Stammzellen aus dem Knochenmark werden seit Jahrzehnten gegen Blutkrebs eingesetzt. In der Zukunft sollen auch andere Krankheiten mit Stammzellen und therapeutischen Genen behandelt werden. Die an der Goethe-Universität geleisteten Vorarbeiten zeigen, dass der Standort wie kaum ein anderer geeignet ist, diese neuen und maßgeschneiderten Verfahren voran zubringen.
Während Bildungspolitiker heftig über die Abschaffung der Hauptschule streiten und Hamburger Bürger sich per Volksentscheid für den Erhalt des Gymnasiums ab Klasse 5 einsetzen, sind sich die meisten Bildungsforscher einig: Entscheidend ist nicht, wo, sondern wie Kinder unterrichtet und betreut werden. Wie kann es gelingen, pädagogische Interventionen den unterschiedlichen Lernvoraussetzungen so anzupassen, dass möglichst alle Schülerinnen und Schüler optimal gefördert werden? Um das herauszufi nden, bedarf es intensiver Anstrengungen in der Lehr-Lernforschung, wie sie in Frankfurt im Forschungszentrum IDeA unternommen werden.
Seit einigen Jahrzehnten wollen Biochemiker, Mediziner, Biologen und Pharmazeuten weltweit nicht mehr auf eine bioanalytische Methode verzichten, an deren Entwicklung der Frankfurter Wissenschaftler Prof. Dr. Michael Karas vom Institut für Pharmazeutische Chemie der Goethe-Universität maßgeblich beteiligt war. Die Rede ist von der Matrix-unterstützten Laser-Desorptions- / Ionisations-Massenspektrometrie – kurz MALDI-MS.
Sie sind nicht weniger intelligent als ihre gleichaltrigen Mitschüler, und doch sind ihre schulischen Leistungen meist schlechter: Kinder, die unter einer Störung mit Aufmerksamkeitsdefi zit und Hyperaktivität (ADHS) leiden, haben es schwer im Schulsystem. Wie können sie lernen, ihre Gefühle, Gedanken und Handlungen bewusst zu beeinfl ussen und sich nicht ständig ablenken zu lassen? Frankfurter Studien zeigen zum Beispiel, dass konkrete "Wenn-Dann-Pläne" ihre Konzentrationsfähigkeit deutlich verbessern können.
Yuniyanti Chuzaifah is one of the founders of "Voice of Concerned Mothers" (Suara Ibu Peduli or SIP) and later joined the "Coalition of Indonesian Women" (Koalisi Perempuan Indonesia or KPI). She studied at the Universities of Leiden and Amsterdam in the Netherlands, and later returned to Indonesia. Since March 2010 she has been the Director of the "Indonesian National Commission on Violence against Women" (Komnas Perempuan). This interview was conducted on 15 March 2010 at the organisation’s premises in Jakarta.
Wie entsteht Schmerz? Und wie kann man ihn lindern? Um diese Fragen beantworten zu können, untersuchen Forscher eine Gruppe von Schlüsselmolekülen, die Eicosanoide, und ihre Abbauprodukte. Dabei machen sie immer wieder überraschende Entdeckungen: Blockiert man etwa durch Schmerzmittel wie Acetylsalicylsäure gezielt die Entstehung des Abbauprodukts Prostaglandin, schützt dies auch vor Krebs. Verhindert man die Entstehung von Leukotrienen, lassen sich allergische Reaktionen wie Asthma, aber auch Krebs, Osteoporose und Herz-Kreislauf-Erkrankungen beeinflussen.
