The intrinsic complexity of the brain can lead one to set aside issues related to its relationships with the body, but the field of embodied cognition emphasizes that understanding brain function at the system level requires one to address the role of the brain-body interface. It has only recently been appreciated that this interface performs huge amounts of computation that does not have to be repeated by the brain, and thus affords the brain great simplifications in its representations. In effect the brain’s abstract states can refer to coded representations of the world created by the body. But even if the brain can communicate with the world through abstractions, the severe speed limitations in its neural circuitry mean that vast amounts of indexing must be performed during development so that appropriate behavioral responses can be rapidly accessed. One way this could happen would be if the brain used a decomposition whereby behavioral primitives could be quickly accessed and combined. This realization motivates our study of independent sensorimotor task solvers, which we call modules, in directing behavior. The issue we focus on herein is how an embodied agent can learn to calibrate such individual visuomotor modules while pursuing multiple goals. The biologically plausible standard for module programming is that of reinforcement given during exploration of the environment. However this formulation contains a substantial issue when sensorimotor modules are used in combination: The credit for their overall performance must be divided amongst them. We show that this problem can be solved and that diverse task combinations are beneficial in learning and not a complication, as usually assumed. Our simulations show that fast algorithms are available that allot credit correctly and are insensitive to measurement noise.
Records of Nesticodes rufipes (Lucas, 1846) from Saxony, Germany are presented. The three females of Nesticodes rufipes were found associated with the zoo trade in the cities of Chemnitz and Plauen in cricket boxes. Female genitalia are illustrated.
The canopy spiders of the floodplain forest in Leipzig have become a focus of ecological studies in recent years. In 2006 we sampled 30 tree canopies in the ‘Burgaue’ nature reserve with pyrethrum knock-down fogging, recording 502 adult spiders belonging to 48 species and 11 families. Based on these data and the results of a previous fogging study, the studied spider community was dominated by forest and forest-edge species with a preference for the shrub and canopy strata as well as by spiders of the web spider feeding guild. The community structure was typical for arboreal spider communities from northern temperate forests but very different from communities in the tropics. Species richness and evenness were similar to the old growth near-primary Białowieża Forest in Poland. The checklist of 96 canopy spider species of the floodplain forest of Leipzig includes 54 additions to the spider fauna of Leipzig and vicinity by recent canopy studies and eight first canopy records for Leipzig from our field work. The theridiid Dipoena torva (Thorell, 1875) was recorded for the first time in Saxony. The floodplain forest of Leipzig sustains a large and species-rich arboreal spider community and is thus a valuable habitat for a large proportion of endangered species (12%).
Cryptachaea blattea : eine weitere nach Deutschland eingeschleppte Spinnenart (Araneae: Theridiidae)

Cryptachaea blattea (Urquhart, 1886) has been recorded for the first time from Germany (Nordstemmen, rural district of Hildesheim, Lower Saxony). One male was found by means of pitfall traps in a tree and shrub nursery. The species was most likely introduced with plants or cargo. Information about its appearance, habitat, and distribution is given.
Pitfall traps were positioned for the investigation of the spider fauna at the northern and southern slopes of three mountain ridges (Chilchberg, Riedberg, and Buechenberg, municipalities Nunningen and Zullwil, canton Solothurn, Switzerland) within the Swiss Jura Mountains. The temperature in the upper litter was measured at three hour intervals. Independent of the weather more or less clear differences between northern and southern slopes could be observed. Maximum day temperature fluctuations of 15.8 °C were measured. There were no significant differences in spider communities based on quantitative comparison methods. However, a qualitative analysis showed major differences in species composition. More than 50% of all species per investigation area showed clear preferences for the northern or the southern slope, with more then two thirds of the individuals only found on either the north or south slopes.
This study presents the development and mapping of simple sequence repeat (SSR) and single nucleotide polymorphism (SNP) markers in chickpea. The mapping population is based on an inter-specific cross between domesticated and non-domesticated genotypes of chickpea (Cicer arietinum ICC 4958 × C. reticulatum PI 489777). This same population has been the focus of previous studies, permitting integration of new and legacy genetic markers into a single genetic map. We report a set of 311 novel SSR markers (designated ICCM—ICRISAT chickpea microsatellite), obtained from an SSR-enriched genomic library of ICC 4958. Screening of these SSR markers on a diverse panel of 48 chickpea accessions provided 147 polymorphic markers with 2–21 alleles and polymorphic information content value 0.04–0.92. Fifty-two of these markers were polymorphic between parental genotypes of the inter-specific population. We also analyzed 233 previously published (H-series) SSR markers that provided another set of 52 polymorphic markers. An additional 71 gene-based SNP markers were developed from transcript sequences that are highly conserved between chickpea and its near relative Medicago truncatula. By using these three approaches, 175 new marker loci along with 407 previously reported marker loci were integrated to yield an improved genetic map of chickpea. The integrated map contains 521 loci organized into eight linkage groups that span 2,602 cM, with an average inter-marker distance of 4.99 cM. Gene-based markers provide anchor points for comparing the genomes of Medicago and chickpea, and reveal extended synteny between these two species. The combined set of genetic markers and their integration into an improved genetic map should facilitate chickpea genetics and breeding, as well as translational studies between chickpea and Medicago.
The impact of shift work induced chronic circadian disruption on IL-6 and TNF-α immune responses

Aim: Sleep disturbances induce proinflammatory immune responses, which might increase cardiovascular disease risk. So far the effects of acute sleep deprivation and chronic sleep illnesses on the immune system have been investigated. The particular impact of shift work induced chronic circadian disruption on specific immune responses has not been addressed so far.
Methods: Pittsburgh-Sleep-Quality-Index (PSQI) questionnaire and blood sampling was performed by 225 shift workers and 137 daytime workers. As possible markers the proinflammatory cytokines IL-6 and TNF-alpha and lymphocyte cell count were investigated. A medical examination was performed and biometrical data including age, gender, height, weight, waist and hip circumference and smoking habits were collected by a structured interview.
Results: Shift workers had a significantly higher mean PSQI score than day workers (6.73 vs. 4.66; p < 0.001). Day workers and shift workers had similar serum levels of IL-6 (2.30 vs. 2.67 resp.; p = 0.276), TNF-alpha (5.58 vs. 5.68, resp.; p = 0.841) or lymphocytes count (33.68 vs. 32.99, resp.; p = 0.404). Furthermore there were no differences in cytokine levels (IL-6 p = 0.761; TNF-alpha p = 0.759) or lymphocyte count (p = 0.593) comparing the sleep quality within the cohorts. When this calculation of sleep quality was stratified by shift and day workers irrespective of their sleep quality day workers and shift workers had similar serum levels of IL-6, TNF-alpha or lymphocytes count. Multiple linear regression analysis showed a significant correlation of lymphocytes count and smoking habits.
Conclusion: Shift work induces chronic sleep debt. Our data reveals that chronic sleep debt might not always lead to an activation of the immune system, as we did not observe differences in lymphocyte count or level of IL-6 or TNF-alpha serum concentration between shift workers and day workers. Therefore chronic sleep restriction might be eased by a long-term compensating immune regulation which (in healthy) protects against an overstimulation of proinflammatory immune mechanisms and moderates metabolic changes, as they are known from short-term sleep deprivation or sleep related breathing disorders.
Background: Diseases associated with smoking are a foremost cause of premature death in the world, both in developed and developing countries. Eliminating smoking can do more to improve health and prolong life than any other measure in the field of preventive medicine. Today's medical students will play a prominent role in future efforts to prevent and control tobacco use.
Methods: A cross-sectional, self-administered, anonymous survey of fifth-year medical students in Berlin, Germany was conducted in November 2007. The study explored the prevalence of smoking among medical students. We assessed their current knowledge regarding tobacco dependence and the effectiveness of smoking cessation methods. Students' perceived competence to counsel smokers and promote smoking cessation treatments was also explored. Analyses were based on responses from 258 students (86.6% response rate).
Results: One quarter of the medical students surveyed were current smokers. The smoking rate was 22.1% among women, 32.4% among men. Students underestimated smoking-related mortality and the negative effect of smoking on longevity. A considerable number of subjects erroneously assumed that nicotine causes coronary artery disease. Students' overall knowledge of the effectiveness of smoking cessation methods was inadequate. Only one third of the students indicated that they felt qualified to counsel patients about tobacco dependence.
Conclusions: This study reveals serious deficiencies in knowledge and counseling skills among medical students in our sample. The curriculum of every medical school should include a tobacco module. Thus, by providing comprehensive training in nicotine dependence interventions to medical students, smokers will have access to the professional expertise they need to quit smoking.
Family members provide most of the patient care and administer most of the treatments to patients with Alzheimer’s disease (AD). Family caregivers have an important impact on clinical outcomes, such as quality of life (QoL). As a consequence of this service, family caregivers suffer high rates of psychological and physical illness as well as social and financial burdens. Hence, it is important to involve family caregivers in multimodal treatment settings and provide interventions that are both suitable and specifically tailored to their needs. In recent years, several clinical guidelines have been presented worldwide for evidence-based treatment of AD and other forms of dementia. Most of these guidelines have considered family advice as integral to the optimal clinical management of AD. This article reviews current and internationally relevant guidelines with emphasis on recommendations concerning family advice.
Es werden 66 Neu- oder Wiederfunde sowie nicht publizierte Funde von Flechten und flechtenbewohnenden Pilzen in Hessen gemeldet und kurz kommentiert. Sechs Arten sind zugleich Neufunde für Deutschland: Absconditella pauxilla, Acremonium antarcticum, Calicium adaequatum, Psammina inflata, Ramonia chrysophaea und Tubeufia heterodermiae. Die hessische Artenliste umfasst mittlerweile annähernd 1200 Flechten und flechtenbewohnende oder flechtenähnliche Pilze.
Im Jahre 2009 wurde erstmals eine umfassende Bestandsaufnahme der ehemaligen und aktuellen hessischen Vorkommen des inzwischen vom Aussterben bedrohten Lungen-Enzians (Gentiana pneumonanthe) durchgeführt. Dabei stellte sich heraus, dass etwa 85 % der früheren Wuchsorte verloren gingen und alle heutigen Vor-kommen – trotz ihrer Lage in südhessischen Schutzgebieten – auch in jüngster Zeit noch Verluste aufweisen. Für das Überleben dieser sich offenbar nur generativ vermehrenden Art in Hessen ist es deshalb notwendig, dass geeignete Schutz- und Hilfsmaßnahmen zur Sicherung und Entwicklung der letzten Bestände ergriffen werden, wobei unbedingt auch phänologische Aspekte zu berücksichtigen sind.
Die Brackwasser-Armleuchteralge (Chara canescens) und die Faden-Armleuchteralge (Chara filiformis) wurden im Juni 2009 erstmals in Hessen im Borkener See, einem Restgewässer des Braunkohleabbaus in Nordhessen, nachgewiesen. Chara filiformis wurde erstmals in Deutschland in einem künstlichen Gewässer nachgewiesen. Chara canescens kommt hier in einem elektrolytarmen Gewässer vor.
Das Gnadenkraut (Gratiola officinalis) gehört zu den seltensten hessischen Pflanzenarten und gilt seit der ersten hessischen Roten Liste der gefährdeten Pflanzenarten als vom Aussterben bedroht. Im Rahmen der von der Botanischen Ver-einigung für Naturschutz in Hessen durchgeführten Artenhilfsprogramme sollte eine umfassende Bestandsaufnahme durchgeführt und für die letzten Bestände Maßnahmen zur Sicherung und Erhaltung vorgeschlagen werden. Zwar konnten aktuell keine Vor-kommen mehr nachgewiesen werden – die Art muß deshalb für Hessen als verschollen gelten – doch erscheint es möglich, aus der noch vorhandenen Samenbank des letzten bekannten Wuchsortes neue Bestände zu begründen.
Zur Schließung von Kenntnislücken zur Flora der Gewässer Hessens wurden 2009 von der BVNH neun Altarme von Rhein und Main untersucht. Die Ergeb-nisse zeigen, dass viele als sehr selten oder verschollen geltende Gewässermakrophyten noch oder wieder in den Altarmen des hessischen Oberrheinabschnitts zu finden sind. Insgesamt wurden 51 Arten nachgewiesen, darunter fünf Armleuchteralgen.

