adult animals and the eggs. Adults of the new species have an oral cavity armature without elongate teeth in the second band, while the processes of the egg chorion have a basal collar and distinct rows of large pores. An updated key to the species of the genus Mesobiotus, including 66 of 70 currently described species, is given. An aquatic mite species from the Lobohalacarus weberi complex (freshwater Halacaridae) co-occurs with M. anastasiae sp. nov., suggesting that the newly described tardigrade inhabits constantly wet moss cushion habitats.
Pathogenic variants in PRRT2, encoding the proline-rich transmembrane protein 2, have been associated with an evolving spectrum of paroxysmal neurologic disorders. Based on a cohort of children with PRRT2-related infantile epilepsy, this study aimed at delineating the broad clinical spectrum of PRRT2-associated phenotypes in these children and their relatives. Only a few recent larger cohort studies are on record and findings from single reports were not confirmed so far. We collected detailed genetic and phenotypic data of 40 previously unreported patients from 36 families. All patients had benign infantile epilepsy and harbored pathogenic variants in PRRT2 (core cohort). Clinical data of 62 family members were included, comprising a cohort of 102 individuals (extended cohort) with PRRT2-associated neurological disease. Additional phenotypes in the cohort of patients with benign sporadic and familial infantile epilepsy consist of movement disorders with paroxysmal kinesigenic dyskinesia in six patients, infantile-onset movement disorders in 2 of 40 individuals, and episodic ataxia after mild head trauma in one girl with bi-allelic variants in PRRT2. The same girl displayed a focal cortical dysplasia upon brain imaging. Familial hemiplegic migraine and migraine with aura were reported in nine families. A single individual developed epilepsy with continuous spikes and waves during sleep. In addition to known variants, we report the novel variant c.843G>T, p.(Trp281Cys) that co-segregated with benign infantile epilepsy and migraine in one family. Our study highlights the variability of clinical presentations of patients harboring pathogenic PRRT2 variants and expands the associated phenotypic spectrum.
The specific temporal evolution of bacterial and phage population sizes, in particular bacterial depletion and the emergence of a resistant bacterial population, can be seen as a kinetic fingerprint that depends on the manifold interactions of the specific phage–host pair during the course of infection. We have elaborated such a kinetic fingerprint for a human urinary tract Klebsiella pneumoniae isolate and its phage vB_KpnP_Lessing by a modeling approach based on data from in vitro co-culture. We found a faster depletion of the initially sensitive bacterial population than expected from simple mass action kinetics. A possible explanation for the rapid decline of the bacterial population is a synergistic interaction of phages which can be a favorable feature for phage therapies. In addition to this interaction characteristic, analysis of the kinetic fingerprint of this bacteria and phage combination revealed several relevant aspects of their population dynamics: A reduction of the bacterial concentration can be achieved only at high multiplicity of infection whereas bacterial extinction is hardly accomplished. Furthermore the binding affinity of the phage to bacteria is identified as one of the most crucial parameters for the reduction of the bacterial population size. Thus, kinetic fingerprinting can be used to infer phage–host interactions and to explore emergent dynamics which facilitates a rational design of phage therapies.
This Ph.D. thesis demonstrates i) the highly precise performance of refined and new analytical setups for clumped isotope analysis (Δ47 and Δ48) and ii) the applicability of clumped isotope analyses to biogenic and abiogenic carbonated apatite (Δ47) and abiogenic carbonates (Δ47 and Δ48) for research related to paleothermophysiology and paleoclimatology, whereas the overall analytical precision has been increased.
A comprehensive Δ47 dataset with 122 replicate analyses is provided from which the temperature dependence of Δ47 for (bio)apatite (Δ47-1/T2) is calculated between 1 °C and 80 °C. The temperature dependence of oxygen isotope equilibrium fractionation between carbonated synthetic apatite and water (1,000ln(αCHAP-H2O)) is experimentally determined. When applied to tooth enameloid from a modern Greenland shark (Somniosus microcephalus), a Late Miocene megatooth shark (Carcharodon megalodon), and an Upper Cretaceous Tyrannosaurus rex, reconstructed Δ47-based temperatures and δ18OH2O are in line with previously published data.
An analytical setup for highly precise clumped isotope analysis is described that allows for the simultaneous measurement of ∆47 and ∆48 in CO2 with external reproducibilities close to the respective shot-noise limits. The analyte gases originate from pure carbonates that were digested in hypersaturated orthophosphoric acid and purified using a fully automated device. Δ47 data sets with 117 replicate analyses in total on 22 pedogenic carbonate nodules from two Spanish Middle Miocene sections reveal the continental Southern European thermal structure during the end of the Middle Miocene Climatic Optimum (MCO) and the complete Middle Miocene Climatic Transition (MMCT; from 15.33 to 12.98 Ma).
Astrozyten erfüllen verschiedene Funktionen im Zentralnervensystem, welche sich in die Bereiche Entwicklung, Durchblutung, Metabolismus, Strukturerhalt und Gliotransmission unterteilen lassen. Astrozyten sind an der synaptischen Informationsverarbeitung beteiligt und wirken an zahlreichen höheren Hirnfunktionen mit. Durch Regulation der synaptischen Transmission und Plastizität sind Astrozyten am Lernverhalten und Erinnerungsvermögen, sowie an der Verhaltensmodulation und Verarbeitung emotionaler Reize involviert. Im Zuge dieser zahlreichen Funktionen können Astrozyten auf externe Stimuli mit der gezielten Freisetzung von Gliotransmittern reagieren.
In kultivierten Astrozyten konnte Keil143 das TGN, bestehend aus Zisternen und Vesikeln, darstellen und mit anti-Rab6 identifizieren. Rab6 mit seinen Subtypen A und B gehört der Superfamilie der monomeren Ras-GTPasen an, die den intrazellulären Membran- und Vesikelverkehr regulieren. Rab6 spielt in HeLa-Zellen beim Transport vesikulärer Organellen vom TGN zur Zellmembran eine wichtige Rolle. Assoziationsanalysen von Rab6A mit vesikulären Glutamattransportern, Serinracemase und Markern der regulierten Exozytose in kultivierten Astrozyten143 deuten darauf hin, dass dieses Rab6A-Organellsystem die ultrastrukturelle Grundlage für die Freisetzung von Gliotransmittern wie D-Serin und Glutamat bildet.
Zur Untersuchung, ob Rab6A tatsächlich ein System der Glia-Neuron-Kommunikation im Gehirn darstellt, war es zunächst unabdingbar das Vorkommen von Rab6A in situ zu untersuchen. Die durchgeführten immunzytochemischen Färbungen an Hirnschnitten der Maus zeigen das gleichmäßige und ubiquitäre Vorkommen von Rab6A in allen untersuchten Hirnregionen. Durch verblindet durchgeführte Kolokalisationsanalysen von Rab6A mit den etablierten astrozytären Markern Glutaminsynthetase (GS), Glial fibrillary acidic protein (GFAP), Aldh1L1 und Sox9 konnte eine Lokalisation von Rab6A in allen Astrozyten gezeigt werden. Weitere Analysen schließen die Lokalisation von Rab6A in Mikroglia (Iba1), NG2-Zellen (NG2) und Oligodendrozyten (CNPase) aus. Die Astrozyten unterscheiden sich in Größe und subzellulärem Verteilungsmuster der Rab6A+ Strukturen, wonach eine Kategorisierung in vier Typen vorgenommen wurde. Anhand der Einteilung kann vermutet werden, dass größere Rab6A+ TGN-Zisternen bis weit in die Zellperipherie transportiert werden und kleine Rab6A+ Vesikel erst dort ausknospen und der Exozytose zugeführt werden. Zur Frage der möglichen astrozytären Subpopulationen konnte gezeigt werden, dass alle untersuchten Astrozyten GS+, Aldh1L1+, Sox9+ und Rab6A+ sind, jedoch nicht GFAP+.
Um die prinzipielle Übertragbarkeit der gewonnenen Befunde auf den Menschen zu überprüfen, wurde reseziertes Cortex-Gewebe von drei Patienten mit unterschiedlicher pathologischer Genese untersucht. Rab6A ist im massiven Ausmaß in humanen Astrozyten lokalisiert, was nahelegt, dass die zuvor an der Maus gewonnenen Ergebnisse auf den Menschen übertragbar sind.
Die mögliche funktionelle Bedeutung von astrozytärem Rab6A im Gehirn wurde an HFS-Schnitten untersucht. Die Untersuchung zeigt einen signifikanten Anstieg der Rab6A+ Intensität in der gesamten Molekularschicht der Fascia dentata der stimulierten im Vergleich zur unstimulierten Seite. Da die HFS ein etabliertes LTP-Modell darstellt, könnte es infolge dieser zu einer strukturellen, intrazellulären Veränderung der Astrozyten mit erhöhter Freisetzung von D-Serin oder Glutamat aus Rab6A+ Vesikeln kommen, was das Lernverhalten beeinflussen könnte. Die dargestellten Ergebnisse legen eine Auswirkung der HFS auf Rab6A nahe.