Introduction: Systemic inflammation (e.g. following surgery) involves Toll-like receptor (TLR) signaling and leads to an endocrine stress response. This study aims to investigate a possible influence of TLR2 and TLR4 single nucleotide polymorphisms (SNPs) on perioperative adrenocorticotropic hormone (ACTH) and cortisol regulation in serum of cardiac surgical patients. To investigate the link to systemic inflammation in this context, we additionally measured 10 different cytokines in the serum. Methods: 338 patients admitted for elective cardiac surgery were included in this prospective observational clinical cohort study. Genomic DNA of patients was screened for TLR2 and TLR4 SNPs. Serum concentrations of ACTH, cortisol, interferon (IFN)-, interleukin (IL)-1, IL-2, IL-4, IL-5, IL-6, IL-8, IL-10, tumor necrosis factor (TNF)- and granulocyte macro-phage-colony stimulating factor (GM-CSF) were determined before surgery, immediately post surgery and on the first postoperative day. Results: 13 patients were identified as TLR2 SNP carrier, 51 as TLR4 SNP carrier and 274 pa-tients as non-carrier. Basal levels of ACTH, cortisol and cytokines did not differ between groups. In all three groups a significant, transient perioperative rise of cortisol could be ob-served. However, only in the non-carrier group this was accompanied by a significant ACTH rise, TLR4 SNP carriers had significant lower ACTH levels compared to non-carriers ((mean[95% confidence intervals]) non-carriers: 201.9[187.7 to 216.1]pg/ml; TLR4 SNP car-riers: 149.9[118.4 to 181.5]pg/ml; TLR2 SNP carriers: 176.4[110.5 to 242.3]pg/ml). Compared to non-carriers, TLR4 SNP carriers showed significant lower serum IL-8, IL-10 and GM-CSF peaks ((mean[95% confidence intervals]): IL-8: non-carriers: 42.6[36.7 to 48.5]pg/ml, TLR4 SNP carriers: 23.7[10.7 to 36.8]pg/ml; IL-10: non-carriers: 83.8[70.3 to 97.4]pg/ml, TLR4 SNP carriers: 54.2[24.1 to 84.2]pg/ml; GM-CSF: non-carriers: 33.0[27.8 to 38.3]pg/ml, TLR4 SNP carriers: 20.2[8.6 to 31.8]pg/ml). No significant changes over time or between the groups were found for the other cytokines. Conclusions: Regulation of the immunoendocrine stress response during systemic inflamma-tion is influenced by the presence of a TLR4 SNP. Cardiac surgical patients carrying this ge-notype showed decreased serum concentrations of ACTH, IL-8, IL-10 and GM-CSF. This finding might have impact on interpreting previous and designing future trials on diagnosing and modulating immunoendocrine dysregulation (e.g. adrenal insufficiency) during systemic inflammation and sepsis.
Adhesion and host cell modulation: critical pathogenicity determinants of Bartonella henselae

Bartonella henselae, the agent of cat scratch disease and the vasculoproliferative disorders bacillary angiomatosis and peliosis hepatis, contains to date two groups of described pathogenicity factors: adhesins and type IV secretion systems. Bartonella adhesin A (BadA), the Trw system and possibly filamentous hemagglutinin act as promiscous or specific adhesins, whereas the virulence locus (Vir)B/VirD4 type IV secretion system modulates a variety of host cell functions. BadA mediates bacterial adherence to endothelial cells and extracellular matrix proteins and triggers the induction of angiogenic gene programming. The VirB/VirD4 type IV secretion system is responsible for, e.g., inhibition of host cell apoptosis, bacterial persistence in erythrocytes, and endothelial sprouting. The Trw-conjugation system of Bartonella spp. mediates host-specific adherence to erythrocytes. Filamentous hemagglutinins represent additional potential pathogenicity factors which are not yet characterized. The exact molecular functions of these pathogenicity factors and their contribution to an orchestral interplay need to be analyzed to understand B. henselae pathogenicity in detail.
PERIOD proteins are central components of the Drosophila and mammalian circadian clocks. The crystal structure of a Drosophila PERIOD (dPER) fragment comprising two PER-ARNT-SIM (PAS) domains (PAS-A and PAS-B) and two additional C-terminal alpha-helices (alphaE and alphaF) has revealed a homodimer mediated by intermolecular interactions of PAS-A with tryptophane 482 in PAS-B and helix alphaF. Here we present the crystal structure of a monomeric PAS domain fragment of dPER lacking the alphaF helix. Moreover, we have solved the crystal structure of a PAS domain fragment of the mouse PERIOD homologue mPER2. The mPER2 structure shows a different dimer interface than dPER, which is stabilized by interactions of the PAS-B beta-sheet surface including tryptophane 419 (equivalent to Trp482dPER). We have validated and quantitatively analysed the homodimer interactions of dPER and mPER2 by site-directed mutagenesis using analytical gel filtration, analytical ultracentrifugation, and co-immunoprecipitation experiments. Furthermore we show, by yeast-two-hybrid experiments, that the PAS-B beta-sheet surface of dPER mediates interactions with TIMELESS (dTIM). Our study reveals quantitative and qualitative differences between the homodimeric PAS domain interactions of dPER and its mammalian homologue mPER2. In addition, we identify the PAS-B beta-sheet surface as a versatile interaction site mediating mPER2 homodimerization in the mammalian system and dPER-dTIM heterodimer formation in the Drosophila system.