Análise da macro e da microestrutura de dicionários bilíngues português-alemão / alemão-português

This paper aims at analyzing macro- and microstructural features of Portuguese-German/German-Portuguese bilingual dictionaries. For this purpose, we will discuss the basic parameters that should guide the conception of a bilingual dictionary: the target users, the direction, the function, and the linguistic anisomorphism.
This article discusses the communicative and the intercultural approaches to language teaching and learning. Firstly, it describes the core theoretical principles, goals, and the roles of teachers and learners in both approaches, as well as the way they are treated in different German textbooks. Secondly, it discusses a set of core principles and concepts (‘activity’, ‘interaction’, ‘motivation’ and ‘communicative posture’), as well as the didactic-pedagogical implications of implementing a communicative approach to language teaching and learning. Finally, it includes a reflection about the necessity for the language teacher to develop an intercultural background, as well as the eclectic use of different linguistic theories and language teaching approaches to make the acquisition of communicative and intercultural competence viable.
It could be said that learner’s dictionaries are the most reliable expression of lexicography in terms of providing the necessary tools to help the learning process of a foreign language. This paper analyses three English learner’s dictionaries in order to establish its stylistic patterns as well as to compare them with four German learner’s dictionaries. Undoubtly, the lexicography of English learner’s dictionaries is a model. However, we argue that this model can not be transfered to German lexicography since each language has its own particularities which demand specific solutions.
This contribution focuses on the conditions of literary production in Germany during the Nazi regime. The article analyzes the work of writer Ricarda Huch as one of the representatives of the so-called "Inner Emigration", literary trend promoted by writers that were not placed to serve the interests of the State, but instead sought to convey - through their work - humanistic values and critics of the status quo, despite the censorship and often putting their lives at risk. The poetic anthology Herbstfeuer ("Fire of Autumn"), published in 1944, is one of the most significant examples of the literature of resistance under the yoke of Nazism.
This study presents a global scale analysis of cropping intensity, crop duration and fallow land extent computed by using the global dataset on monthly irrigated and rainfed crop areas MIRCA2000. MIRCA2000 was mainly derived from census data and crop calendars from literature. Global cropland extent was 16 million km2 around the year 2000 of which 4.4 million km2 (28%) was fallow, resulting in an average cropping intensity of 0.82 for total cropland extent and of 1.13 when excluding fallow land. The lowest cropping intensities related to total cropland extent were found for Southern Africa (0.45), Central America (0.49) and Middle Africa (0.54), while highest cropping intensities were computed for Eastern Asia (1.04) and Southern Asia (1.0). In remote or arid regions where shifting cultivation is practiced, fallow periods last 3–10 years or even longer. In contrast, crops are harvested two or more times per year in highly populated, often irrigated tropical or subtropical lowlands where multi-cropping systems are common. This indicates that intensification of agricultural land use is a strategy that may be able to significantly improve global food security. There exist large uncertainties regarding extent of cropland, harvested crop area and therefore cropping intensity at larger scales. Satellite imagery and remote sensing techniques provide opportunities for decreasing these uncertainties and to improve the MIRCA2000 inventory.
We report on screening tests of 66 extracts obtained from 35 marine sponge species from the Caribbean Sea (Curaçao) and from eight species from the Great Barrier Reef (Lizard Island). Extracts were prepared in aqueous and organic solvents and were tested for hemolytic, hemagglutinating, antibacterial and anti-acetylcholinesterase (AChE) activities, as well as their ability to inhibit or activate cell protein phosphatase 1 (PP1). The most interesting activities were obtained from extracts of Ircinia felix, Pandaros acanthifolium, Topsentia ophiraphidites, Verongula rigida and Neofibularia nolitangere. Aqueous and organic extracts of I. felix and V. rigida showed strong antibacterial activity. Topsentia aqueous and some organic extracts were strongly hemolytic, as were all organic extracts from I. felix. The strongest hemolytic activity was observed in aqueous extracts from P. acanthifolium. Organic extracts of N. nolitangere and I. felix inhibited PP1. The aqueous extract from Myrmekioderma styx possessed the strongest hemagglutinating activity, whilst AChE inhibiting activity was found only in a few sponges and was generally weak, except in the methanolic extract of T. ophiraphidites.
Spontaneous symmetry breaking is a general principle that constitutes the underlying concept of a vast number of physical phenomena ranging from ferromagnetism and superconductivity in condensed matter physics to the Higgs mechanism in the standard model of elementary particles. I focus on manifestations of spontaneously broken symmetries in systems that are not Lorentz invariant, which include both nonrelativistic systems as well as relativistic systems at nonzero density, providing a self-contained review of the properties of spontaneously broken symmetries specific to such theories. Topics covered include: (i) Introduction to the mathematics of spontaneous symmetry breaking and the Goldstone theorem. (ii) Minimization of Higgs-type potentials for higher-dimensional representations. (iii) Counting rules for Nambu–Goldstone bosons and their dispersion relations. (iv) Construction of effective Lagrangians. Specific examples in both relativistic and nonrelativistic physics are worked out in detail.
Surface measurements of aerosol and ice nuclei (IN) at a Central European mountain site during an episode of dust transport from the Sahara are presented. Transport is simulated by the Eulerian regional dust model DREAM. Ice nuclei and mineral dust are significantly correlated. The highest correlation is found between IN concentration and aerosol surface area. The ice nucleating characteristics of the aerosol with respect to temperature and supersaturation are similar during the dust episode than during the course of the year. This suggests that dust is always a dominant constituent of ice nucleating aerosols in Central Europe.
Surface measurements of aerosol and ice nuclei (IN) at a Central European mountain site during an episode of dust transport from the Sahara are presented. Ice nuclei were sampled by electrostatic precipitation on silicon wafers and were analyzed in an isothermal static vapor diffusion chamber. The transport of mineral dust is simulated by the Eulerian regional dust model DREAM. Ice nuclei and mineral dust are significantly correlated, in particular IN number concentration and aerosol surface area. The ice nucleating characteristics of the aerosol as analyzed with respect to temperature and supersaturation are similar during the dust episode than during the course of the year. This suggests that dust may be a main constituent of ice nucleating aerosols in Central Europe.
This paper presents an analysis of the recent tropospheric molecular hydrogen (H2) budget with a particular focus on soil uptake and surface emissions. A variational inversion scheme is combined with observations from the RAMCES and EUROHYDROS atmospheric networks, which include continuous measurements performed between mid-2006 and mid-2009. Net H2 surface flux, soil uptake distinct from surface emissions and finally, soil uptake, biomass burning, anthropogenic emissions and N2 fixation-related emissions separately were inverted in several scenarios. The various inversions generate an estimate for each term of the H2 budget. The net H2 flux per region (High Northern Hemisphere, Tropics and High Southern Hemisphere) varies between −8 and 8 Tg yr−1. The best inversion in terms of fit to the observations combines updated prior surface emissions and a soil deposition velocity map that is based on soil uptake measurements. Our estimate of global H2 soil uptake is −59 ± 4.0 Tg yr−1. Forty per cent of this uptake is located in the High Northern Hemisphere and 55% is located in the Tropics. In terms of surface emissions, seasonality is mainly driven by biomass burning emissions. The inferred European anthropogenic emissions are consistent with independent H2 emissions estimated using a H2/CO mass ratio of 0.034 and CO emissions considering their respective uncertainties. To constrain a more robust partition of H2 sources and sinks would need additional constraints, such as isotopic measurements.
In this paper we present evidence that the observed increase in tropical upwelling after the year 2000 may be attributed to a change in the Brewer-Dobson circulation pattern. For this purpose, we use the concept of transit times derived from residual circulation trajectories and different in-situ measurements of ozone and nitrous dioxide. Observations from the Canadian midlatitude ozone profile record, probability density functions of in-situ N2O observations and a shift of the N2O-O3 correlation slopes, taken together, indicate that the increased upwelling in the tropics after the year 2000 appears to have triggered an intensification of tracer transport from the tropics into the extratropics in the lower stratosphere below about 500 K. This finding is corroborated by the fact that transit times along the shallow branch of the residual circulation into the LMS have decreased for the same time period (1993–2003). On a longer time scale (1979–2009), the transit time of the shallow residual circulation branch show a steady decrease of about −1 month/decade over the last 30 years, while the transit times of the deep branch remain unchanged. This highlights the fact that a change in the upwelling across the tropical tropopause is not a direct indicator for changes of the whole Brewer-Dobson circulation.
Palystes kreutzmanni sp. n. is described from habitats close to Kleinmond, in the Western Cape Province, South Africa. Spiders of this new species live in the typical fynbos vegetation of the Western Cape region. They build retreats between apical leaves of Leucadendron bushes. The systematic position of Palystes kreutzmanni sp. n. is discussed. Male and female show characters of different species groups, especially the female copulatory organ seems to be unique within the genus Palystes L. Koch, 1875.
The design, construction, and commissioning of the ALICE Time-Projection Chamber (TPC) is described. It is the main device for pattern recognition, tracking, and identification of charged particles in the ALICE experiment at the CERN LHC. The TPC is cylindrical in shape with a volume close to 90 m3 and is operated in a 0.5 T solenoidal magnetic field parallel to its axis.
In this paper we describe in detail the design considerations for this detector for operation in the extreme multiplicity environment of central Pb–Pb collisions at LHC energy. The implementation of the resulting requirements into hardware (field cage, read-out chambers, electronics), infrastructure (gas and cooling system, laser-calibration system), and software led to many technical innovations which are described along with a presentation of all the major components of the detector, as currently realized. We also report on the performance achieved after completion of the first round of stand-alone calibration runs and demonstrate results close to those specified in the TPC Technical Design Report.
New projects, services and collaborations have recently brought the infrastructural services for African Studies a big step forward. This report gives an account of new subject gateways and digitisation projects. It discusses recent European cooperation ventures in the field of librarianship. Additionally, new developments and services of the Africa Collection at Frankfurt University Library are presented, which help to address the changing needs of researchers and to handle information overload, while keeping up with the latest developments. Nevertheless, the fragmentation and compartmentalisation of the different services still hinder more integrated information services.
This paper introduces a new methodology for the fabrication of strain-sensor elements for MEMS and NEMS applications based on the tunneling effect in nano-granular metals. The strain-sensor elements are prepared by the maskless lithography technique of focused electron-beam-induced deposition (FEBID) employing the precursor trimethylmethylcyclopentadienyl platinum [MeCpPt(Me)3]. We use a cantilever-based deflection technique to determine the sensitivity (gauge factor) of the sensor element. We find that its sensitivity depends on the electrical conductivity and can be continuously tuned, either by the thickness of the deposit or by electron-beam irradiation leading to a distinct maximum in the sensitivity. This maximum finds a theoretical rationale in recent advances in the understanding of electronic charge transport in nano-granular metals.
Although throughout the history of anthropology the ethnography of urban societies was never an important topic, investigations on cities in Africa contributed to the early theoretical development of urban studies in social sciences. As the ethnography of rural migrants in towns made clear, cultural diversity and creativity are foundational and permanent elements of urban cultures in Africa (and beyond). Currently, two new aspects complement these insights: 1) Different forms of mobility have received a new awareness through the concept of transnationalism. They are much more complex, including not only rural–urban migration, but also urban–urban migration, and migrations with a destination beyond the continent. 2) Urban life-worlds also include the appropriation of globally circulating images and lifestyles, which contribute substantially to the current cultural dynamics of cities in Africa. These two aspects are the reasons for the high complexity of urban contexts in Africa. Therefore, whether it is still appropriate to speak about the “locality” of these life-worlds has become questionable. At the same time, these new aspects explain the self-consciousness of members of urban cultures in Africa. They contribute to the expansive character of these societies and to the impression that cities in Africa host the most innovative and creative societies worldwide.