Zur Bestätigung der immunzytochemischen Untersuchungen wurde die mRNA-Expression von Rab6A in Astrozyten bereits publizierter Transkriptomanalysen untersucht. Die in den Publikationen verwendeten Genom-Chips treffen allenfalls indirekt eine Aussage zu Rab6A, da Rab6 allgemein und nur Rab6B spezifisch untersucht wurde, jedoch keine spezifische Rab6A Sonde erwähnt wird.
Zusammenfassend kann Rab6A als spezifisches und selektiv in Astrozyten vorkommendes Protein dargestellt und als neuer astrozytärer Marker etabliert werden, der auch Astrozyten des humanen Gewebes markiert. Durch die gewonnenen Befunde kann in nachfolgenden Studien die mögliche Bedeutung von Rab6A in neuropathologischen und neurophysiologischen Prozessen untersucht werden.
Complete reosseointegration after treatment of periimplantitis was never published yet. This short scientific communication reports about results of a randomized controlled preclinical study. An electrolytic approach was compared to a classical modality (ablative, cotton pellets soaked with sodium chloride solution and H2O2. For electrolytic cleaning a complete reosseointegration was achieved in several cases serving as a proof of concept.
Cyrtandra argentii Olivar, H.J.Atkins & Muellner sp. nov., endemic to the Philippines and named after George Argent, is herein described and illustrated. Collections associated with this new species are often confused with three other species, namely C. ferruginea Merr., C. villosissima Merr., and C. hirtigera H.J.Atkins & Cronk. Distinguishing characters including keys, updated descriptions, distribution maps, and photos of live specimens are provided to aid identification of the four species. Following the International Union for the Conservation of Nature (IUCN) criteria, C. argentii sp. nov. is considered to be Near Threatened (NT) due to its distribution in a zone susceptible to anthropogenic pressure and the lack of any formal protection.
Die Sicherstellung der hausärztlichen Versorgung ist vor allem im ländlichen Raum mit regional unterschiedlich starker Ausprägung zunehmend gefährdet. Ein wesentlicher Grund liegt in der stetig sinkenden Zahl an hausärztlich tätigen Ärzten/innen. Ursächlich hierfür sind einerseits die hohen „Bruttoabgänge“ von Hausärzten/innen, zumeist aufgrund altersbedingten Ausscheidens, und einem andererseits eklatanten Nachwuchsproblem.
Um dieser problematischen Entwicklung entgegenzuwirken, kommt der Gewinnung hausärztlichen Nachwuchses eine Schlüsselrolle zu. In Flächenländern wie Australien, Kanada oder den USA, die ähnlichen Herausforderungen schon seit längerer Zeit gegenüberstehen, existieren seit den 1970er Jahren universitäre Schwerpunktprogramme, die die Allgemeinmedizin bereits in der medizinischen Ausbildung fördern. Breit angelegte Evaluationsstudien zeigen dabei, dass die Teilnahme an longitudinalen Längsschnittcurricula einen positiven Effekt auf die Wahrscheinlichkeit hat, nach Abschluss des Studiums eine Weiterbildung im Fach Allgemeinmedizin aufzunehmen und sich darüber hinaus hausärztlich (im ländlichen Raum) niederzulassen.
Unter der Annahme, dass eine allgemeinmedizinische Schwerpunktsetzung im Studium das Interesse am selbigen Fach erhöht und darüber hinaus eine hausärztliche Karriereplanung positiv beeinflusst, soll die hier vorliegende Promotionsarbeit folgende Forschungsfrage beantworten: Wie kann ein longitudinales, fachbereichsweites Lehrangebot konzeptionell gestaltet werden, welches es Medizinstudierenden ermöglicht, Allgemeinmedizin im ländlichen Raum kennenzulernen?
Zur Beantwortung der Fragestellung wurde ein triangulierender Forschungsansatz gewählt, der aus mehreren Arbeitsschritten besteht: 1. Erarbeitung einer Übersichtarbeit bestehend aus einer Literaturrecherche und der Kontaktaufnahme zu hiesigen Experten, 2. schriftliche und telefonische Befragung aller medizinischen Fakultäten Deutschlands 3. webbasierte Befragung von Medizinstudierenden der Goethe Universität, Frankfurt, 4. konzeptionelle Entwicklung und Implementierung eines universitären Schwerpunktprogramms zur Förderung der Allgemeinmedizin in ländlichen Regionen auf Basis der in Schritt eins bis drei gewonnen Ergebnisse.
Mittels der verschiedenen methodischen Entwicklungsschritte konnte im Zeitraum von 2015 bis 2016 das longitudinale Schwerpunktprogramm „Landpartie 2.0“ konzeptionell entwickelt und ab dem Wintersemester 2016/2017 in das Medizinstudium der Goethe-Universität, Frankfurt am Main, implementiert werden.
Das entwickelte Lehrangebote richtet sich pro Jahr in der Regel an bis zu 15 Studierende ab dem klinischen Studienabschnitt. Im Kern beinhaltet das mehrsemestrige Angebot wiederkehrende Praxisphasen in ausgewählten und geschulten Hausarztpraxen in ländlichen Regionen. Begleitet werden die Praktika von vor- und nachbereitenden Seminaren an der Universität, dem Kurs Allgemeinmedizin in einer ländlichen Hausarztpraxis und einem jährlichen Tagesausflug zu innovativen Gesundheitsmodellen. Seit Einführung des Programms konnten 62 Studierende in die „Landpartie 2.0“ aufgenommen werden.
Erste Evaluationsergebnisse belegen eine sehr hohe Zufriedenheit mit den einzelnen Programmbestandteilen unter den Teilnehmenden. Langfristig sollen darüber hinaus in einer Verbleibstudie Effekte auf die Motivation für eine hausärztliche Tätigkeit (auf dem Land) sowie Karriereverläufe abgebildet werden.
Insgesamt erwies sich das gewählte methodische Vorgehen als zielführend. Mittels der einzelnen Entwicklungsschritte konnte ein abgestimmtes, umfassendes und den wissenschaftlichen Erkenntnissen berücksichtigendes Längsschnittcurriculum am Fachbereich Medizin der Goethe Universität, Frankfurt am Main, erfolgreich konzeptioniert und implementiert werden.

We use the quantum null energy condition in strongly coupled two-dimensional field theories (QNEC2) as diagnostic tool to study a variety of phase structures, including crossover, second and first order phase transitions. We find a universal QNEC2 constraint for first order phase transitions with kinked entanglement entropy and discuss in general the relation between the QNEC2-inequality and monotonicity of the Casini-Huerta c-function. We then focus on a specific example, the holographic dual of which is modelled by three-dimensional Einstein gravity plus a massive scalar field with one free parameter in the self-interaction potential. We study translation invariant stationary states dual to domain walls and black branes. Depending on the value of the free parameter we find crossover, second and first order phase transitions between such states, and the c-function either flows to zero or to a finite value in the infrared. Strikingly, evaluating QNEC2 for ground state solutions allows to predict the existence of phase transitions at finite temperature.
Risk stratification for bipolar disorder using polygenic risk scores among young high-risk adults

Objective: Identifying high-risk groups with an increased genetic liability for bipolar disorder (BD) will provide insights into the etiology of BD and contribute to early detection of BD. We used the BD polygenic risk score (PRS) derived from BD genome-wide association studies (GWAS) to explore how such genetic risk manifests in young, high-risk adults. We postulated that BD-PRS would be associated with risk factors for BD.
Methods: A final sample of 185 young, high-risk German adults (aged 18–35 years) were grouped into three risk groups and compared to a healthy control group (n = 1,100). The risk groups comprised 117 cases with attention deficit hyperactivity disorder (ADHD), 45 with major depressive disorder (MDD), and 23 help-seeking adults with early recognition symptoms [ER: positive family history for BD, (sub)threshold affective symptomatology and/or mood swings, sleeping disorder]. BD-PRS was computed for each participant. Logistic regression models (controlling for sex, age, and the first five ancestry principal components) were used to assess associations of BD-PRS and the high-risk phenotypes.
Results: We observed an association between BD-PRS and combined risk group status (OR = 1.48, p < 0.001), ADHD diagnosis (OR = 1.32, p = 0.009), MDD diagnosis (OR = 1.96, p < 0.001), and ER group status (OR = 1.7, p = 0.025; not significant after correction for multiple testing) compared to healthy controls.
Conclusions: In the present study, increased genetic risk for BD was a significant predictor for MDD and ADHD status, but not for ER. These findings support an underlying shared risk for both MDD and BD as well as ADHD and BD. Improving our understanding of the underlying genetic architecture of these phenotypes may aid in early identification and risk stratification.