In the next years the Facility for Antiproton and Ion Research FAIR will be constructed at the GSI Helmholtzzentrum fur Schwerionenforschung in Darmstadt, Germany. This new accelerator complex will allow for unprecedented and pathbreaking research in hadronic, nuclear, and atomic physics as well as in applied sciences. This manuscript will discuss some of these research opportunities, with a focus on few-body physics.
The energy dependence of the local and violation in Au+Au and Cu+Cu collisions in a large energy range is estimated within a simple phenomenological model. It is expected that at LHC the chiral magnetic effect will be about 20 times weaker than at RHIC. At lower energy range, covered by the low-energy scan at RHIC and future NICA/FAIR facilities, the created magnetic field strength and energy density of deconfined matter are rather high providing necessary conditions for the chiral magnetic effect. However, the particular model for the chiral magnetic effect predicts that this effect should vanish sharply at energy somewhere above the top SPS one. To elucidate CME background effects the Hadron-String-Dynamics (HSD) transport model including electromagnetic fields is put forward. Importance of new planning experiments at LHC and for the low-energy RHIC scan program is emphasized.
Effects of nuclear orientation on fusion and fission in the reaction using 238U target nucleus
(2010)
Fission fragment mass distributions in the reaction of 30Si+238U were measured around the Coulomb barrier. At the above-barrier energies, the mass distribution showed a Gaussian shape. At the subbarrier energies, triple-humped distribution was observed, which consists of symmetric fission and asymmetric fission peaked at AL/AH ~ 90/178. The asymmetric fission should be attributed to quasifission from the results of the measured evaporation residue (ER) cross-sections for 30Si+238U. The cross-section for 263Sg at the abovebarrier energy agree with the statistical model calculation which assumes that the measured fission cross-section originates from fusion-fission, whereas the one for 264 Sg measured at the sub-barrier energy is smaller than the calculation, which suggests the presence of quasifission.
We examine the scaling trends in particle multiplicity and flow observables between SPS, RHIC and LHC, and discuss their compatibility with popular theoretical models. We examine the way scaling trends between SPS and RHIC are broken at LHC energies, and suggest experimental measurements which can further clarify the situation.
The mass-dependent structure of the composite nucleus is shown based on three-dimensional timedependent Hartree-Fock calculations with Skyrme interactions (SLy4d and SkM*). One remarkable result is that the isovector monopole excitation dominantly appears for collisions of heavy nuclei, and the isovector dipole excitation for those of light ones. Such a difference found in the dynamical structure of composite nucleus plays a role in the equilibration of charge.
We derive the equations of second order dissipative fluid dynamics from the relativistic Boltzmann equation following the method of W. Israel and J. M. Stewart [1]. We present a frame independent calculation of all first- and second-order terms and their coefficients using a linearised collision integral. Therefore, we restore all terms that were previously neglected in the original papers of W. Israel and J. M. Stewart.
We present results on Hanbury Brown-Twiss (HBT) radii extracted from the Ultra-relativistic Molecular Dynamics (UrQMD) approach to relativistic heavy ion collisions. The present investigation provides a comparison of results from pure hadronic transport calculations to a Boltzmann + Hydrodynamic hybrid approach with an intermediate hydrodynamic phase. For the hydrodynamic phase different Equations of State (EoS) have been employed, i.e. bag model, hadron resonance gas and a chiral EoS. The influence of various freeze-out scenarios has been investigated and shown to be negligible if hadronic rescatterings after the hydrodynamic evolution are included. Furthermore, first results of the source tilt from azimuthal sensitive HBT and the direct extraction from the transport model are presented and exhibit a very good agreement with E895 data at AGS.
A mechanism for locally density-dependent dynamic parton rearrangement and fusion has been implemented into the Ultrarelativistic Quantum Molecular Dynamics (UrQMD) approach. The same mechanism has been previously built in the Quark Gluon String Model (QGSM). This rearrangement and fusion approach based on parton coalescence ideas enables the description of multi-particle interactions, namely 3 -> 3 and 3 -> 2, between (pre)hadronic states in addition to standard binary interactions. The UrQMD model (v2.3) extended by these additional processes allows to investigate implications of multi-particle interactions on the reaction dynamics of ultrarelativistic heavy ion collisions. The mechanism, its implementation and first results of this investigation are presented and discussed.
We present the current status of hybrid approaches to describe heavy ion collisions and their future challenges and perspectives. First we present a hybrid model combining a Boltzmann transport model of hadronic degrees of freedom in the initial and final state with an optional hydrodynamic evolution during the dense and hot phase. Second, we present a recent extension of the hydrodynamical model to include fluctuations near the phase transition by coupling a chiral field to the hydrodynamic evolution.