Background: For M1 pancreatic adenocarcinomas pancreatic resection is usually not indicated. However, in highly selected patients synchronous metastasectomy may be appropriate together with pancreatic resection when operative morbidity is low.
Materials and Methods: From January 1, 2004 to December, 2007 a total of 20 patients with pancreatic malignancies were retrospectively evaluated who underwent pancreatic surgery with synchronous resection of hepatic, adjacent organ, or peritoneal metastases for proven UICC stage IV periampullary cancer of the pancreas. Perioperative as well as clinicopathological parameters were evaluated.
Results: There were 20 patients (9 men, 11 women; mean age 58 years) identified. The primary tumor was located in the pancreatic head (n=9, 45%), in pancreatic tail (n=9, 45%), and in the papilla Vateri (n=2, 10%). Metastases were located in the liver (n=14, 70%), peritoneum (n=5, 25%), and omentum majus (n=2, 10%). Lymphnode metastases were present in 16 patients (80%). All patients received resection of their tumors together with metastasectomy. Pylorus preserving duodenopancreatectomy was performed in 8 patients, distal pancreatectomy in 8, duodenopancreatectomy in 2, and total pancreatectomy in 2. Morbidity was 45% and there was no perioperative mortality. Median postoperative survival was 10.7 months (2.6–37.7 months) which was not significantly different from a matched-pair group of patients who underwent pancreatic resection for UICC adenocarcinoma of the pancreas (median survival 15.6 months; =.1).
Conclusion: Pancreatic resection for M1 periampullary cancer of the pancreas can be performed safely in well-selected patients. However, indication for surgery has to be made on an individual basis.
In the past, the genetically diabetic-obese diabetes/diabetes (db/db) and obese/obese (ob/ob) mouse strains were used to investigate mechanisms of diabetes-impaired wound healing. Here we determined patterns of skin repair in genetically normal C57Bl/6J mice that were fed using a high fat diet (HFD) to induce a diabetes-obesity syndrome. Wound closure was markedly delayed in HFD-fed mice compared to mice which had received a standard chow diet (CD). Impaired wound tissue of HFD mice showed a marked prolongation of wound inflammation. Expression of vascular endothelial growth factor (VEGF) was delayed and associated with the disturbed formation of wound margin epithelia and an impaired angiogenesis in the reduced granulation tissue. Normal wound contraction was retarded and disordered. Wound disorders in obese C57Bl/6J mice were paralleled by a prominent degradation of the inhibitor of NFκB (IκB-α) in the absence of an Akt activation. By contrast to impaired wound conditions in ob/ob mice, late wounds of HFD mice did not develop a chronic inflammatory state and were epithelialized after 11 days of repair. Thus, only genetically obese and diabetic ob/ob mice finally developed chronic wounds and therefore represent a better suited experimental model to investigate diabetes-induced wound healing disorders.
Cells can respond to stress in various ways ranging from the activation of survival pathways to the initiation of cell death that eventually eliminates damaged cells. Whether cells mount a protective or destructive stress response depends to a large extent on the nature and duration of the stress as well as the cell type. Also, there is often the interplay between these responses that ultimately determines the fate of the stressed cell. The mechanism by which a cell dies (i.e., apoptosis, necrosis, pyroptosis, or autophagic cell death) depends on various exogenous factors as well as the cell's ability to handle the stress to which it is exposed. The implications of cellular stress responses to human physiology and diseases are manifold and will be discussed in this review in the context of some major world health issues such as diabetes, Parkinson's disease, myocardial infarction, and cancer.
One of the hallmarks of human cancers is the intrinsic or acquired resistance to apoptosis. Evasion of apoptosis can be part of a cellular stress response to ensure the cell's survival upon exposure to stressful stimuli. Apoptosis resistance may contribute to carcinogenesis, tumor progression, and also treatment resistance, since most current anticancer therapies including chemotherapy as well as radio- and immunotherapies primarily act by activating cell death pathways including apoptosis in cancer cells. Hence, a better understanding of the molecular mechanisms regarding how cellular stress stimuli trigger antiapoptotic mechanisms and how this contributes to tumor resistance to apoptotic cell death is expected to provide the basis for a rational approach to overcome apoptosis resistance mechanisms in cancers.

Editorial: This special issue on Cell Stress and Cell Death is aimed at bringing together recent developments in the fields of cellular stress and cell death and, in particular, the interplay between cell stress responses and cell death. The special issue opens with a review by S. Fulda et al. which provides an overview of how cells can respond to stress in a variety of ways ranging from the activation of survival pathways to the initiation of cell death that eventually eliminates damaged cells. Whether cells mount a protective response or succumb to death depends to a large extent on the nature and duration of the stress as well as the cell type. For example, milder stresses can lead to protection through activation of the heat shock response or the unfolded protein response (UPR). This review also describes several types of cell death (e.g., apoptosis, necrosis, pyroptosis, or autophagic cell death) and the mechanism by which a cell dies often depends on various exogenous factors as well as the cell’s ability to handle the stress to which it is exposed. The implications of cellular stress responses for human physiology and disease are multifold and are discussed in this review in the context of some major world health issues such as diabetes, Parkinson’s disease, myocardial infarction, and cancer. ...
An information system is more than just the information technology; it is the system that emerges from the complex interactions and relationships between the information technology and the organization. However, what impact information technology has on an organization and how organizational structures and organizational change influence information technology remains an open question. We propose a theory to explain how communication structures emerge and adapt to environmental changes. We operationalize the interplay of information technology and organization as language communities whose members use and develop domain-specific languages for communication. Our theory is anchored in the philosophy of language. In developing it as an emergent perspective, we argue that information systems are self-organizing and that control of this ability is disseminated throughout the system itself, to the members of the language community. Information technology influences the dynamics of this adaptation process as a fundamental constraint leading to perturbations for the information system. We demonstrate how this view is separated from the entanglement in practice perspective and show that this understanding has far-reaching consequences for developing, managing, and examining information systems.
Theory building is not only underdeveloped in IT services management research, but in
general in IS. Given the paradigm shift that comes from the development away from a
networked economy towards a network economy, the lack of spending enough attention to
theorizing in IS becomes even more obvious. In the light of other "megatrends" in IS
research, such as the increasing professionalization and use of statistical methods and the
exploitation of extremely large sets of data (often harvested from social media sites), we
might lose interest in theorizing in the presence of the tremendous amount of available
empirical data. In this position paper, the author advocates that services science researchers
should focus on rigor and relevance in their research approaches.
Ao apelo da Universidade de Coimbra para que, no âmbito da sua semana Cultural, as Faculdades e outros organismos que nela se acolhem glosem o tema da “Imaginação”, responde o Instituto de Estudos Alemães com o colóquio Imaginação do mesmo: a diferença na repetição. No texto que enquadra o conjunto de contributos que hoje aqui se apresentam, se afirma, a dado passo, que “Há inovação e transformação não na impossível invenção de uma origem, mas sim na capacidade de articulação produtiva”. Pode esta ser uma boa síntese daquilo que a moderna descendência da espécie homo leva a cabo, enquanto homo sapiens (sapiens) e, portanto, homo loquens, no exercício da faculdade que é, a um tempo, o seu mais poderoso instrumento de cognição e o mais eficaz e económico veículo de comunicação – competição e cooperação são, recordo, os principais pilares em que assenta o processo da sua sobrevivência enquanto espécie.

Zarathustra in den Tropen : der Text als Spirale der Wiederholung in Nietzsche und Robert Müller

Der Einfluss Nietzsches auf die Autoren des Fin-de-Siècle und des so genannten „Expressionistischen Jahrzehnts“ wird unumstritten von der Kritik akzeptiert. Jedoch beschränkt sich diese Einschätzung auch in den jüngsten Abhandlungen (Ester 2001) aufgrund von Thesen aus den 70er Jahren (zum Beispiel die des kanonischen Bandes von Martens von 1971) auf den Parameter des Vitalismus. Die Untersuchungen zur Rezeption Nietzsches in dieser Zeit konzentrieren sich größtenteils auf 'Also sprach Zarathustra' und ignorieren oder unterschätzen somit andere philosophische Texte des Autors. Auf jeden Fall kann man den deutlichen Einfluss Nietzsches auf den deutschsprachigen Modernismus weder auf den 'Zarathustra' und sein vorherrschendes Thema den Übermenschen, noch auf die metaphysische Dimension seiner Philosophie beschränken, ein Umstand, den auch das fiktionale und essayistische Werk des Wiener Autors Robert Müller (1887-1924) musterhaft unter Beweis stellt. 'Der Mythos der Reise. Urkunden eines deutschen Ingenieurs' (1915) ist ein Beispiel für einen essayistischen Roman, der nicht nur direkt von der Philosophie Nietzsches, sondern auch gerade von der jeweiligen begrifflichen und diskursiven Methodologie sowie dem jeweiligen textuellen Konzept profitiert. Es handelt sich damit um Faktoren, die auch unter dem Zeichen des Essayismus gesehen werden müssen.