The current pandemic situation caused by the Betacoronavirus SARS-CoV-2 (SCoV2) highlights the need for coordinated research to combat COVID-19. A particularly important aspect is the development of medication. In addition to viral proteins, structured RNA elements represent a potent alternative as drug targets. The search for drugs that target RNA requires their high-resolution structural characterization. Using nuclear magnetic resonance (NMR) spectroscopy, a worldwide consortium of NMR researchers aims to characterize potential RNA drug targets of SCoV2. Here, we report the characterization of 15 conserved RNA elements located at the 5′ end, the ribosomal frameshift segment and the 3′-untranslated region (3′-UTR) of the SCoV2 genome, their large-scale production and NMR-based secondary structure determination. The NMR data are corroborated with secondary structure probing by DMS footprinting experiments. The close agreement of NMR secondary structure determination of isolated RNA elements with DMS footprinting and NMR performed on larger RNA regions shows that the secondary structure elements fold independently. The NMR data reported here provide the basis for NMR investigations of RNA function, RNA interactions with viral and host proteins and screening campaigns to identify potential RNA binders for pharmaceutical intervention.
1H, 13C, and 15N backbone chemical shift assignments of coronavirus-2 non-structural protein Nsp10

The international Covid19-NMR consortium aims at the comprehensive spectroscopic characterization of SARS-CoV-2 RNA elements and proteins and will provide NMR chemical shift assignments of the molecular components of this virus. The SARS-CoV-2 genome encodes approximately 30 different proteins. Four of these proteins are involved in forming the viral envelope or in the packaging of the RNA genome and are therefore called structural proteins. The other proteins fulfill a variety of functions during the viral life cycle and comprise the so-called non-structural proteins (nsps). Here, we report the near-complete NMR resonance assignment for the backbone chemical shifts of the non-structural protein 10 (nsp10). Nsp10 is part of the viral replication-transcription complex (RTC). It aids in synthesizing and modifying the genomic and subgenomic RNAs. Via its interaction with nsp14, it ensures transcriptional fidelity of the RNA-dependent RNA polymerase, and through its stimulation of the methyltransferase activity of nsp16, it aids in synthesizing the RNA cap structures which protect the viral RNAs from being recognized by the innate immune system. Both of these functions can be potentially targeted by drugs. Our data will aid in performing additional NMR-based characterizations, and provide a basis for the identification of possible small molecule ligands interfering with nsp10 exerting its essential role in viral replication.
We use holography to study the dynamics of a strongly-coupled gauge theory in four-dimensional de Sitter space with Hubble rate H. The gauge theory is non-conformal with a characteristic mass scale M. We solve Einstein’s equations numerically and determine the time evolution of homogeneous gauge theory states. If their initial energy density is high compared with H4 then the early-time evolution is well described by viscous hydrodynamics with a non-zero bulk viscosity. At late times the dynamics is always far from equilibrium. The asymptotic late-time state preserves the full de Sitter symmetry group and its dual geometry is a domain-wall in AdS5. The approach to this state is characterised by an emergent relation of the form P = w E that is different from the equilibrium equation of state in flat space. The constant w does not depend on the initial conditions but only on H/M and is negative if the ratio H/M is close to unity. The event and the apparent horizons of the late-time solution do not coincide with one another, reflecting its non-equilibrium nature. In between them lies an “entanglement horizon” that cannot be penetrated by extremal surfaces anchored at the boundary, which we use to compute the entanglement entropy of boundary regions. If the entangling region equals the observable universe then the extremal surface coincides with a bulk cosmological horizon that just touches the event horizon, while for larger regions the extremal surface probes behind the event horizon.
We use the quantum null energy condition in strongly coupled two-dimensional field theories (QNEC2) as diagnostic tool to study a variety of phase structures, including crossover, second and first order phase transitions. We find a universal QNEC2 constraint for first order phase transitions with kinked entanglement entropy and discuss in general the relation between the QNEC2-inequality and monotonicity of the Casini-Huerta c-function. We then focus on a specific example, the holographic dual of which is modelled by three-dimensional Einstein gravity plus a massive scalar field with one free parameter in the self-interaction potential. We study translation invariant stationary states dual to domain walls and black branes. Depending on the value of the free parameter we find crossover, second and first order phase transitions between such states, and the c-function either flows to zero or to a finite value in the infrared. Strikingly, evaluating QNEC2 for ground state solutions allows to predict the existence of phase transitions at finite temperature.
Netzwerkmodelle spielen in verschiedenen Wissenschaftsdisziplinen eine wichtige Rolle und dienen unter anderem der Beschreibung realistischer Graphen.
Sie werden häufig als Zufallsgraphen formuliert und stellen somit Wahrscheinlichkeitsverteilungen über Graphen dar.
Meist ist die Verteilung dabei parametrisiert und ergibt sich implizit, etwa über eine randomisierten Konstruktionsvorschrift.
Ein früher Vertreter ist das G(n,p) Modell, welches über allen ungerichteten Graphen mit n Knoten definiert ist und jede Kante unabhängig mit Wahrscheinlichkeit p erzeugt.
Ein aus G(n,p) gezogener Graph hat jedoch kaum strukturelle Ähnlichkeiten zu Graphen, die zumeist in Anwendungen beobachtet werden.
Daher sind populäre Modelle so gestaltet, dass sie mit hinreichend hoher Wahrscheinlichkeit gewünschte topologische Eigenschaften erzeugen.
Beispielsweise ist es ein gängiges Ziel die nur unscharf definierte Klasse der sogenannten komplexen Netzwerke nachzubilden, der etwa viele soziale Netze zugeordnet werden.
Unter anderem verfügen diese Graphen in der Regel über eine Gradverteilung mit schweren Rändern (heavy-tailed), einen kleinen Durchmesser, eine dominierende Zusammenhangskomponente, sowie über überdurchschnittlich dichte Teilbereiche, sogenannte Communities.
Die Einsatzmöglichkeiten von Netzwerkmodellen gehen dabei weit über das ursprüngliche Ziel, beobachtete Effekte zu erklären, hinaus.
Ein gängiger Anwendungsfall besteht darin, Daten systematisch zu produzieren.
Solche Daten ermöglichen oder unterstützen experimentelle Untersuchungen, etwa zur empirischen Verifikation theoretischer Vorhersagen oder zur allgemeinen Bewertung von Algorithmen und Datenstrukturen.
Hierbei ergeben sich insbesondere für große Probleminstanzen Vorteile gegenüber beobachteten Netzen.
So sind massive Eingaben, die auf echten Daten beruhen, oft nicht in ausreichender Menge verfügbar, nur aufwendig zu beschaffen und zu verwalten, unterliegen rechtlichen Beschränkungen, oder sind von unklarer Qualität.
In der vorliegenden Arbeit betrachten wir daher algorithmische Aspekte der Generierung massiver Zufallsgraphen.
Um Anwendern Reproduzierbarkeit mit vorhandenen Studien zu ermöglichen, fokussieren wir uns hierbei zumeist auf getreue Implementierungen etablierter Netzwerkmodelle,
etwa Preferential Attachment-Prozesse, LFR, simple Graphen mit vorgeschriebenen Gradsequenzen, oder Graphen mit hyperbolischer (o.Ä.) Einbettung.
Zu diesem Zweck entwickeln wir praktisch sowie analytisch effiziente Generatoren.
Unsere Algorithmen sind dabei jeweils auf ein geeignetes Maschinenmodell hin optimiert.
Hierzu entwerfen wir etwa klassische sequentielle Generatoren für Registermaschinen, Algorithmen für das External Memory Model, und parallele Ansätze für verteilte oder Shared Memory-Maschinen auf CPUs, GPUs, und anderen Rechenbeschleunigern.
Introduction: From the beginning of the corona pandemic until August 19, 2020, more than 21,989,366 cases have been reported worldwide – 228,495 in Germany alone, including 12,648 children aged 0–14. In many countries, the proportion of infected children in the total population is comparatively low; in addition, children often have no or milder symptoms and are less likely to transmit the pathogen to adults than the other way round. Based on the registration data in Frankfurt am Main, Germany, the symptoms of children in comparison with adults and the likely routes of transmission are presented below.
Materials and methods: The documentation of the mandatory reports includes personal data (name, date of birth, gender, place of residence), disease characteristics (date of report, date of onset of the disease, symptoms), possible contact persons (family, others) and i.a. possible activity or care in children’s community facilities. All reports were viewed, especially with regard to likely transmission routes.
Results: From March 1 to July 31, 2020, 1,977 infected people were reported, including 138 children between the ages of 0 and 14 years. Children had fewer and milder symptoms than adults. None of the children experienced severe respiratory symptoms or the need for ventilation. 62% of the children had no symptoms at all (19% adults), 5% of the children were hospitalized (24% adults), and none of the children died (3.8% adults).
After excluding a cluster of 34 children from refugee accommodations and 14 children from a parish, 78% of the remaining 90 children had been infected by an adult within the family, and only 4% were likely to have a reverse transmission route. In 5.5% of cases, transmission in a community facility was likely.