Fast thermalization and a strong build up of elliptic flow of QCD matter were investigated within the pQCD based 3+1 dimensional parton transport model BAMPS including bremsstrahlung 2 <-> 3 processes. Within the same framework quenching of gluonic jets in Au+Au collisions at RHIC can be understood. The development of conical structure by gluonic jets is investigated in a static box for the regimes of small and large dissipation. Furthermore we demonstrate two different approaches to extract the shear viscosity coefficient n from a microscopical picture.
We study the kinetic and chemical equilibration in 'infinite' parton-hadron matter within the Parton-Hadron-String Dynamics transport approach, which is based on a dynamical quasiparticle model for partons matched to reproduce lattice-QCD results – including the partonic equation of state – in thermodynamic equilibrium. The 'infinite' matter is simulated within a cubic box with periodic boundary conditions initialized at different baryon density (or chemical potential) and energy density. The transition from initially pure partonic matter to hadronic degrees of freedom (or vice versa) occurs dynamically by interactions. Different thermody-namical distributions of the strongly-interacting quark-gluon plasma (sQGP) are addressed and discussed.
Heavy quark and charmonium production as well as their space-time evolution are studied in transport simulations of heavy-ion collisions at RHIC and LHC. In the partonic transport model Boltzmann Approach of MultiParton Scatterings (BAMPS) heavy quarks can be produced in initial hard parton scatterings or during the evolution of the quark-gluon plasma. Subsequently, they interact with the medium via binary scatterings with a running coupling and a more precise Debye screening which is derived from hard thermal loop calculations, participate in the flow and lose energy. We present results of the elliptic flow and nuclear modification factor of heavy quarks and compare them to available data. Furthermore, preliminary results on J/psi suppression at forward and mid-rapidity are reported for central and non-central collisions at RHIC. For this, we study cold nuclear matter effects and the dissociation as well as regeneration of J/psi in the quark-gluon plasma. The eukaryotic glyoxalase system consists of two enzymatic components, glyoxalase I (lactoylglutathionelyase) and glyoxalase II (hydroxyacylglutathione hydrolase). These enzymes are dedicated to the removal of toxic alpha-oxoaldehydes like methylglyoxal (MG). MG is formed as a by-product of glycolysis and MG toxicity results from its damaging capability leading to modifications of proteins, lipids and nucleic acids. An efficient removal of MG appears to be essential to ensure cellular functionality and viability. Here we study the effects of the genetic modulation of genes encoding the components of the glyoxalase system in the filamentous ascomycete and aging model Podospora anserina. Overexpression of PaGlo1 leads to a lifespan reduction on glucose rich medium, probably due to depletion of reduced glutathione. Deletion of PaGlo1 leads to hypersensitivity against MG added to the growth medium. A beneficial effect on lifespan is observed when both PaGlo1 and PaGlo2 are overexpressed and the corresponding strains are grown on media containing increased glucose concentrations. Notably, the double mutant has a ‘healthy’ phenotype without physiological impairments. Moreover, PaGlo1/PaGlo2_OEx strains are not long-lived on media containing standard glucose concentrations suggesting a tight correlation between the efficiency and capacity to remove MG within the cell, the level of available glucose and lifespan. Overall, our results identify the up-regulation of both components of the glyoxalase system as an effective intervention to increase lifespan in P. anserina. TIM23-mediated insertion of transmembrane alpha-helices into the mitochondrial inner membrane

While overall hydrophobicity is generally recognized as the main characteristic of transmembrane (TM) alpha-helices, the only membrane system for which there are detailed quantitative data on how different amino acids contribute to the overall efficiency of membrane insertion is the endoplasmic reticulum (ER) of eukaryotic cells. Here, we provide comparable data for TIM23-mediated membrane protein insertion into the inner mitochondrial membrane of yeast cells. We find that hydrophobicity and the location of polar and aromatic residues are strong determinants of membrane insertion. These results parallel what has been found previously for the ER. However, we see striking differences between the effects elicited by charged residues flanking the TM segments when comparing the mitochondrial inner membrane and the ER, pointing to an unanticipated difference between the two insertion systems. G-quadruplex topologies of telomeric repeat sequences from vertebrates were investigated in the presence of molecular crowding (MC) mimetics, namely polyethylene glycol 200 (PEG), Ficoll 70 as well as Xenopus laevis egg extract by CD and NMR spectroscopy and native PAGE. Here, we show that the conformational behavior of the telomeric repeats in X. laevis egg extract or in Ficoll is notably different from that observed in the presence of PEG. While the behavior of the telomeric repeat in X. laevis egg extract or in Ficoll resembles results obtained under dilute conditions, PEG promotes the formation of high-order parallel topologies. Our data suggest that PEG should not be used as a MC mimetic.