"Navigare necesse est" [...] Ao longo dos tempos, esta expressão de ousadia conservou-se no discurso literário da cultura ocidental. No século XX, serviram-se dela, por exemplo, dois autores indelevelmente inscritos na memória cultural da língua portuguesa: o poeta Fernando Pessoa, como vimos, e ainda o cantautor Caetano Veloso. […] Nesta linha de apropriação se situa também o escritor austríaco Stefan Zweig quando, em 1937, intitula “Navigare necesse est” o capítulo introdutório da biografia “Magellan. Der Mann und seine Tat”. Ao contrário dos autores de língua portuguesa acima mencionados, Zweig adopta apenas o primeiro segmento do axioma, pois no fenómeno de intertextualidade [...]. Zweig aproveita a expressão no enquadramento semântico original – as navegações –, para a aplicar à linhagem de bravos homens do mar e de eruditos cosmógrafos que tornaram possível a era dos Descobrimentos. Nesse capítulo inicial da narração biográfica, a voz autoral traça uma panorâmica da primeira fase das Descobertas, mostrando como o móbil económico, que desencadeou todo o processo, acabou por trazer à Humanidade ganhos de outra natureza. Assim, os Descobrimentos não terão permitido apenas chegar à Índia e às tão desejadas especiarias: a demanda corajosa e continuada ter-se-á justificado principalmente por desafiar os mitos do oceano, afastar os medos do mar e alargar a nossa dimensão do mundo.
Passaram já mais de vinte anos sobre a apresentação no Centro Pompidou, em Paris, da exposição 'Vienne, 1880-1938, naissance d'un siècle'. Tratou-se, como alguns se recordarão, de um acontecimento de grande repercussão em toda a Europa. A definitiva consolidação do mito cultural da “Viena de 1900” está estreitamente associada a este acontecimento. A ressonância do evento, contudo, não poderá ser cabalmente entendida se não se tiver presente que, em meados dos anos oitenta a controvérsia sobre o pós-modernismo estava no auge. A verdade é que o comissário da exposição, Jean Clair, programou-a expressamente como uma espécie de manifesto pós-moderno, à luz de uma interpretação da modernidade vienense como um sítio arqueológico maior da pós-modernidade. […] É evidente que esta afirmação distintamente pós-histórica e pós-filosófica se baseia numa imagem da 'Wiener Moderne' como lugar de uma critica implícita à narrativa do progresso como perpétua inovação. A actualidade da Viena de 1900 reside, aos olhos de Jean Clair, justamente, numa específica noção de temporalidade que é hostil à ideia do novo como ruptura irreversível […]. É este, de facto, o núcleo da “hipótese vienense”, para usar a expressão proposta na época por Paolo Portoghesi, um dos mais influentes teóricos da arquitectura pós-modernista nos anos oitenta (Portoghesi 1984). Não vou ocupar-me aqui dos equívocos, por vezes grotescos, gerados em torno dessa hipótese no âmbito da teoria do pós-modernismo. Vou, simplesmente, concentrar-me no conceito de repetição e nas suas consequências para um entendimento mais lato do paradigma modernista, já que, na verdade, a “hipótese vienense”, as lições literalmente excêntricas da 'Wiener Moderne', representam, no fundamental, a percepção do carácter aberto e plural das respostas modernistas à condição moderna.
O romance em apreço poderá […] ser compreendido com o resultado desta duplicidade identitária e interpretado como uma metáfora da amnésia social generalizada que o autor observa na sociedade onde actua. Neste texto é sua intenção criticar a ausência de altruísmo, a indiferença, o preconceito e as generalizações a que muitas das personagens do romance são sujeitas. Enquanto algumas destas figuras, especialmente as que pertencem a minorias negligenciadas, aspiram à sua integridade pessoal e ao reconhecimento social, as restantes figuras personificam a incapacidade de relembrar da qual a sociedade civil genericamente parece sofrer. Na visão de Rabinovici, devido a este esquecimento colectivo a sociedade incorre nos mesmos erros do passado, do passado nacional-socialista, na medida em que, adoptando o papel de mera espectadora, continua passivamente a ignorar a necessidade de legitimação e de aceitação de um conjunto de indivíduos.
Die Eigenheit der letzteren Welle von literarischen Texten mit dem Hintergrund der Flucht und Vertreibung – unter anderen Günter Grass 'Im Krebsgang' (2002), Tanja Dückers 'Himmelskörper' (2003), Christoph Heins 'Landnahme' (2004) – liegt nicht in der Thematik per se (seit dem Nachkrieg wurde die Flucht und Vertreibung von Schriftstellern aus verschiedenen ideologischen Hintergründen thematisiert und fiktionalisiert), sondern in der Problematik der transgenerationellen Übertragung dieser traumatischen Erfahrungen. Schwerpunkt dieser Werke ist nicht darzustellen, wie die Kriegsgeneration den Zusammenbruch des Dritten Reichs erlebte, sondern die Überlieferung und die Verarbeitung dieser Ereignisse im Familienkreis, d.h. die Vermittlung der Wirkung solcher Erlebnisse auf die folgenden Generationen (Kinder, Enkelkinder). […] Viele dieser Werke weisen auf die Massenvergewaltigungen (oder zumindest auf die Panik davor) im Kontext von Flucht und Vertreibung hin, widmen aber der sexuellen Gewalt keine besondere Aufmerksamkeit im Text. Ausnahmen sind Hans-Ulrich Treichels (BRD, 1952) 'Der Verlorene' (1998) und Reinhard Jirgls (DDR, 1953) 'Die Unvollendeten' (2003). Für die Mütter der Erzähler dieser Texte waren die deutsche Niederlage und der Heimatverlust eng mit sexuellem Missbrauch verbunden. Als unbewältigte Schatten auf dem Familienleben müssen solche Erfahrungen bei der Analyse der intrafamiliären und intergenerationellen Beziehungen unbedingt berücksichtigt werden. Ziel dieses Referats ist es, zu untersuchen, welche Strategien beide Texte für die Repräsentation der Vergewaltigung bevorzugen, welche Rolle die sexuelle Gewalt als Chiffre der deutschen Niederlage in ihnen spielt, und inwieweit diese traumatische Erfahrung als Trope für die Nachkriegsrealitäten der beiden deutschen Staaten und der Integration der Vertriebenen fungiert.
Vor mehr als zehn Jahren beschrieb ich bereits Theorie und Praxis des Deutschunterrichts an der Universidade Aberta, ein Studienbereich, der 1996 auf Initiative von Maria Laura Bettencourt Pires (Direktorin des Bereichs der Fernstudien do Ensino a Distância) entstanden war. Doch sollte die Umsetzung des Regelwerks von Bologna an unserer Institution noch bis 2007 dauern. Portugal hatte diesen Vorgaben, die einen gemeinsamen europäischen Hochschulraum ermöglichen sollten, bereits 1999 zugestimmt. Sie werden im geänderten Hochschulrahmengesetz von 2005 berücksichtigt. Ihre Einführung an der Universidade Aberta fiel sowohl mit der Methode des E-learnings als auch mit der Einbettung der Germanistik in ein neues Kurssystem zusammen, die u.a. die Semestralisierung der Lehrfächer zur Vorraussetzung hatte. Diese Semestralisierung bedeutete die Verdoppelung der zu unterrichtenden Fächer, sowie ein Vielfaches der bisherigen Anzahl der Examen. Außerdem galt es zu berücksichtigen, dass der Lehrstoff der Literaturfächer (franz. und dt. Literatur) bis zu den Anfängen der jeweiligen nationalen Literaturen ausgedehnt werden sollte. Bis zu jenem Zeitpunkt wurde an der Universidade Aberta das Fach ‚Neuere deutsche Literaturgeschichte vom 18. Jh. (Aufklärung) bis zur Gegenwartsliteratur‘ gelehrt. Für unsere Germanistikabteilung war ich in einer Arbeitsgruppe dafür zuständig, Vorschläge zur Umstrukturierung (inkl. Semestralisierung) und Anpassung der ‚Línguas e Literaturas Modernas‘ (LLM) an das Regelwerk von Bologna auszuarbeiten.
Background: Many disabling human retinal disorders involve the central retina, particularly the macula. However, the commonly used rodent models in research, mouse and rat, do not possess a macula. The purpose of this study was to identify small laboratory rodents with a significant central region as potential new models for macular research.
Methodology/Principal Findings: Gerbillus perpallidus, Meriones unguiculatus and Phodopus campbelli, laboratory rodents less commonly used in retinal research, were subjected to confocal scanning laser ophthalmoscopy (cSLO), fluorescein and indocyanine green angiography, and spectral-domain optical coherence tomography (SD-OCT) using standard equipment (Heidelberg Engineering HRA1 and Spectralis™) adapted to small rodent eyes. The existence of a visual streak-like pattern was assessed on the basis of vascular topography, retinal thickness, and the topography of retinal ganglion cells and cone photoreceptors. All three species examined showed evidence of a significant horizontal streak-like specialization. cSLO angiography and retinal wholemounts revealed that superficial retinal blood vessels typically ramify and narrow into a sparse capillary net at the border of the respective area located dorsal to the optic nerve. Similar to the macular region, there was an absence of larger blood vessels in the streak region. Furthermore, the thickness of the photoreceptor layer and the population density of neurons in the ganglion cell layer were markedly increased in the visual streak region.
Conclusions/Significance: The retinal specializations of Gerbillus perpallidus, Meriones unguiculatus and Phodopus campbelli resemble features of the primate macula. Hence, the rodents reported here may serve to study aspects of macular development and diseases like age-related macular degeneration and diabetic macular edema, and the preclinical assessment of therapeutic strategies.
Activation of Notch1 signaling in neural progenitor cells (NPCs) induces self-renewal and inhibits neurogenesis. Upon neuronal differentiation, NPCs overcome this inhibition, express proneural genes to induce Notch ligands, and activate Notch1 in neighboring NPCs. The molecular mechanism that coordinates Notch1 inactivation with initiation of neurogenesis remains elusive. Here, we provide evidence that Prox1, a transcription repressor and downstream target of proneural genes, counteracts Notch1 signaling via direct suppression of Notch1 gene expression. By expression studies in the developing spinal cord of chick and mouse embryo, we showed that Prox1 is limited to neuronal precursors residing between the Notch1+ NPCs and post-mitotic neurons. Physiological levels of Prox1 in this tissue are sufficient to allow binding at Notch1 promoter and they are critical for proper Notch1 transcriptional regulation in vivo. Gain-of-function studies in the chick neural tube and mouse NPCs suggest that Prox1-mediated suppression of Notch1 relieves its inhibition on neurogenesis and allows NPCs to exit the cell cycle and differentiate. Moreover, loss-of-function in the chick neural tube shows that Prox1 is necessary for suppression of Notch1 outside the ventricular zone, inhibition of active Notch signaling, down-regulation of NPC markers, and completion of neuronal differentiation program. Together these data suggest that Prox1 inhibits Notch1 gene expression to control the balance between NPC self-renewal and neuronal differentiation.
Türkiye’de son yıllarda birçok üniversitede çeşitli dillere yönelik “mütercim-tercümanlık”, daha yeni bir tanımla “çeviribilim” bölümlerinin açılmasıyla birlikte çeviri eğitimi, bilimsel açıdan araştırmaya değer bir konu haline gelmiştir. Sayıları yıldan yıla artarak bugün 20’ye ulaşan çeviri bölümlerinin hedef ve temel ilkeleri, son on yıllarla ifade edilen evreli bir gelişim sürecinde kurumsallaşmaya yaklaşan “çeviribilim” alanı açısından araştırılmaya değer olgusal durum olarak karşımızda durmaktadır. Bu çalışmanın amacı, bir yandan Türkiye’deki üniversitelerin çeviri programlarını temel alarak sürdürülen eğitim-öğretim etkinliklerini, ortaya koydukları hedefler açısından ele alıp irdelemek; diğer yandan ise çeviri eğitimi uygulamaları ile çeviri eğitimine dönük olarak ortaya atılan yaklaşımlar arasındaki benzerlikleri - farklılıkları tartışmak, çeviri bölümlerinin eğitim-öğretim izlenceleri üzerinden çeviri eğitiminde görülen olası eksikliklere ve yetersizliklere ilişkin çözüm önerileri geliştirmektir.
Sucrose- and H+-dependent charge movements associated with the gating of sucrose transporter ZmSUT1