Discussion: The results of the registration data from Frankfurt am Main, Germany confirm the results published in other countries: Children are less likely to become infected, and if infected, their symptoms are less severe than in adults, and they are apparently not the main drivers of virus transmission. Therefore, scientific medical associations strongly recommend reopening schools.
Das Internet findet auf unterschiedlichste Weise Eingang in den Film: Digitale Formate wie Webserien, Podcasts oder sogar Tweets werden im Medienwechsel Grundlage filmischer Adaptionen, filmische Experimente mit interaktiven und virtuellen Technologien generieren neue, zwischen Film und Computerspiel angesiedelte Medienkombinationen, transmediale Erweiterungen führen auf verschiedene Arten Film- und Serienuniversen im digitalen Raum fort und intermediale Bezüge erzählen durch die Imitation einer digitalen Ästhetik nicht (nur) über das Altermedium, sondern oft auch durch das andere Medium. Zu letzterer intermedialer Kategorie gehörende Phänomene der Thematisierung, Evozierung oder Simulierung sollen hier im Kontext der Darstellung des Internets analysiert werden. Aufgrund der Ubiquität digitaler Medien im Alltag spielen seit einigen Jahren neuere Technologien als Bezugsmedien eine zentrale Rolle in vielen Filmen und Serien. Filmische Internetanwendungen werden dabei vor allem als grafische Benutzeroberfläche, als Nutzungsschnittstelle zwischen Anwender und technischem Gerät visualisiert, die Repräsentation der Hardware erscheint meist nachrangig. Nicht die Darstellung von Computern und Smartphones, sondern die Inszenierung von vernetzten Systemen, Räumen und Kommunikationsstrukturen steht daher im Fokus dieses Artikels. Eingegangen werden soll in diesem Zusammenhang insbesondere auf intermediale Evozierungen des Altermediums durch die Nachahmung digitaler Ästhetiken vermittels des Formenrepertoires des Films, simulierte Screen- und Desktopfilme und auf die Darstellung der dominant schrift- und zeichenbasierten digitalen Kultur durch die Integration von Schrift im Filmbild. Begonnen wird die Untersuchung mit einer Betrachtung von visuellen Metaphern und Strategien der Sichtbarmachung virtueller Räume.
Inschriften sind Formen, die durch eine besondere mediale Disposition charakterisiert sind. Was Inschriften auszeichnet, ist, neben ihrem engen Bezug zu einem materiellen Träger, ihre eigentümliche Position auf der Schwelle von Schrift und Bild. [...] Die Eigenart der Inschrift, ein Wort oder einen Text als sichtbare Zeichenfolge auszustellen, hat der italienische Epigraphieforscher Armando Petrucci im Begriff der 'scrittura esposta' zum Ausdruck gebracht. [...] Versucht man, die damit berührte spezifische Potenz der Inschrift genauer zu erfassen, liegt es nahe, zunächst auf die visuelle Dimension zurückzukommen. Es ist, so darf man annehmen, die Fähigkeit der Inschrift, als Bild zu erscheinen, die es ihr erlaubt, in den Blick des Betrachters zu treten und sich jenem als exponierte Figur vor Augen zu stellen. Mit dieser bildhaften Erscheinungsform, so ließe sich das Argument weiterführen, verbinden sich ästhetische Qualitäten der sinnlichen Eindrücklichkeit und Präsenz, die der Inschrift die ihr eigentümliche Ausdrucks- und Aussagekraft verleihen. [...] Mit dieser Erklärung ist unterdessen nur die eine Seite der Inschrift und ihrer medien- und wirkungsästhetischen Beschaffenheit erfasst. Das Besondere der Inschrift erschöpft sich nicht in deren Eigenart als ausgestellter, exponierter Zeichenformation. Die Inschrift ist nicht nur 'esposta', sondern ebenso 'scrittura'. Die besondere Gestaltungs- und Wirkungsweise der Inschrift beruht mithin nicht allein auf deren bildhafter Disposition. Die Wirkkraft der Inschrift verdankt sich, so die hier vorgeschlagene These, dem Umstand, dass diese, auch wenn sie sich als exponierte, eingängig und weithin sichtbare Gestalt zur Geltung bringt, zugleich ihren Charakter als Schrift bewahrt und diesen nicht weniger deutlich hervorkehrt. Wer eine Inschrift betrachtet, der erblickt in ihrer bildhaften Gestaltung zugleich die visuelle Form eines Textes, einer sprachlichen Äußerung. Durch ihre Gestaltung als 'scrittura' erscheint die Inschrift somit in einer Form, die in spezifischer Weise mit Momenten der Macht und Autorität versehen ist. Ist doch die Schrift dasjenige Medium, in dem uns, in einer von der Antike bis in die Neuzeit und Moderne reichenden Tradition, das Gesetz, die aufgezeichnete und materialisierte 'Stimme des Souveräns' entgegentritt. Das Besondere der Inschrift scheint also, so lässt sich vorläufig festhalten, darin zu bestehen, dass sie die Medien von Bild und Schrift in einer spezifischen Weise miteinander verknüpft. In ihr sind mediale und ästhetische Qualitäten wirksam, die teils dem Bild, teils der Schrift angehören. Auf diesem Zusammenspiel beruht auch das eigentümliche Wirkungspotential, das sich mit dieser Äußerungsform verbindet. In der Folge wird es darum gehen, dieses Zusammenwirken bildlicher und skripturaler Aspekte genauer zu erkunden und vor diesem Hintergrund die Bedeutung und Wirkkraft inschriftlicher Zeichen insbesondere in politischen Kontexten zu untersuchen.
Die Verunsicherung auf dem Feld zeitgenössischer Kunst berührt nicht nur die Frage nach der Qualität von Kunst, sondern auch jene der Grenze zwischen Kunst(werk) und ihrem (bzw. seinem) jeweiligen Außen. [...] Kunst, die einen herkömmlichen Werkbegriff in Frage stellt (und vom breiten Publikum oft abgelehnt wird), aber doch verortet und verortbar und daher, zumindest weitestgehend, als Kunst erkennbar ist, soll im folgenden Gegenwartskunst genannt werden, die in den Alltag integrierte und intervenierende und manchmal nicht als Kunst wahrgenommene Kunst als Situationskunst. Gegenwartskunst setzt ihre Autonomie und eine klare Grenze zwischen Kunst und Nicht-Kunst voraus, Situationskunst (die man als eine radikale Ausformung und somit als Teil der Gegenwartskunst ansehen könnte) sät Zweifel an der Kunstautonomie, auch wenn sie diese häufig als Argument gegen Anrufungen oder Übergriffe von Politik, Religion oder Alltagswirklichkeit verwendet bzw. verwenden 'muss'. Bei beiden Formen, die sich in vielen Fällen überschneiden, wird im herkömmlichen Sinne nichts mehr erschaffen ('poesis'), sondern etwas gefunden bzw. letztlich 'einfach' etwas getan ('praxis'). In beiden Fällen versteht sich nichts mehr von selbst: Es ist in der Rezeption - zumindest im ersten Moment - unklar, ob wir es überhaupt mit Kunst zu tun haben. In anderen Worten: Wir können uns im Moment des Ausstellungsbesuches also nicht auf unsere Sinneswahrnehmungen, auf unsere Erfahrung und auf unser implizites (Vor-)Wissen verlassen, wenn wir wissen wollen, womit wir es zu tun haben und was das alles soll. Wir benötigen also nicht zuletzt Erklärungen und Erläuterungen (die wieder zu implizitem Wissen gerinnen können) - und das ist ein Grund, warum zeitgenössische Kunst für die Komparatistik interessant sein könnte. Davon wird noch zu sprechen sein. Die Begriffe Gegenwarts- und Situationskunst decken einen sehr weiten Bereich von Phänomenen ab. Daher wird das Folgende eine kursorische Skizze werden, bei der in erster Linie auf solche Phänomene und ihre Gemeinsamkeiten abgezielt werden soll, die für die Komparatistik von Interesse sind. Im Zentrum steht nicht eine genaue Analyse und Interpretation von Phänomenen, sondern die Frage, was im Hinblick auf die Disziplin der Komparatistik spannend für Analyse und Interpretation wäre. Die im Folgenden diskutierten Phänomene und Beispiele befinden sich auf jeden Fall in der Peripherie der Komparatistik mit allen Nachteilen, welche die Arbeit in Peripherien mit sich bringt.