Background: In contrast to man the majority of higher plants use sucrose as mobile carbohydrate. Accordingly proton-driven sucrose transporters are crucial for cell-to-cell and long-distance distribution within the plant body. Generally very negative plant membrane potentials and the ability to accumulate sucrose quantities of more than 1 M document that plants must have evolved transporters with unique structural and functional features.
Methodology/Principal Findings: To unravel the functional properties of one specific high capacity plasma membrane sucrose transporter in detail, we expressed the sucrose/H+ co-transporter from maize ZmSUT1 in Xenopus oocytes. Application of sucrose in an acidic pH environment elicited inward proton currents. Interestingly the sucrose-dependent H+ transport was associated with a decrease in membrane capacitance (Cm). In addition to sucrose Cm was modulated by the membrane potential and external protons. In order to explore the molecular mechanism underlying these Cm changes, presteady-state currents (Ipre) of ZmSUT1 transport were analyzed. Decay of Ipre could be best fitted by double exponentials. When plotted against the voltage the charge Q, associated to Ipre, was dependent on sucrose and protons. The mathematical derivative of the charge Q versus voltage was well in line with the observed Cm changes. Based on these parameters a turnover rate of 500 molecules sucrose/s was calculated. In contrast to gating currents of voltage dependent-potassium channels the analysis of ZmSUT1-derived presteady-state currents in the absence of sucrose (I = Q/τ) was sufficient to predict ZmSUT1 transport-associated currents.
Conclusions: Taken together our results indicate that in the absence of sucrose, ‘trapped’ protons move back and forth between an outer and an inner site within the transmembrane domains of ZmSUT1. This movement of protons in the electric field of the membrane gives rise to the presteady-state currents and in turn to Cm changes. Upon application of external sucrose, protons can pass the membrane turning presteady-state into transport currents.
Background: Occupational demands of educators are not very well researched. Nevertheless their work is subject to several requirements. Whether these demands have an effect on the work ability and the health status of employees has also not been examined. Furthermore it is unclear if the ownership type of day care centres have an influence on job satisfaction and work ability of the pedagogical staff and what kind of resources do exist. Previous studies were mainly based on questionnaire data. Objective data does not exist. Therefore the aim of this investigation is to collect precise data relating to work of educators.
Methods: Effects of different types of ownership of day care centres on job satisfaction and work ability of educators will be assessed with the help of objective real time studies in combination with multi-level psycho diagnostic measurements.
Discussion: The present study is the first of its kind. Up to now there are no computer-based real time studies on workflow of pedagogical staff with regard to assess their work-related stress. Following an exhaustive documentation of educators work processes the day-to-day task can be estimated and approaches for prevention can be developed. This can substantially contribute to an overall improvement of child care in Germany.
Background: Due to constantly rising air pollution levels as well as an increasing awareness of the hazardousness of air pollutants, new laws and rules have recently been passed. Although there has been a large amount of research on this topic, bibliometric data is still to be collected. Thus this study provides a scientometric approach to the material published on this subject so far.
Methods: For this purpose, data retrieved from the "Web of Science" provided by the Thomson Scientific Institute was analyzed and visualized both with density-equalizing methods and classic data-processing methods such as tables and charts.
Results: For the time span between 1955 and 2006, 26,253 items were listed and related to the topic of air pollution, published by 124 countries in 24 different languages. General citation activity has been constantly increasing since the beginning of the examined period. However, beginning with the year 1991, citation levels have been rising exponentially each year, reaching 39,220 citations in the year 2006. The United States, the UK and Germany were the three most productive countries in the area, with English and German ranked first and second in publishing languages, followed by French. An article published by Dockery, Pope, Xu et al. was both the most cited in total numbers and in average citation rate. J. Schwartz was able to claim the highest total number of citations on his publications, while D.W. Dockery has the highest citation rate per publication. As to the subject areas the items are assigned with, the most item were published in Environmental Sciences, followed by Meteorology & Atmospheric Sciences and Public, Environmental & Occupational Health. Nine out of the ten publishing journals with more than 300 entries dealt with environmental interests and one dealt with epidemiology.
Conclusions: Using the method of density-equalizing mapping and further common data processing procedures, it can be concluded that scientific work concerning air pollution and related topics enjoys unbrokenly growing scientific interest. This can be observed both in publication numbers and in citation activity.
Background: Leishmaniasis is a chronic disease that is found in various countries of the world. The aim of the current study was to investigate the impact of leishmaniasis on the world's research output. The present study assessed benchmarking of research output for the period between 1957 and 2006. Using large database analyses, research in the field of leishmaniasis was evaluated. Furthermore, cooperation between different countries was identified.
Results: The number of publications increased with time. Most publications came from Western countries such as the US, UK or Germany. Interestingly, countries like Brazil and India had a high research output. We found a substantial amount of cooperation between countries.
Conclusion: Although leishmaniasis is of limited geographic distribution it attracts a wide research interest. The central hub of research cooperation is the USA.
Soil biogenic NO emissions (SNOx) play important direct and indirect roles in chemical processes of the troposphere. The most widely applied algorithm to calculate SNOx in global models was published 15 years ago by Yienger and Levy (1995), was based on very few measurements. Since then numerous new measurements have been published, which we used to build up a atabase of field measurements conducted world wide covering the period from 1978 to 2009, including 108 publications with 560 measurements.
Recently, several satellite based top-down approaches, which recalculated the different sources of NOx (fossil fuel, biomass burning, soil and lightning), have shown an underestimation of SNOx by the algorithm of Yienger and Levy (1995). Nevertheless, to our knowledge no general improvements of this algorithm have yet been published.
Here we present major improvements to the algorithm, which should help to optimize the representation of SNOx in atmospheric-chemistry global climate models, without modifying the underlying principal or mathematical equations. The changes include: 1) Using a new up to date land cover map, with twice the number of land cover classes, and using annually varying fertilizer application rates; 2) Adopting the fraction of SNOx induced by fertilizer application based on our database; 3) Switching from soil water column to volumetric soil moisture, to distinguish between the wet and dry state; 4) Tuning the emission factors to reproduce the measured emissions in our database and calculate the emissions based on their mean value. These steps lead us to increased global yearly SNOx, and our total SNOx source ends up being close to one of the top-down approaches. In some geographical regions the new results agree better with the top-down approach, but there are also distinct differences in other regions. This suggests that a ombination of both top-down and bottom-up approaches could be combined in a future attempt to provide an even better calculation of SNOx.
Residual circulation trajectories and transit times into the extratropical lowermost stratosphere

Transport into the extratropical lowermost stratosphere (LMS) can be divided into a slow part (time-scale of several months to years) associated with the global-scale stratospheric residual circulation and a fast part (time-scale of days to a few months) associated with (mostly quasi-horizontal) mixing (i.e. two-way irreversible transport, including stratosphere-troposphere exchange). The stratospheric residual circulation can be considered to consist of two branches: a deep branch more strongly associated with planetary waves breaking in the middle to upper stratosphere, and a shallow branch more strongly associated with synoptic-scale waves breaking in the subtropical lower stratosphere. In this study the contribution due to the stratospheric residual circulation alone to transport into the LMS is quantified using residual circulation trajectories, i.e. trajectories driven by the (time-dependent) residual mean meridional and vertical velocities. This contribution represents the advective part of the overall transport into the LMS and can be viewed as providing a background onto which the effect of mixing has to be added. Residual mean velocities are obtained from a comprehensive chemistry-climate model as well as from reanalysis data. Transit times of air traveling from the tropical tropopause to the LMS along the residual circulation streamfunction are evaluated and compared to recent mean age of air estimates. A clear time-scale separation with much smaller transit times into the mid-latitudinal LMS than into polar LMS is found that is indicative of a clear separation of the shallow from the deep branch of the residual circulation. This separation between the shallow and the deep circulation branch is further manifested in a clear distinction in the aspect ratio of the vertical to meridional extent of the trajectories as well as the integrated mass flux along the residual circulation trajectories. The residual transit time distribution reproduces qualitatively the observed seasonal cycle of youngest air in the extratropical LMS in fall and oldest air in spring.
Tubular carbonate concretions of up to 1 m in length and perpendicular to bedding, occur abundantly in the Upper Pliensbachian (upper Amaltheus margaritatus Zone, Gibbosus Subzone) in outcrops (Fontaneilles section) in the vicinity of Rivière-sûr-Tarn, southern France. Stable isotope analyses of these concretions show negative δ13C values that decrease from the rim to the center from −18.8‰ to −25.7‰ (V-PDB), but normal marine δ18O values (−1.8‰). Carbon isotope analyses of Late Pliensbachian bulk carbonate (matrix) samples from the Fontaneilles section show clearly decreasing C-isotope values across the A. margaritatus Zone, from +1‰ to −3‰ (V-PDB). Isotope analyses of coeval belemnite rostra do not document such a negative C-isotope trend with values remaining stable around +2‰ (V-PDB). Computer tomographic (CT) scanning of the tubular concretions show multiple canals that are lined or filled entirely with pyrite. Previously, the formation of these concretions with one, two, or more central tubes, has been ascribed to the activity of an enigmatic organism, possibly with annelid or arthropod affinities, known as Tisoa siphonalis. Our results suggest tisoan structures are abiogenic. Based on our geochemical analyses and sedimentological observations we suggest that these concretions formed as a combination of the anaerobic oxidation of methane (AOM) and sulfate reduction within the sediment. Fluids rich in methane and/or hydrocarbons likely altered local bulk rock carbon isotope records, but did not affect the global carbon cycle. Interestingly, Tisoa siphonalis has been described from many locations in the Grands Causses Basin in southern France, and from northern France and Luxemburg, always occurring at the same stratigraphic level. Upper Pliensbachian authigenic carbonates thus possibly cover an area of many thousand square kilometers. Greatly reduced sedimentation rates are needed to explain the stabilization of the sulfate-methane transition zone in the sedimentary column in order for the tubular concretions to form. Late Pliensbachian cooling, reducing run-off, and/or the influx of colder water and more vigorous circulation could be responsible for a halt in sedimentation. At the same time (thermogenic) methane may have destabilized during a major phase of Late Pliensbachian sea level fall. As such Tisoa siphonalis is more than a geological curiosity, and its further study could prove pivotal in understanding Early Jurassic paleoenvironmental change.
Floodplains play an important role in the terrestrial water cycle and are very important for biodiversity. Therefore, an improved representation of the dynamics of floodplain water flows and storage in global hydrological and land surface models is required. To support model validation, we combined monthly time series of satellite-derived inundation areas (Papa et al., 2010) with data on irrigated rice areas (Portmann et al., 2010). In this way, we obtained global-scale time series of naturally inundated areas (NIA), with monthly values of inundation extent during 1993–2004 and a spatial resolution of 0.5°. For most grid cells (0.5°×0.5°), the mean annual maximum of NIA agrees well with the static open water extent of the Global Lakes and Wetlands database (GLWD) (Lehner and Döll, 2004), but in 16% of the cells NIA is larger than GLWD. In some regions, like Northwestern Europe, NIA clearly overestimates inundated areas, probably because of confounding very wet soils with inundated areas. In other areas, such as South Asia, it is likely that NIA can help to enhance GLWD. NIA data will be very useful for developing and validating a floodplain modeling algorithm for the global hydrological model WGHM. For example, we found that monthly NIAs correlate with observed river discharges.
Introduction: Researchers experience increasing pressures to connect with bodies that finance their projects. In this climate, critical scholars face many obstacles as they seek to navigate the treacherous waters of securing external funds. To debate these challenges, the ACME Editorial Collective organized a panel for the 2009 Annual Meeting of the Association of American Geographers in Las Vegas. This intervention represents a follow-up discussion and collective writing process among some of the panelists and members of the audience who attended the panel.
Below, we examine the neoliberalization of the current funding systems, discuss the implications for research practice, and make suggestions for critical engagement and transformation. Our suggestions, however, will not be easy to implement, as we can infer from the experience of the radical scholars of the post-1968 generation whose ascension into the upper echelons of North American and European university systems was also associated with the neoliberalization of the funding systems. This intervention represents a modest contribution in the tradition of critical research practice of creating the possibilities for progressive change.