Briefe, das Gespräch zweier Abwesender miteinander, spielen in vielen Filmen eine große Rolle. Sie werden eingeblendet oder per 'voice over' vorgelesen, man sieht Lese- und Schreibszenen, die mit der Vieldeutigkeit des Geschriebenen spielen. Der Brief sei, so Christina Bartz, "wegen der kommunikativen Verbindung über zeitliche und räumliche Distanzen hinweg" "besonders anschlussfähig für den Film", der ebenfalls durch die Montage räumlich und zeitlich Getrenntes zusammenbringt. Im Gegensatz zum Film ist der Brief jedoch kein Massenmedium sondern Individualkommunikation. Das Zeigen des Mediums Brief oder das Ersetzen dieses historischen Mediums durch ein aktuelleres im Film bietet immer auch die Möglichkeit der Medienreflexion. In meinem Beitrag möchte ich anhand zweier prominenter Beispiele zum einen beobachten, wie in filmischen Adaptionen briefgeprägter literarischer Texte mit Briefen umgegangen wird, und zum anderen, wie anhand der Briefthematik eine Medienreflexion stattfindet. Ich stelle dazu zwei Melodramen vor, in denen Briefe und das damit einhergehende Erkennen und Verkennen eine zentrale Rolle spielen: Max Ophüls' "Letter from an unknown woman" (USA 1948), der Verfilmung von Stefan Zweigs Novelle "Brief einer Unbekannten" (1922), und "Atonement" (2007), die Adaption von Ian McEwans gleichnamigen Roman von 2001.
Wie kaum ein anderes Bildmotiv machen schmelzende Gletscher den Klimawandel sichtbar. Sie spielen deshalb eine zentrale Rolle für die Klimaforschung selbst, für die Popularisierung ihrer alarmierenden Erkenntnisse sowie für die zeitgenössische Kunst, die im Lichte dieser Einsichten nach einer adäquaten neuen Ästhetik sucht. Entsprechend umfangreich fällt inzwischen auch die kulturwissenschaftliche Auseinandersetzung mit Gletscherbildern aus. Zahlreiche Ausstellungskataloge und umfangreiche Studien verfolgen deren Entwicklung vom frühen 17. Jahrhundert, auf das die ersten bildlichen Darstellungen datiert sind, bis in die Gegenwart, in der Gletscher und ihr Verschwinden zum Emblem der globalen Erwärmung geworden sind. Der Heuristik des Vergleichs kommt dabei eine wichtige Funktion zu: Nicht nur bildet sie die Basis etwa für klassisch kunsthistorische Untersuchungen, deren Augenmerk dem Wandel der Ausdrucksformen und Abbildungskonventionen von Gletscherbildern (etwa auf einer Skala zwischen Idealisierung und Realismus) gilt. Überdies und insbesondere ist auch der Prozess des Verschwindens auf den vergleichenden Blick angewiesen, denn dieser offenbart sich ja erst auf diese Weise in seiner ganzen Dramatik. Dieser Aufsatz jedoch wählt eine andere Perspektive: In begrifflicher Anlehnung an Jussi Parikkas 'Mediengeologie' und vor dem Hintergrund des umfassenden Felds der Medienökologie wird im Folgenden eine "Medienglaziologie" umrissen, die Gletscher selbst als Medien versteht. Ganz im Sinne des medienkomparatistischen Forschungsparadigmas, dass sich spezifische Medialitäten erst aus einer medienvergleichenden Perspektive erschließen, wird der Frage nachgegangen, wie sich dieses "Medien-Werden" der Gletscher im und durch den Vergleich mit anderen (technischen) Medien vollzieht. Dabei konzentriere ich mich zeitlich auf das 19. und frühe 20. Jahrhundert und regional auf die Alpengletscher, deren wissenschaftliche Erforschung die Disziplin der Glaziologie begründete.
The elliptic flow (v2) of (anti-)3He is measured in Pb–Pb collisions at √sNN=5.02TeV in the transverse-momentum (pT) range of 2–6 GeV/c for the centrality classes 0–20%, 20–40%, and 40–60% using the event-plane method. This measurement is compared to that of pions, kaons, and protons at the same center-of-mass energy. A clear mass ordering is observed at low pT, as expected from relativistic hydrodynamics. The violation of the scaling of v2 with the number of constituent quarks at low pT, already observed for identified hadrons and deuterons at LHC energies, is confirmed also for (anti-)3He. The elliptic flow of (anti-)3He is underestimated by the Blast-Wave model and overestimated by a simple coalescence approach based on nucleon scaling. The elliptic flow of (anti-)3He measured in the centrality classes 0–20% and 20–40% is well described by a more sophisticated coalescence model where the phase-space distributions of protons and neutrons are generated using the iEBE-VISHNU hybrid model with AMPT initial conditions.
Macro-finance theory predicts that financial fragility builds up when volatility is low. This “volatility paradox’” challenges traditional systemic risk measures. I explore a new dimension of systemic risk, spillover persistence, which is the average time horizon at which a firm’s losses increase future risk in the financial system. Using firm-level data covering more than 30 years and 50 countries, I document that persistence declines when fragility builds up: before crises, during stock market booms, and when banks take more risks. In contrast, persistence increases with loss amplification: during crises and fire sales. These findings support key predictions of recent macrofinance models.
The impact of the appropriate and inappropriately applied statistical metrics to verify the State of Control of pharmaceutical manufacturing has been reviewed from an auditor’s perspective. Good and bad statistical practices have been presented in an attempt for manufactures to appreciate the risks of using these metrics. Conclusions concerning (1) control charts to be used instead of line/run charts for trend analysis, (2) Ppk as the preferred capability index (but still with an ambition to get processes into statistical control), (3) show process capability indices along with their respective control charts (4) determine which Manufacturing State of Control the product/process lies in (5) an effective Control Strategy can only be implemented if the Manufacturing State of Control is understood, (6) when presenting data consider what is truly representative of the product/process and not the average (7) Management should align with ICH Q10 more effectively to provide statistical resources for their personnel.
Linking epigenetic signature and metabolic phenotype in IDH mutant and IDH wildtype diffuse glioma

Aims: Changes in metabolism are known to contribute to tumour phenotypes. If and how metabolic alterations in brain tumours contribute to patient outcome is still poorly understood. Epigenetics impact metabolism and mitochondrial function. The aim of this study is a characterisation of metabolic features in molecular subgroups of isocitrate dehydrogenase mutant (IDHmut) and isocitrate dehydrogenase wildtype (IDHwt) gliomas. Methods: We employed DNA methylation pattern analyses with a special focus on metabolic genes, large-scale metabolism panel immunohistochemistry (IHC), qPCR-based determination of mitochondrial DNA copy number and immune cell content using IHC and deconvolution of DNA methylation data. We analysed molecularly characterised gliomas (n = 57) for in depth DNA methylation, a cohort of primary and recurrent gliomas (n = 22) for mitochondrial copy number and validated these results in a large glioma cohort (n = 293). Finally, we investigated the potential of metabolic markers in Bevacizumab (Bev)-treated gliomas (n = 29). Results: DNA methylation patterns of metabolic genes successfully distinguished the molecular subtypes of IDHmut and IDHwt gliomas. Promoter methylation of lactate dehydrogenase A negatively correlated with protein expression and was associated with IDHmut gliomas. Mitochondrial DNA copy number was increased in IDHmut tumours and did not change in recurrent tumours. Hierarchical clustering based on metabolism panel IHC revealed distinct subclasses of IDHmut and IDHwt gliomas with an impact on patient outcome. Further quantification of these markers allowed for the prediction of survival under anti-angiogenic therapy. Conclusion: A mitochondrial signature was associated with increased survival in all analyses, which could indicate tumour subgroups with specific metabolic vulnerabilities.
Simple Summary: Targeted therapies are of growing interest to physicians in cancer treatment. These drugs target specific genes and proteins involved in the growth and survival of cancer cells. Brain tumor therapy is complicated by the fact that not all drugs can penetrate the blood brain barrier and reach their target. We explored the non-invasive method, Magnetic Resonance Spectroscopy, for monitoring drug penetration and its effects in live animals bearing brain tumors. We were able to show the presence of the investigated drug in mouse brains and its on-target activity.
Abstract: Background: BAY1436032 is a fluorine-containing inhibitor of the R132X-mutant isocitrate dehydrogenase (mIDH1). It inhibits the mIDH1-mediated production of 2-hydroxyglutarate (2-HG) in glioma cells. We investigated brain penetration of BAY1436032 and its effects using 1H/19F-Magnetic Resonance Spectroscopy (MRS). Methods: 19F-Nuclear Magnetic Resonance (NMR) Spectroscopy was conducted on serum samples from patients treated with BAY1436032 (NCT02746081 trial) in order to analyze 19F spectroscopic signal patterns and concentration-time dynamics of protein-bound inhibitor to facilitate their identification in vivo MRS experiments. Hereafter, 30 mice were implanted with three glioma cell lines (LNT-229, LNT-229 IDH1-R132H, GL261). Mice bearing the IDH-mutated glioma cells received 5 days of treatment with BAY1436032 between baseline and follow-up 1H/19F-MRS scan. All other animals underwent a single scan after BAY1436032 administration. Mouse brains were analyzed by liquid chromatography-mass spectrometry (LC-MS/MS). Results: Evaluation of 1H-MRS data showed a decrease in 2-HG/total creatinine (tCr) ratios from the baseline to post-treatment scans in the mIDH1 murine model. Whole brain concentration of BAY1436032, as determined by 19F-MRS, was similar to total brain tissue concentration determined by Liquid Chromatography with tandem mass spectrometry (LC-MS/MS), with a signal loss due to protein binding. Intratumoral drug concentration, as determined by LC-MS/MS, was not statistically different in models with or without R132X-mutant IDH1 expression. Conclusions: Non-invasive monitoring of mIDH1 inhibition by BAY1436032 in mIDH1 gliomas is feasible.