Augustins frühe Schrift De musica ist neben Boethius’ De institutione musica und weiteren Äußerungen Augustins über Musik in anderen Schriften eine der zentralen Quellen für die Frühzeit der christlich-abendländischen Musikanschauung und hat diese bis in die Neuzeit hinein geprägt. Die sechs Bücher dieses Werkes haben in der Rezeption allerdings unterschiedliche Beachtung gefunden, da das sechste mehr christlich-philosophisch ausgerichtete Buch sich in Stil und Inhalt von den anderen fünf eher technisch ausgerichteten Büchern deutlich unterscheidet. Dies hat in der Forschung zu unterschiedlichen Spekulationen über die literarische und inhaltliche Einheitlichkeit von De musica geführt, zumal sich eine Überarbeitung des sechsten Buches tatsächlich nachweisen lässt. Es hat auch dazu geführt, dass in Untersuchungen zu dieser Schrift oft nur vom sechsten Buch ausgegangen und von den ersten fünf Büchern abgesehen wurde. Auch der in der folgenden Darstellung akzentuierte – und bisher kaum beachtete – Aspekt wird hauptsächlich im sechsten Buch greifbar. Dennoch wird als Neuansatz versucht, diesen in die als einheitlich zu erweisende Gesamtkonzeption aller sechs Bücher einzubetten. Dieser Aspekt betrifft die ethische Dimension der Schrift. Sie, so die Grundthese, stellt vom ersten Buch an einen zentralen Strang des Werkes dar. ...

Tracer measurements in the tropical tropopause layer during the AMMA/SCOUT-O3 aircraft campaign

We present airborne in situ measurements made during the AMMA (African Monsoon Multidisciplinary Analysis)/SCOUT-O3 campaign between 31 July and 17 August 2006 on board the M55 Geophysica aircraft, based in Ouagadougou, Burkina Faso. CO<sub>2</sub> and N<sub>2</sub>O were measured with the High Altitude Gas Analyzer (HAGAR), CO was measured with the Cryogenically Operated Laser Diode (COLD) instrument, and O<sub>3</sub> with the Fast Ozone ANalyzer (FOZAN). We analyze the data obtained during five local flights to study the dominant transport processes controlling the tropical tropopause layer (TTL) above West-Africa: deep convection up to the level of main convective outflow, overshooting of deep convection, horizontal inmixing across the subtropical tropopause, and horizontal transport across the subtropical barrier. Except for the flight of 13 August, distinct minima in CO<sub>2</sub> indicate convective outflow of boundary layer air in the TTL. The CO<sub>2</sub> profiles show that the level of main convective outflow was mostly located between 350 and 360 K, and for 11 August reached up to 370 K. While the CO<sub>2</sub> minima indicate quite significant convective influence, the O<sub>3</sub> profiles suggest that the observed convective signatures were mostly not fresh, but of older origin. When compared with the mean O<sub>3</sub> profile measured during a previous campaign over Darwin in November 2005, the O<sub>3</sub> minimum at the main convective outflow level was less pronounced over Ouagadougou. Furthermore O<sub>3</sub> mixing ratios were much higher throughout the whole TTL and, unlike over Darwin, rarely showed low values observed in the regional boundary layer. Signatures of irreversible mixing following overshooting of convective air were scarce in the tracer data. Some small signatures indicative of this process were found in CO<sub>2</sub> profiles between 390 and 410 K during the flights of 4 and 8 August, and in CO data at 410 K on 7 August. However, the absence of expected corresponding signatures in other tracer data makes this evidence inconclusive, and overall there is little indication from the observations that overshooting convection has a profound impact on TTL composition during AMMA. We find the amount of photochemically aged air isentropically mixed into the TTL across the subtropical tropopause to be not significant. Using the N<sub>2</sub>O observations we estimate the fraction of aged extratropical stratospheric air in the TTL to be 0.0±0.1 up to 370 K during the local flights, increasing above this level to 0.2±0.15 at 390 K. The subtropical barrier, as indicated by the slope of the correlation between N<sub>2</sub>O and O<sub>3</sub> between 415 and 490 K, does not appear as a sharp border between the tropics and extratropics, but rather as a gradual transition region between 10 and 25° N latitude where isentropic mixing between these two regions may occur.
Background: Circulating progenitor cells (CPC) contribute to the homeostasis of the vessel wall, and a reduced CPC count predicts cardiovascular morbidity and mortality. We tested the hypothesis that CPC count improves cardiovascular risk stratification and that this is modulated by low-grade inflammation. Methodology/Principal Findings: We pooled data from 4 longitudinal studies, including a total of 1,057 patients having CPC determined and major adverse cardiovascular events (MACE) collected. We recorded cardiovascular risk factors and high-sensitive C-reactive protein (hsCRP) level. Risk estimates were derived from Cox proportional hazard analyses. CPC count and/or hsCRP level were added to a reference model including age, sex, cardiovascular risk factors, prevalent CVD, chronic renal failure (CRF) and medications. The sample was composed of high-risk individuals, as 76.3% had prevalent CVD and 31.6% had CRF. There were 331 (31.3%) incident MACE during an average 1.7±1.1 year follow-up time. CPC count was independently associated with incident MACE even after correction for hsCRP. According to C-statistics, models including CPC yielded a non-significant improvement in accuracy of MACE prediction. However, the integrated discrimination improvement index (IDI) showed better performance of models including CPC compared to the reference model and models including hsCRP in identifying MACE. CPC count also yielded significant net reclassification improvements (NRI) for CV death, non-fatal AMI and other CV events. The effect of CPC was independent of hsCRP, but there was a significant more-than-additive interaction between low CPC count and raised hsCRP level in predicting incident MACE. Conclusions/Significance: In high risk individuals, a reduced CPC count helps identifying more patients at higher risk of MACE over the short term, especially in combination with a raised hsCRP level.
Although a variety of genetic strategies have been developed to inhibit HIV replication, few direct comparisons of the efficacy of these inhibitors have been carried out. Moreover, most studies have not examined whether genetic inhibitors are able to induce a survival advantage that results in an expansion of genetically-modified cells following HIV infection. We evaluated the efficacy of three leading genetic strategies to inhibit HIV replication: 1) an HIV-1 tat/rev-specific small hairpin (sh) RNA; 2) an RNA antisense gene specific for the HIV-1 envelope; and 3) a viral entry inhibitor, maC46. In stably transduced cell lines selected such that >95% of cells expressed the genetic inhibitor, the RNA antisense envelope and viral entry inhibitor maC46 provided the strongest inhibition of HIV-1 replication. However, when mixed populations of transduced and untransduced cells were challenged with HIV-1, the maC46 fusion inhibitor resulted in highly efficient positive selection of transduced cells, an effect that was evident even in mixed populations containing as few as 1% maC46-expressing cells. The selective advantage of the maC46 fusion inhibitor was also observed in HIV-1-infected cultures of primary T lymphocytes as well as in HIV-1-infected humanized mice. These results demonstrate robust inhibition of HIV replication with the fusion inhibitor maC46 and the antisense Env inhibitor, and importantly, a survival advantage of cells expressing the maC46 fusion inhibitor both in vitro and in vivo. Evaluation of the ability of genetic inhibitors of HIV-1 replication to confer a survival advantage on genetically-modified cells provides unique information not provided by standard techniques that may be important in the in vivo efficacy of these genes.
Background: ClC-7 is a ubiquitous transporter which is broadly expressed in mammalian tissues. It is implied in the pathogenesis of lysosomal storage disease and osteopetrosis. Because of its endosomal/lysosomal localization it is still poorly characterized. Methodology/Principal Findings: An electrophysiological characterization of rat ClC-7 using solid-supported membrane-based electrophysiology is presented. The measured currents show the characteristics of ClC-7 and confirm its function as a Cl−/H+-antiporter. We have used rat ClC-7 in CHO cells as a model system to investigate the functionality and cellular localization of the wt transporter and its variant G213R ClC-7 which is the analogue of human G215R ClC-7 responsible for autosomal dominant osteopetrosis type II. Our study shows that rat G213R ClC-7 is functional but has a localization defect in CHO cells which prevents it from being correctly targeted to the lysosomal membrane. The electrophysiological assay is tested as a tool for drug discovery. The assay is validated with a number of drug candidates. It is shown that ClC-7 is inhibited by DIDS, NPPB and NS5818 at micromolar concentrations. Conclusions/Significance: It is suggested that the scenario found in the CHO model system also applies to the human transporter and that mislocalization rather than impaired functionality of G215R ClC-7 is the primary cause of the related autosomal dominant osteopetrosis type II. Furthermore, the robust solid-supported membrane-based electrophysiological assay is proposed for rapid screening for potential ClC-7 inhibitors which are discussed for treatment of osteoporosis.
We established a protocol of the SuperSAGE technology combined with next-generation sequencing, coined “High-Throughput (HT-) SuperSAGE”. SuperSAGE is a method of digital gene expression profiling that allows isolation of 26-bp tag fragments from expressed transcripts. In the present protocol, index (barcode) sequences are employed to discriminate tags from different samples. Such barcodes allow researchers to analyze digital tags from transcriptomes of many samples in a single sequencing run by simply pooling the libraries. Here, we demonstrated that HT-SuperSAGE provided highly sensitive, reproducible and accurate digital gene expression data. By increasing throughput for analysis in HT-SuperSAGE, various applications are foreseen and several examples are provided in the present study, including analyses of laser-microdissected cells, biological replicates and tag extraction using different anchoring enzymes.
Background: One virulence property of Borrelia burgdorferi is its resistance to innate immunity, in particular to complement-mediated killing. Serum-resistant B. burgdorferi express up to five distinct complement regulator-acquiring surface proteins (CRASP) which interact with complement regulator factor H (CFH) and factor H-like protein 1 (FHL1) or factor H-related protein 1 (CFHR1). In the present study we elucidate the role of the infection-associated CRASP-3 and CRASP-5 protein to serve as ligands for additional complement regulatory proteins as well as for complement resistance of B. burgdorferi. Methodology/Principal Findings: To elucidate whether CRASP-5 and CRASP-3 interact with various human proteins, both borrelial proteins were immobilized on magnetic beads. Following incubation with human serum, bound proteins were eluted and separated by Glycine-SDS-PAGE. In addition to CFH and CFHR1, complement regulators CFHR2 and CFHR5 were identified as novel ligands for both borrelial proteins by employing MALDI-TOF. To further assess the contributions of CRASP-3 and CRASP-5 to complement resistance, a serum-sensitive B. garinii strain G1 which lacks all CFH-binding proteins was used as a valuable model for functional analyses. Both CRASPs expressed on the B. garinii outer surface bound CFH as well as CFHR1 and CFHR2 in ELISA. In contrast, live B. garinii bound CFHR1, CFHR2, and CFHR5 and only miniscute amounts of CFH as demonstrated by serum adsorption assays and FACS analyses. Further functional analysis revealed that upon NHS incubation, CRASP-3 or CRASP-5 expressing borreliae were killed by complement. Conclusions/Significance: In the absence of CFH and the presence of CFHR1, CFHR2 and CFHR5, assembly and integration of the membrane attack complex was not efficiently inhibited indicating that CFH in co-operation with CFHR1, CFHR2 and CFHR5 supports complement evasion of B. burgdorferi.
Quantification of circulating endothelial progenitor cells using the modified ISHAGE protocol