The production of light neutral mesons in AA collisions probes the physics of the Quark-Gluon Plasma (QGP), which is formed in heavy-ion collisions at the LHC. More specifically, the centrality dependent neutral meson spectra in AA collisions compared to its spectra in minimum-bias pp collisions, scaled with the number of hard collisions, provides information on the energy loss of partons traversing the QGP. The measurement allows to test with high precision the predictions of theoretical model calculations. In addition, the decay of the π0 and η mesons are the dominant back- grounds for all direct photon measurements. Therefore, pushing the limits of the precision of neutral meson production is key to learning about the temperature and space-time evolution of the QGP.
In the ALICE experiment neutral mesons can be detected via their decay into two photons. The latter can be reconstructed using the two calorimeters EMCal and PHOS or via conversions in the detector material. The excellent momentum resolution of the conversion photons down to very low pT and the high reconstruction efficiency and triggering capability of calorimeters at high pT, allow us to measure the pT dependent invariant yield of light neutral mesons over a wide kinematic range.
Combining state-of-the-art reconstruction techniques with the high statistics delivered by the LHC in Run 2 gives us the opportunity to enhance the precision of our measurements. In these proceedings, new ALICE run 2 preliminary results for neutral meson production in pp and Pb–Pb collisions at LHC energies are presented.
Nature affects human well-being in multiple ways. However, the association between species diversity and human well-being at larger spatial scales remains largely unexplored. Here, we examine the relationship between species diversity and human well-being at the continental scale, while controlling for other known drivers of well-being. We related socio-economic data from more than 26,000 European citizens across 26 countries with macroecological data on species diversity and nature characteristics for Europe. Human well-being was measured as self-reported life-satisfaction and species diversity as the species richness of several taxonomic groups (e.g. birds, mammals and trees). Our results show that bird species richness is positively associated with life-satisfaction across Europe. We found a relatively strong relationship, indicating that the effect of bird species richness on life-satisfaction may be of similar magnitude to that of income. We discuss two, non-exclusive pathways for this relationship: the direct multisensory experience of birds, and beneficial landscape properties which promote both bird diversity and people's well-being. Based on these results, this study argues that management actions for the protection of birds and the landscapes that support them would benefit humans. We suggest that political and societal decision-making should consider the critical role of species diversity for human well-being.
Introduction: Recommendations for venous thromboembolism and deep venous thrombosis (DVT) prophylaxis using graduated compression stockings (GCS) is historically based and has been critically examined in current publications. Existing guidelines are inconclusive as to recommend the general use of GCS.
Patients/Methods: 24 273 in-patients (general surgery and orthopedic patients) undergoing surgery between 2006 and 2016 were included in a retrospectively analysis from a single center. From January 2006 to January 2011 perioperative GCS was employed additionally to drug prophylaxis and from February 2011 to March 2016 patients received drug prophylaxis alone. According to german guidelines all patients received venous thromboembolism prophylaxis with weight-adapted LMWH. Risk stratification (low risk, moderate risk, high risk) was based on the guideline of the American College of Chest Physicians. Data analysis was performed before and after propensity matching (PM). The defined primary endpoint was the incidence of symptomatic or fatal pulmonary embolism (PE). A secondary endpoint was the incidence of deep venous thromboembolism (DVT).
Results: After risk stratification (low risk n = 16 483; moderate risk n = 4464; high risk n = 3326) a total of 24 273 patient were analyzed. Before to PM the relative risk for the occurrence of a PE or DVT was not increased by abstaining from GCS. After PM two groups of 11 312 patients each, one with and one without GCS application, were formed. When comparing the two groups, the relative risk (RR) for the occurrence of a pulmonary embolism was: Low Risk 0.99 [CI95% 0.998–1.000]; Moderate Risk 0.999 [CI95% 0.95–1.003]; High Risk 0.996 [CI95% 0.992–1.000] (p > 0.05). The incidence of PE in the total group LMWH alone was 0.1% (n = 16). In the total group using LMWH + GCS, the incidence was 0.3% (n = 29). RR after PM was 0.999 [CI95% 0.998–1.00].
Conclusion: In comparison to prior studies with only small numbers of patients our trial shows in a large group of patients with moderate and high risk developing VTE we can support the view that abstaining from GCS-use does not increase the incidence of symptomatic or fatal PE and symptomatic DVT.
Over the last 15 years the Diagnostic Center of Acute Leukemia (DCAL) at the Frankfurt University has diagnosed and elucidated the Mixed Lineage Leukemia (MLL) recombinome with >100 MLL fusion partners. When analyzing all these different events, balanced chromosomal translocations were found to comprise the majority of these cases (~70%), while other types of genetic rearrangements (3-way-translocations, spliced fusions, 11q inversions, interstitial deletions or insertion of chromosomal fragments into other chromosomes) account for about 30%. In nearly all those complex cases, functional fusion proteins can be produced by transcription, splicing and translation. With a few exceptions (10 out of 102 fusion genes which were per se out-of-frame), all these genetic rearrangements produced a direct MLL fusion gene, and in 94% of cases an additional reciprocal fusion gene. So far, 114 patients (out of 2454 = ~5%) have been diagnosed only with the reciprocal fusion allele, displaying no MLL-X allele. The fact that so many MLL rearrangements bear at least two fusion alleles, but also our findings that several direct MLL fusions were either out-of-frame fusions or missing, raises the question about the function and importance of reciprocal MLL fusions. Recent findings also demonstrate the presence of reciprocal MLL fusions in sarcoma patients. Here, we want to discuss the role of reciprocal MLL fusion proteins for leukemogenesis and beyond.
Inhibitors against the NS3-4A protease of hepatitis C virus (HCV) have proven to be useful drugs in the treatment of HCV infection. Although variants have been identified with mutations that confer resistance to these inhibitors, the mutations do not restore replicative fitness and no secondary mutations that rescue fitness have been found. To gain insight into the molecular mechanisms underlying the lack of fitness compensation, we screened known resistance mutations in infectious HCV cell culture with different genomic backgrounds. We observed that the Q41R mutation of NS3-4A efficiently rescues the replicative fitness in cell culture for virus variants containing mutations at NS3-Asp168. To understand how the Q41R mutation rescues activity, we performed protease activity assays complemented by molecular dynamics simulations, which showed that protease-peptide interactions far outside the targeted peptide cleavage sites mediate substrate recognition by NS3-4A and support protease cleavage kinetics. These interactions shed new light on the mechanisms by which NS3-4A cleaves its substrates, viral polyproteins and a prime cellular antiviral adaptor protein, the mitochondrial antiviral signaling protein MAVS. Peptide binding is mediated by an extended hydrogen-bond network in NS3-4A that was effectively optimized for protease-MAVS binding in Asp168 variants with rescued replicative fitness from NS3-Q41R. In the protease harboring NS3-Q41R, the N-terminal cleavage products of MAVS retained high affinity to the active site, rendering the protease susceptible for potential product inhibition. Our findings reveal delicately balanced protease-peptide interactions in viral replication and immune escape that likely restrict the protease adaptive capability and narrow the virus evolutionary space.

Abstract
To promote non-motorised travel, many travel behaviour studies acknowledge the importance of the built environment to modal choice, for example with its density or mix of uses. From a mobility design theory perspective, however, objects and environments affect human perceptions, assessments and behaviour in at least three different ways: by their practical, aesthetic and emblematic functions. This review of existing evidence will argue that travel behaviour research has so far mainly focused on the practical function of the built environment. For that purpose, we systematically identified 56 relevant studies on the impacts of the built environment on non-motorised travel behaviour in the Web of Science database. The focus of research on the practical design function primary involves land use distribution, street network connectivity and the presence of walking and cycling facilities. Only a small number of papers address the aesthetic and emblematic functions. These show that the perceived attractiveness of an environment and evoked feelings of traffic safety increase the likelihood of walking and cycling. However, from a mobility design perspective, the results of the review indicate a gap regarding comprehensive research on the effects of the aesthetic and emblematic functions of the built environment. Further research involving these functions might contribute to a better understanding of how to promote non-motorised travel more effectively. Moreover, limitations related to survey techniques, regional distribution and the comparability of results were identified.