Aims: Circulating endothelial progenitor cells (EPC), involved in endothelial regeneration, neovascularisation, and determination of prognosis in cardiovascular disease can be characterised with functional assays or using immunofluorescence and flow cytometry. Combinations of markers, including CD34+KDR+ or CD133+KDR+, are used. This approach, however may not consider all characteristics of EPC. The lack of a standardised protocol with regards to reagents and gating strategies may account for the widespread inter-laboratory variations in quantification of EPC. We, therefore developed a novel protocol adapted from the standardised so-called ISHAGE protocol for enumeration of haematopoietic stem cells to enable comparison of clinical and laboratory data. Methods and Results: In 25 control subjects, 65 patients with coronary artery disease (CAD; 40 stable CAD, 25 acute coronary syndrome/acute myocardial infarction (ACS)), EPC were quantified using the following approach: Whole blood was incubated with CD45, KDR, and CD34. The ISHAGE sequential strategy was used, and finally, CD45dimCD34+ cells were quantified for KDR. A minimum of 100 CD34+ events were collected. For comparison, CD45+CD34+ and CD45-CD34+ were analysed simultaneously. The number of CD45dimCD34+KDR+ cells only were significantly higher in healthy controls compared to patients with CAD or ACS (p = 0.005 each, p<0.001 for trend). An inverse correlation of CD45dimCD34+KDR+ with disease activity (r = -0.475, p<0.001) was confirmed. Only CD45dimCD34+KDR+ correlated inversely with the number of diseased coronaries (r = -0.344; p<0.005). In a second study, a 4-week de-novo treatment of atorvastatin in stable CAD evoked an increase only of CD45dimCD34+KDR+ EPC (p<0.05). CD45+CD34+KDR+ and CD45-CD34+KDR+ were indifferent between the three groups. Conclusion: Our newly established protocol adopted from the standardised ISHAGE protocol achieved higher accuracy in EPC enumeration confirming previous findings with respect to the correlation of EPC with disease activity and the increase of EPC during statin therapy. The data of this study show the CD45dim fraction to harbour EPC.
Background: Clock genes and their protein products regulate circadian rhythms in mammals but have also been implicated in various physiological processes, including bone formation. Osteoblasts build new mineralized bone whereas osteoclasts degrade it thereby balancing bone formation. To evaluate the contribution of clock components in this process, we investigated mice mutant in clock genes for a bone volume phenotype. Methodology/Principal Findings: We found that Per2Brdm1 mutant mice as well as mice lacking Cry2-/- displayed significantly increased bone volume at 12 weeks of age, when bone turnover is high. Per2Brdm1 mutant mice showed alterations in parameters specific for osteoblasts whereas mice lacking Cry2-/- displayed changes in osteoclast specific parameters. Interestingly, inactivation of both Per2 and Cry2 genes leads to normal bone volume as observed in wild type animals. Importantly, osteoclast parameters affected due to the lack of Cry2, remained at the level seen in the Cry2-/- mutants despite the simultaneous inactivation of Per2. Conclusions/Significance: This indicates that Cry2 and Per2 affect distinct pathways in the regulation of bone volume with Cry2 influencing mostly the osteoclastic cellular component of bone and Per2 acting on osteoblast parameters.
Background: An inducible release of soluble junctional adhesion molecule-A (sJAM-A) under pro-inflammatory conditions was described in cultured non-CNS endothelial cells (EC) and increased sJAM-A serum levels were found to indicate inflammation in non-CNS vascular beds. Here we studied the regulation of JAM-A expression in cultured brain EC and evaluated sJAM-A as a serum biomarker of blood-brain barrier (BBB) function. Methodology/Principal Findings: As previously reported in non-CNS EC types, pro-inflammatory stimulation of primary or immortalized (hCMEC/D3) human brain microvascular EC (HBMEC) induced a redistribution of cell-bound JAM-A on the cell surface away from tight junctions, along with a dissociation from the cytoskeleton. This was paralleled by reduced immunocytochemical staining of occludin and zonula occludens-1 as well as by increased paracellular permeability for dextran 3000. Both a self-developed ELISA test and Western blot analysis detected a constitutive sJAM-A release by HBMEC into culture supernatants, which importantly was unaffected by pro-inflammatory or hypoxia/reoxygenation challenge. Accordingly, serum levels of sJAM-A were unaltered in 14 patients with clinically active multiple sclerosis compared to 45 stable patients and remained unchanged in 13 patients with acute ischemic non-small vessel stroke over time. Conclusion: Soluble JAM-A was not suited as a biomarker of BBB breakdown in our hands. The unexpected non-inducibility of sJAM-A release at the human BBB might contribute to a particular resistance of brain EC to inflammatory stimuli, protecting the CNS compartment.
Respiratory chain complexes in dynamic mitochondria display a patchy distribution in life cells

Background: Mitochondria, the main suppliers of cellular energy, are dynamic organelles that fuse and divide frequently. Constraining these processes impairs mitochondrial is closely linked to certain neurodegenerative diseases. It is proposed that functional mitochondrial dynamics allows the exchange of compounds thereby providing a rescue mechanism. Methodology/Principal Findings: The question discussed in this paper is whether fusion and fission of mitochondria in different cell lines result in re-localization of respiratory chain (RC) complexes and of the ATP synthase. This was addressed by fusing cells containing mitochondria with respiratory complexes labelled with different fluorescent proteins and resolving their time dependent re-localization in living cells. We found a complete reshuffling of RC complexes throughout the entire chondriome in single HeLa cells within 2–3 h by organelle fusion and fission. Polykaryons of fused cells completely re-mixed their RC complexes in 10–24 h in a progressive way. In contrast to the recently described homogeneous mixing of matrix-targeted proteins or outer membrane proteins, the distribution of RC complexes and ATP synthase in fused hybrid mitochondria, however, was not homogeneous but patterned. Thus, complete equilibration of respiratory chain complexes as integral inner mitochondrial membrane complexes is a slow process compared with matrix proteins probably limited by complete fusion. In co-expressing cells, complex II is more homogenously distributed than complex I and V, resp. Indeed, this result argues for higher mobility and less integration in supercomplexes. Conclusion/Significance: Our results clearly demonstrate that mitochondrial fusion and fission dynamics favours the re-mixing of all RC complexes within the chondriome. This permanent mixing avoids a static situation with a fixed composition of RC complexes per mitochondrion.
This paper discusses the sustainability impact (contribution to sustainability, reduction of adverse environmental impacts) of online second-hand trading. A survey of eBay users shows that a relationship between the trading of used goods and the protection of natural resources is hardly realized. Secondly, the environmental motivation and the willingness to act in a sustainable manner differ widely between groups of consumers. Given these results from a user perspective, the paper tries to find some objective hints of online second-hand trading’s environmental impact. The greenhouse gas emissions resulting from the energy used for the trading transactions seem to be considerably lower than the emissions due to the (avoided) production of new goods. The paper concludes with a set of recommendations for second-hand trade and consumer policy. Information about the sustainability benefits of purchasing second-hand goods should be included in general consumer information, and arguments for changes in behavior should be targeted to different groups of consumers. Background: The apoptosis-inducing serine protease granzyme B (GrB) is an important factor contributing to lysis of target cells by cytotoxic lymphocytes. Expression of enzymatically active GrB in recombinant form is a prerequisite for functional analysis and application of GrB for therapeutic purposes. Methods and Findings: We investigated the influence of bacterial maltose-binding protein (MBP) fused to GrB via a synthetic furin recognition motif on the expression of the MBP fusion protein also containing an N-terminal alpha-factor signal peptide in the yeast Pichia pastoris. MBP markedly enhanced the amount of GrB secreted into culture supernatant, which was not the case when GrB was fused to GST. MBP-GrB fusion protein was cleaved during secretion by an endogenous furin-like proteolytic activity in vivo, liberating enzymatically active GrB without the need of subsequent in vitro processing. Similar results were obtained upon expression of a recombinant fragment of the ErbB2/HER2 receptor protein or GST as MBP fusions. Conclusions: Our results demonstrate that combination of MBP as a solubility enhancer with specific in vivo cleavage augments secretion of processed and functionally active proteins from yeast. This strategy may be generally applicable to improve folding and increase yields of recombinant proteins.
Background: Decoding of frequency-modulated (FM) sounds is essential for phoneme identification. This study investigates selectivity to FM direction in the human auditory system. Methodology/Principal Findings: Magnetoencephalography was recorded in 10 adults during a two-tone adaptation paradigm with a 200-ms interstimulus-interval. Stimuli were pairs of either same or different frequency modulation direction. To control that FM repetition effects cannot be accounted for by their on- and offset properties, we additionally assessed responses to pairs of unmodulated tones with either same or different frequency composition. For the FM sweeps, N1m event-related magnetic field components were found at 103 and 130 ms after onset of the first (S1) and second stimulus (S2), respectively. This was followed by a sustained component starting at about 200 ms after S2. The sustained response was significantly stronger for stimulation with the same compared to different FM direction. This effect was not observed for the non-modulated control stimuli. Conclusions/Significance: Low-level processing of FM sounds was characterized by repetition enhancement to stimulus pairs with same versus different FM directions. This effect was FM-specific; it did not occur for unmodulated tones. The present findings may reflect specific interactions between frequency separation and temporal distance in the processing of consecutive FM sweeps.
Variants resistant to compounds specifically targeting HCV are observed in clinical trials. A multi-variant viral dynamic model was developed to quantify the evolution and in vivo fitness of variants in subjects dosed with monotherapy of an HCV protease inhibitor, telaprevir. Variant fitness was estimated using a model in which variants were selected by competition for shared limited replication space. Fitness was represented in the absence of telaprevir by different variant production rate constants and in the presence of telaprevir by additional antiviral blockage by telaprevir. Model parameters, including rate constants for viral production, clearance, and effective telaprevir concentration, were estimated from 1) plasma HCV RNA levels of subjects before, during, and after dosing, 2) post-dosing prevalence of plasma variants from subjects, and 3) sensitivity of variants to telaprevir in the HCV replicon. The model provided a good fit to plasma HCV RNA levels observed both during and after telaprevir dosing, as well as to variant prevalence observed after telaprevir dosing. After an initial sharp decline in HCV RNA levels during dosing with telaprevir, HCV RNA levels increased in some subjects. The model predicted this increase to be caused by pre-existing variants with sufficient fitness to expand once available replication space increased due to rapid clearance of wild-type (WT) virus. The average replicative fitness estimates in the absence of telaprevir ranged from 1% to 68% of WT fitness. Compared to the relative fitness method, the in vivo estimates from the viral dynamic model corresponded more closely to in vitro replicon data, as well as to qualitative behaviors observed in both on-dosing and long-term post-dosing clinical data. The modeling fitness estimates were robust in sensitivity analyses in which the restoration dynamics of replication space and assumptions of HCV mutation rates were varied.
The continuous progress in the structural and functional characterization of aquaporins increasingly attracts attention to study their roles in certain mammalian diseases. Although several structures of aquaporins have already been solved by crystallization, the challenge of producing sufficient amounts of functional proteins still remains. CF (cell free) expression has emerged in recent times as a promising alternative option in order to synthesize large quantities of membrane proteins, and the focus of this report was to evaluate the potential of this technique for the production of eukaryotic aquaporins. We have selected the mouse aquaporin 4 as a representative of mammalian aquaporins. The protein was synthesized in an E. coli extract based cell-free system with two different expression modes, and the efficiencies of two modes were compared. In both, the P-CF (cell-free membrane protein expression as precipitate) mode generating initial aquaporin precipitates as well as in the D-CF (cell-free membrane protein expression in presence of detergent) mode, generating directly detergent solubilized samples, we were able to obtain mg amounts of protein per ml of cell-free reaction. Purified aquaporin samples solubilized in different detergents were reconstituted into liposomes, and analyzed for the water channel activity. The calculated Pf value of proteoliposome samples isolated from the D-CF mode was 133 µm/s at 10°C, which was 5 times higher as that of the control. A reversible inhibitory effect of mercury chloride was observed, which is consistent with previous observations of in vitro reconstituted aquaporin 4. In this study, a fast and convenient protocol was established for functional expression of aquaporins, which could serve as basis for further applications such as water filtration.
Background: A delta and C fibers are the major pain-conducting nerve fibers, activate only partly the same brain areas, and are differently involved in pain syndromes. Whether a stimulus excites predominantly A delta or C fibers is a commonly asked question in basic pain research but a quick test was lacking so far. Methodology/Principal Findings: Of 77 verbal descriptors of pain sensations, "pricking", "dull" and "pressing" distinguished best (95% cases correctly) between A delta fiber mediated (punctate pressure produced by means of von Frey hairs) and C fiber mediated (blunt pressure) pain, applied to healthy volunteers in experiment 1. The sensation was assigned to A delta fibers when "pricking" but neither "dull" nor "pressing" were chosen, and to C fibers when the sum of the selections of "dull" or "pressing" was greater than that of the selection of "pricking". In experiment 2, with an independent cohort, the three-descriptor questionnaire achieved sensitivity and specificity above 0.95 for distinguishing fiber preferential non-mechanical induced pain (laser heat, exciting A delta fibers, and 5-Hz electric stimulation, exciting C fibers). Conclusion: A three-item verbal rating test using the words "pricking", "dull", and "pressing" may provide sufficient information to characterize a pain sensation evoked by a physical stimulus as transmitted via A delta or via C fibers. It meets the criteria of a screening test by being easy to administer, taking little time, being comfortable in handling, and inexpensive while providing high specificity for relevant information.
Activation of hypoxia inducible factor 1 is a general phenomenon in infections with human pathogens

Background: Hypoxia inducible factor (HIF)-1 is the key transcriptional factor involved in the adaptation process of cells and organisms to hypoxia. Recent findings suggest that HIF-1 plays also a crucial role in inflammatory and infectious diseases. Methodology/Principal Findings: Using patient skin biopsies, cell culture and murine infection models, HIF-1 activation was determined by immunohistochemistry, immunoblotting and reporter gene assays and was linked to cellular oxygen consumption. The course of a S. aureus peritonitis was determined upon pharmacological HIF-1 inhibition. Activation of HIF-1 was detectable (i) in all ex vivo in biopsies of patients suffering from skin infections, (ii) in vitro using cell culture infection models and (iii) in vivo using murine intravenous and peritoneal S. aureus infection models. HIF-1 activation by human pathogens was induced by oxygen-dependent mechanisms. Small colony variants (SCVs) of S. aureus known to cause chronic infections did not result in cellular hypoxia nor in HIF-1 activation. Pharmaceutical inhibition of HIF-1 activation resulted in increased survival rates of mice suffering from a S. aureus peritonitis. Conclusions/Significance: Activation of HIF-1 is a general phenomenon in infections with human pathogenic bacteria, viruses, fungi and protozoa. HIF-1-regulated pathways might be an attractive target to modulate the course of life-threatening infections.
Background: R-flurbiprofen, one of the enantiomers of flurbiprofen racemate, is inactive with respect to cyclooxygenase inhibition, but shows analgesic properties without relevant toxicity. Its mode of action is still unclear. Methodology/Principal Findings: We show that R-flurbiprofen reduces glutamate release in the dorsal horn of the spinal cord evoked by sciatic nerve injury and thereby alleviates pain in sciatic nerve injury models of neuropathic pain in rats and mice. This is mediated by restoring the balance of endocannabinoids (eCB), which is disturbed following peripheral nerve injury in the DRGs, spinal cord and forebrain. The imbalance results from transcriptional adaptations of fatty acid amide hydrolase (FAAH) and NAPE-phospholipase D, i.e. the major enzymes involved in anandamide metabolism and synthesis, respectively. R-flurbiprofen inhibits FAAH activity and normalizes NAPE-PLD expression. As a consequence, R-Flurbiprofen improves endogenous cannabinoid mediated effects, indicated by the reduction of glutamate release, increased activity of the anti-inflammatory transcription factor PPAR gamma and attenuation of microglia activation. Antinociceptive effects are lost by combined inhibition of CB1 and CB2 receptors and partially abolished in CB1 receptor deficient mice. R-flurbiprofen does however not cause changes of core body temperature which is a typical indicator of central effects of cannabinoid-1 receptor agonists. Conclusion: Our results suggest that R-flurbiprofen improves the endogenous mechanisms to regain stability after axonal injury and to fend off chronic neuropathic pain by modulating the endocannabinoid system and thus constitutes an attractive, novel therapeutic agent in the treatment of chronic, intractable pain.
Background: Due to the use of organophosphates (OP) as pesticides and the availability of OP-type nerve agents, an effective medical treatment for OP poisonings is still a challenging problem. The acute toxicity of an OP poisoning is mainly due to the inhibition of acetylcholinesterase (AChE) in the peripheral and central nervous systems (CNS). This results in an increase in the synaptic concentration of the neurotransmitter acetylcholine, overstimulation of cholinergic receptors and disorder of numerous body functions up to death. The standard treatment of OP poisoning includes a combination of a muscarinic antagonist and an AChE reactivator (oxime). However, these oximes can not cross the blood-brain barrier (BBB) sufficiently. Therefore, new strategies are needed to transport oximes over the BBB. Methodology/Principal Findings: In this study, we combined different oximes (obidoxime dichloride and two different HI 6 salts, HI 6 dichloride monohydrate and HI 6 dimethanesulfonate) with human serum albumin nanoparticles and could show an oxime transport over an in vitro BBB model. In general, the nanoparticulate transported oximes achieved a better reactivation of OP-inhibited AChE than free oximes. Conclusions/Significance: With these nanoparticles, for the first time, a tool exists that could enable a transport of oximes over the BBB. This is very important for survival after severe OP intoxication. Therefore, these nanoparticulate formulations are promising formulations for the treatment of the peripheral and the CNS after OP poisoning.
In dyadic communication, both interlocutors adapt to each other linguistically, that is, they align interpersonally. In this article, we develop a framework for modeling interpersonal alignment in terms of the structural similarity of the interlocutors’ dialog lexica. This is done by means of so-called two-layer time-aligned network series, that is, a time-adjusted graph model. The graph model is partitioned into two layers, so that the interlocutors’ lexica are captured as subgraphs of an encompassing dialog graph. Each constituent network of the series is updated utterance-wise. Thus, both the inherent bipartition of dyadic conversations and their gradual development are modeled. The notion of alignment is then operationalized within a quantitative model of structure formation based on the mutual information of the subgraphs that represent the interlocutor’s dialog lexica. By adapting and further developing several models of complex network theory, we show that dialog lexica evolve as a novel class of graphs that have not been considered before in the area of complex (linguistic) networks. Additionally, we show that our framework allows for classifying dialogs according to their alignment status. To the best of our knowledge, this is the first approach to measuring alignment in communication that explores the similarities of graph-like cognitive representations. Iločki jezikoslovni razmišljaji : (Marko Samardžija: Devet iločkih priopćenja i jedno warszawsko)

U radu se predstavljaju rezultati terenskog istraživanja o nazivima za uskršnju pletenicu, provedenog na gotovo dvjesto punktova duž hrvatske obale te u unutrašnjosti Istre i nadopunjenog podatcima iz objavljene literature. Istraživanjem su, radi usporedbe, zahvaćena i neka naselja u Gorskom kotaru te u unutrašnjosti Dalmacije. Analiza prikupljenog korpusa ukazuje na snažnu prisutnost slike sadržaja ‘ptica’ u našim jadranskim nazivima za uskršnju pletenicu, bilo da je riječ o nazivima slavenskog ili romanskog (dalmatskog, istroromanskog ili mletačkog) podrijetla. Na temelju tih zapažanja ponovno se razmatraju neka dosad predložena etimološka rješenja (Skok, Vinja) te donose novi etimološki prijedlozi.
U ovome su radu obradena 232 obiteljska nadimka u Puciscima na otoku Bracu. Obiteljski su nadimci, kao dodatan vid identifikacije koji se razvio još u pretprezimenskome razdoblju, a kasnije je sve zastupljeniji zbog brojnosti nositelja pojedinih prezimena, svojevrsni specifikum hrvatskih otoka koji dosad nije dostatno proucen. U Puciscima se obiteljski nadimci bilježe od konca 16. st. te se na temelju njihove motivacije može djelomicno rekonstruirati fond osobnih imena (odnos hrvatskih narodnih imena te hrvatskih i novijih romanskih prilagodenica kršcanskih imena), vanjština (posebice tjelesne mane), karakterne crte (uglavnom nekonvencionalne) te podrijetlo i svakodnevni život Puciscana. Fond je obiteljskih nadimaka znatno otvoreniji inojezicnim sustavima (poglavito romanskim) te je odraz svojevrsne tisucljetne hrvatsko-romanske simbioze na istocnoj obali Jadranskoga mora.