This research examines the impact of online display advertising and paid search advertising relative to offline advertising on firm performance and firm value. Using proprietary data on annualized advertising expenditures for 1651 firms spanning seven years, we document that both display advertising and paid search advertising exhibit positive effects on firm performance (measured by sales) and firm value (measured by Tobin's q). Paid search advertising has a more positive effect on sales than offline advertising, consistent with paid search being closest to the actual purchase decision and having enhanced targeting abilities. Display advertising exhibits a relatively more positive effect on Tobin's q than offline advertising, consistent with its long-term effects. The findings suggest heterogeneous economic benefits across different types of advertising, with direct implications for managers in analyzing advertising effectiveness and external stakeholders in assessing firm performance.
The US Treasury recently permitted deferred longevity income annuities to be included in pension plan menus as a default payout solution, yet little research has investigated whether more people should convert some of the $18 trillion they hold in employer-based defined contribution plans into lifelong income streams. We investigate this innovation using a calibrated lifecycle consumption and portfolio choice model embodying realistic institutional considerations. Our welfare analysis shows that defaulting a modest portion of retirees’ 401(k) assets (over a threshold) is an attractive way to enhance retirement security, enhancing welfare by up to 20% of retiree plan accruals.
In this paper a new method of experimental data analysis, the Particle-Set Identification method, is presented. The method allows to reconstruct moments of multiplicity distribution of identified particles. The difficulty the method copes with is due to incomplete particle identification – a particle mass is frequently determined with a resolution which does not allow for a unique determination of the particle type. Within this method the moments of order k are calculated from mean multiplicities of k-particle sets of a given type. The Particle-Set Identification method remains valid even in the case of correlations between mass measurements for different particles. This distinguishes it from the Identity method introduced by us previously to solve the problem of incomplete particle identification in studies of particle fluctuations.
Deubiquitinases (DUBs) are vital for the regulation of ubiquitin signals, and both catalytic activity of and target recruitment by DUBs need to be tightly controlled. Here, we identify asparagine hydroxylation as a novel posttranslational modification involved in the regulation of Cezanne (also known as OTU domain–containing protein 7B (OTUD7B)), a DUB that controls key cellular functions and signaling pathways. We demonstrate that Cezanne is a substrate for factor inhibiting HIF1 (FIH1)- and oxygen-dependent asparagine hydroxylation. We found that FIH1 modifies Asn35 within the uncharacterized N-terminal ubiquitin-associated (UBA)-like domain of Cezanne (UBACez), which lacks conserved UBA domain properties. We show that UBACez binds Lys11-, Lys48-, Lys63-, and Met1-linked ubiquitin chains in vitro, establishing UBACez as a functional ubiquitin-binding domain. Our findings also reveal that the interaction of UBACez with ubiquitin is mediated via a noncanonical surface and that hydroxylation of Asn35 inhibits ubiquitin binding. Recently, it has been suggested that Cezanne recruitment to specific target proteins depends on UBACez. Our results indicate that UBACez can indeed fulfill this role as regulatory domain by binding various ubiquitin chain types. They also uncover that this interaction with ubiquitin, and thus with modified substrates, can be modulated by oxygen-dependent asparagine hydroxylation, suggesting that Cezanne is regulated by oxygen levels.

Abstract
Given the ubiquitous presence of microplastics in aquatic environments, an evaluation of their toxicity is essential. Microplastics are a heterogeneous set of materials that differ not only in particle properties, like size and shape, but also in chemical composition, including polymers, additives and side products. Thus far, it remains unknown whether the plastic chemicals or the particle itself are the driving factor for microplastic toxicity. To address this question, we exposed Daphnia magna for 21 days to irregular polyvinyl chloride (PVC), polyurethane (PUR) and polylactic acid (PLA) microplastics as well as to natural kaolin particles in high concentrations (10, 50, 100, 500 mg/L, ≤ 59 μm) and different exposure scenarios, including microplastics and microplastics without extractable chemicals as well as the extracted and migrating chemicals alone. All three microplastic types negatively affected the life-history of D. magna. However, this toxicity depended on the endpoint and the material. While PVC had the largest effect on reproduction, PLA reduced survival most effectively. The latter indicates that bio-based and biodegradable plastics can be as toxic as their conventional counterparts. The natural particle kaolin was less toxic than microplastics when comparing numerical concentrations. Importantly, the contribution of plastic chemicals to the toxicity was also plastic type-specific. While we can attribute effects of PVC to the chemicals used in the material, effects of PUR and PLA plastics were induced by the mere particle. Our study demonstrates that plastic chemicals can drive microplastic toxicity. This highlights the importance of considering the individual chemical composition of plastics when assessing their environmental risks. Our results suggest that less studied polymer types, like PVC and PUR, as well as bioplastics are of particular toxicological relevance and should get a higher priority in ecotoxicological studies.
Decline in physical activity in the weeks preceding sustained ventricular arrhythmia in women

Background: Heightened risk of cardiac arrest following physical exertion has been reported. Among patients with an implantable defibrillator, an appropriate shock for sustained ventricular arrhythmia was preceded by a retrospective self-report of engaging in mild-to-moderate physical activity. Previous studies evaluating the relationship between activity and sudden cardiac arrest lacked an objective measure of physical activity and women were often underrepresented.
Objective: To determine the relationship between physical activity, recorded by accelerometer in a wearable cardioverter-defibrillator (WCD), and sustained ventricular arrhythmia among female patients.
Methods: A dataset of female adult patients prescribed a WCD for a diagnosis of myocardial infarction or dilated cardiomyopathy was compiled from a commercial database. Curve estimation, to include linear and nonlinear interpolation, was applied to physical activity as a function of time (days before arrhythmia).
Results: Among women who received an appropriate WCD shock for sustained ventricular arrhythmia (N = 120), a quadratic relationship between time and activity was present prior to shock. Physical activity increased starting at the beginning of the 30-day period up until day -16 (16 days before the ventricular arrhythmia) when activity begins to decline.
Conclusion: For patients who received treatment for sustained ventricular arrhythmia, a decline in physical activity was found during the 2 weeks preceding the arrhythmic event. Device monitoring for a sustained decline in physical activity may be useful to identify patients at near-term risk of a cardiac arrest.
Aim: To assess volumetric tissue changes at peri‐implantitis sites following combined surgical therapy of peri‐implantitis over a 6‐month follow‐up period.
Materials and Methods: Twenty patients (n = 28 implants) diagnosed with peri‐implantitis underwent access flap surgery, implantoplasty at supracrestally or bucally exposed implant surfaces and augmentation at intra‐bony components using a natural bone mineral and application of a native collagen membrane during clinical routine treatments. The peri‐implant region of interest (ROI) was intra‐orally scanned pre‐operatively (S0), and after 1 (S1) and 6 (S2) months following surgical therapy. Digital files were converted to standard tessellation language (STL) format for superimposition and assessment of peri‐implant volumetric variations between time points. The change in thickness was assessed at a standardized ROI, subdivided into three equidistant sections (i.e. marginal, medial and apical). Peri‐implant soft tissue contour area (STCA) (mm2) and its corresponding contraction rates (%) were also assessed.
Results: Peri‐implant tissues revealed a mean thickness change (loss) of −0.11 and −0.28 mm at 1 and 6 months. S0 to S1 volumetric variations pointed to a thickness change of −0.46, 0.08 and 0.4 mm at marginal, medial and apical regions, respectively. S0 to S2 analysis exhibited corresponding thickness changes of −0.61, −0.25 and −0.09 mm, respectively. The thickness differences between the areas were statistically significant at both time periods. The mean peri‐implant STCA totalled to 189.2, 175 and 158.9 mm2 at S0, S1 and S2, showing a significant STCA contraction rate of 7.9% from S0 to S1 and of 18.5% from S0 to S2. Linear regression analysis revealed a significant association between the pre‐operative width of keratinized mucosa (KM) and STCA contraction rate.
Conclusions: The peri‐implant mucosa undergoes considerable volumetric changes after combined surgical therapy. However, tissue contraction appears to be influenced by the width of KM.
In resource-limited or point-of-care settings, rapid diagnostic tests (RDTs), that aim to simultaneously detect HIV antibodies and p24 capsid (p24CA) antigen with high sensitivity, can pose important alternatives to screen for early infections. We evaluated the performance of the antibody and antigen components of the old and novel version of the Determine™ HIV-1/2 Ag/Ab Combo RDTs in parallel to quantifications in a fourth-generation antigen/antibody immunoassay (4G-EIA), p24CA antigen immunoassay (p24CA-EIA), immunoblots, and nucleic acid quantification. We included plasma samples of acute, treatment-naïve HIV-1 infections (Fiebig stages I–VI, subtypes A1, B, C, F, CRF02_AG, CRF02_AE, URF) or chronic HIV-1 and HIV-2 infections. The tests’ antigen component was evaluated also for a panel of subtype B HIV-1 transmitted/founder (T/F) viruses, HIV-2 strains and HIV-2 primary isolates. Furthermore, we assessed the analytical sensitivity of the RDTs to detect p24CA using a highly purified HIV-1NL4-3 p24CA standard. We found that 77% of plasma samples from acutely infected, immunoblot-negative HIV-1 patients in Fiebig stages II–III were identified by the new RDT, while only 25% scored positive in the old RDT. Both RDTs reacted to all samples from chronically HIV-1-infected and acutely HIV-1-infected patients with positive immunoblots. All specimens from chronically infected HIV-2 patients scored positive in the new RDT. Of note, the sensitivity of the RDTs to detect recombinant p24CA from a subtype B virus ranged between 50 and 200 pg/mL, mirrored also by the detection of HIV-1 T/F viruses only at antigen concentrations tenfold higher than suggested by the manufacturer. The RTD failed to recognize any of the HIV-2 viruses tested. Our results indicate that the new version of the Determine™ HIV-1/2 Ag/Ab Combo displays an increased sensitivity to detect HIV-1 p24CA-positive, immunoblot-negative plasma samples compared to the precursor version. The sensitivity of 4G-EIA and p24CA-EIA to detect the major structural HIV antigen, and thus to diagnose acute infections prior to seroconversion, is still superior.
Mitochondria have a central role in regulating a range of cellular activities and host responses upon bacterial infection. Multiple pathogens affect mitochondria dynamics and functions to influence their intracellular survival or evade host immunity. On the other side, major host responses elicited against infections are directly dependent on mitochondrial functions, thus placing mitochondria centrally in maintaining homeostasis upon infection. In this review, we summarize how different bacteria and viruses impact morphological and functional changes in host mitochondria and how this manipulation can influence microbial pathogenesis as well as the host cell metabolism and immune responses.
In this proceeding, we review our recent work using deep convolutional neural network (CNN) to identify the nature of the QCD transition in a hybrid modeling of heavy-ion collisions. Within this hybrid model, a viscous hydrodynamic model is coupled with a hadronic cascade “after-burner”. As a binary classification setup, we employ two different types of equations of state (EoS) of the hot medium in the hydrodynamic evolution. The resulting final-state pion spectra in the transverse momentum and azimuthal angle plane are fed to the neural network as the input data in order to distinguish different EoS. To probe the effects of the fluctuations in the event-by-event spectra, we explore different scenarios for the input data and make a comparison in a systematic way. We observe a clear hierarchy in the predictive power when the network is fed with the event-by-event, cascade-coarse-grained and event-fine-averaged spectra. The carefully-trained neural network can extract high-level features from pion spectra to identify the nature of the QCD transition in a realistic simulation scenario.
Previous studies reported on the safety and applicability of mesenchymal stem/stromal cells (MSCs) to ameliorate pulmonary inflammation in acute respiratory distress syndrome (ARDS). Thus, multiple clinical trials assessing the potential of MSCs for COVID-19 treatment are underway. Yet, as SARS-inducing coronaviruses infect stem/progenitor cells, it is unclear whether MSCs could be infected by SARS-CoV-2 upon transplantation to COVID-19 patients. We found that MSCs from bone marrow, amniotic fluid, and adipose tissue carry angiotensin-converting enzyme 2 and transmembrane protease serine subtype 2 at low levels on the cell surface under steady-state and inflammatory conditions. We did not observe SARS-CoV-2 infection or replication in MSCs at steady state under inflammatory conditions, or in direct contact with SARS-CoV-2-infected Caco-2 cells. Further, indoleamine 2,3-dioxygenase 1 production in MSCs was not impaired in the presence of SARS-CoV-2. We show that MSCs are resistant to SARS-CoV-2 infection and retain their immunomodulation potential, supporting their potential applicability for COVID-19 treatment.
Knowledge of consumers' willingness to pay (WTP) is a prerequisite to profitable price-setting. To gauge consumers' WTP, practitioners often rely on a direct single question approach in which consumers are asked to explicitly state their WTP for a product. Despite its popularity among practitioners, this approach has been found to suffer from hypothetical bias. In this paper, we propose a rigorous method that improves the accuracy of the direct single question approach. Specifically, we systematically assess the hypothetical biases associated with the direct single question approach and explore ways to de-bias it. Our results show that by using the de-biasing procedures we propose, we can generate a de-biased direct single question approach that is accurate enough to be useful for managerial decision-making. We validate this approach with two studies in this paper.
Aims: Acetylsalicylic acid (ASA) is widely used for the prevention of atherothrombotic events in patients with chronic coronary artery disease (CAD) and peripheral artery disease (PAD), but the risk of vascular events remains high. We aimed at identifying randomised controlled trials (RCTs) on antithrombotic treatments in patients with chronic CAD or PAD.
Methods: Searches were conducted on MEDLINE, EMBASE, and CENTRAL on March 1st, 2018. This systematic review (SR) uses a narrative synthesis to summarize the evidence for the efficacy and safety of antiplatelet and anticoagulant therapies in the population of both chronic CAD or PAD patients.
Results: Four RCTs from 27 publications were included. Study groups included 15,603 to 27,395 patients. ASA alone was the most extensively studied (n = 3); other studies included rivaroxaban with or without ASA (n = 1), vorapaxar alone (n = 1), and clopidogrel with (n = 1) or without ASA (n = 1). Clopidogrel alone and clopidogrel plus ASA compared to ASA presented similar efficacy with comparable safety profile. Rivaroxaban plus ASA significantly reduced the risk of the composite of cardiovascular death, myocardial infarction, and stroke compared to ASA alone, although major bleeding with rivaroxaban plus ASA increased.
Conclusion: There is limited and heterogeneous evidence on the prevention of atherothrombotic events in patients with chronic CAD or PAD. Clopidogrel alone and clopidogrel plus ASA did not demonstrate superiority over ASA alone. A combination of rivaroxaban plus ASA may offer significant additional benefit in reducing cardiovascular outcomes, yet it may increase the risk of bleeding, compared to ASA alone.
Objectives: Evaluation of surgical and non-surgical air-polishing in vitro efficacy for implant surface decontamination.
Material and methods: One hundred eighty implants were distributed to three differently angulated bone defect models (30°, 60°, 90°). Biofilm was imitated using indelible red color. Sixty implants were used for each defect, 20 of which were air-polished with three different types of glycine air powder abrasion (GAPA1–3) combinations. Within 20 equally air-polished implants, a surgical and non-surgical (with/without mucosa mask) procedure were simulated. All implants were photographed to determine the uncleaned surface. Changes in surface morphology were assessed using scanning electron micrographs (SEM).
Results: Cleaning efficacy did not show any significant differences between GAPA1–3 for surgical and non-surgical application. Within a cleaning method significant (p < 0.001) differences for GAPA2 between 30° (11.77 ± 2.73%) and 90° (7.25 ± 1.42%) in the non-surgical and 30° (8.26 ± 1.02%) and 60° (5.02 ± 0.84%) in the surgical simulation occurred. The surgical use of air-polishing (6.68 ± 1.66%) was significantly superior (p < 0.001) to the non-surgical (10.13 ± 2.75%). SEM micrographs showed no surface damages after use of GAPA.
Conclusions: Air-polishing is an efficient, surface protective method for surgical and non-surgical implant surface decontamination in this in vitro model. No method resulted in a complete cleaning of the implant surface.
Clinical relevance: Air-polishing appears to be promising for implant surface decontamination regardless of the device.
Objectives: To immunohistochemically characterize and correlate macrophage M1/M2 polarization status with disease severity at peri-implantitis sites.
Materials and methods: A total of twenty patients (n = 20 implants) diagnosed with peri-implantitis (i.e., bleeding on probing with or without suppuration, probing depths ≥ 6 mm, and radiographic marginal bone loss ≥ 3 mm) were included. The severity of peri-implantitis was classified according to established criteria (i.e., slight, moderate, and advanced). Granulation tissue biopsies were obtained during surgical therapy and prepared for immunohistological assessment and macrophage polarization characterization. Macrophages, M1, and M2 phenotypes were identified through immunohistochemical markers (i.e., CD68, CD80, and CD206) and quantified through histomorphometrical analyses.
Results: Macrophages exhibiting a positive CD68 expression occupied a mean proportion of 14.36% (95% CI 11.4–17.2) of the inflammatory connective tissue (ICT) area. Positive M1 (CD80) and M2 (CD206) macrophages occupied a mean value of 7.07% (95% CI 5.9–9.4) and 5.22% (95% CI 3.8–6.6) of the ICT, respectively. The mean M1/M2 ratio was 1.56 (95% CI 1–12–1.9). Advanced peri-implantitis cases expressed a significantly higher M1 (%) when compared with M2 (%) expression. There was a significant correlation between CD68 (%) and M1 (%) expression and probing depth (PD) values.
Conclusion: The present immunohistochemical analysis suggests that macrophages constitute a considerable proportion of the inflammatory cellular composition at peri-implantitis sites, revealing a significant higher expression for M1 inflammatory phenotype at advanced peri-implantitis sites, which could possibly play a critical role in disease progression.
Clinical relevance: Macrophages have critical functions to establish homeostasis and disease.