Background: FTY720, an immunomodulator derived from a fungal metabolite which reduces circulating lymphocyte counts by increasing the homing of lymphocytes to the lymph nodes has recently gained interest in stroke research. The aim of this study was to evaluate the protective efficacy of FTY720 in cerebral ischemia in two different application paradigms and to gather first data on the effect of FTY720 on the rate of spontaneous bacterial infections in experimental stroke. Methods: Middle cerebral artery occlusion (MCAO) in C57BL/6 mice (strain J, groups of 10 animals) was performed with two different durations of ischemia (90 min and 3 h) and FTY720 was applied 2 h after vessel occlusion to study the impact of reperfusion on the protective potency of FTY720. Lesion size was determined by TTC staining. Mice treated with FTY720 or vehicle were sacrificed 48 h after 90 min MCAO to determine the bacterial burden in lung and blood. Results: FTY720 1 mg/kg significantly reduced ischemic lesion size when administered 2 h after the onset of MCAO for 3 h (45.4 +/- 22.7 mm3 vs. 84.7 +/- 23.6 mm3 in control mice, p = 0.001) and also when administered after reperfusion, 2 h after the onset of MCAO for 90 min (31.1 +/- 28.49 mm3 vs. 69.6 +/- 27.2 mm3 in control mice, p = 0.013). Bacterial burden of lung homogenates 48 h after stroke did not increase in the group treated with the immunomodulator FTY720 while there was no spontaneous bacteremia 48 h after MCAO in treated and untreated animals. Conclusions: Our results corroborate the experimental evidence of the protective effect of FTY720 seen in different rodent stroke models. Interestingly, we found no increase in bacterial lung infections even though FTY720 strongly reduces the number of circulating leukocytes.
Background: Studies on allele length polymorphism designate several glacial refugia for Norway spruce (Picea abies) in the South Carpathian Mountains, but infer only limited expansion from these refugia after the last glaciation. To better understand the genetic dynamics of a South Carpathian spruce lineage, we compared ancient DNA from 10,700 and 11,000-year-old spruce pollen and macrofossils retrieved from Holocene lake sediment in the Retezat Mountains with DNA extracted from extant material from the same site. We used eight primer pairs that amplified short and variable regions of the spruce cpDNA. In addition, from the same lake sediment we obtained a 15,000-years-long pollen accumulation rate (PAR) record for spruce that helped us to infer changes in population size at this site. Results: We obtained successful amplifications for Norway spruce from 17 out of 462 pollen grains tested, while the macrofossil material provided 22 DNA sequences. Two fossil sequences were found to be unique to the ancient material. Population genetic statistics showed higher genetic diversity in the ancient individuals compared to the extant ones. Similarly, statistically significant Ks and Kst values showed a considerable level of differentiation between extant and ancient populations at the same loci. Lateglacial and Holocene PAR values suggested that population size of the ancient population was small, in the range of 1/10 or 1/5 of the extant population. PAR analysis also detected two periods of rapid population growths (from ca. 11,100 and 3900 calibrated years before present (cal yr BP)) and three bottlenecks (around 9180, 7200 and 2200 cal yr BP), likely triggered by climatic change and human impact. Conclusion: Our results suggest that the paternal lineages observed today in the Retezat Mountains persisted at this site at least since the early Holocene. Combination of the results from the genetic and the PAR analyses furthermore suggests that the higher level of genetic variation found in the ancient populations and the loss of ancient allele types detected in the extant individuals were likely due to the repeated bottlenecks during the Holocene. This study demonstrates how past population size changes inferred from PAR records can be efficiently used in combination with ancient DNA studies. The joint application of palaeoecological and population genetic analyses proved to be a powerful tool to understand the influence of past population demographic changes on the haplotype diversity and genetic composition of forest tree species.
Anfang des 20. Jahrhunderts regierte Europa über ca. 85% des globalen Territoriums in Form von Kolonien, Protektoraten und Dependancen. Die koloniale Expansion war ein exorbitanter und gewalttätiger Prozess, der durch Ausbeutung, Versklavung und Diebstahl charakterisiert war. Es stellt sich deswegen die Frage, warum sich innerhalb der westlichen wissenschaftlichen Disziplinen lange Zeit nur eine kleine Minderheit diesem Ereignis analytisch angenommen hat. Keine große intellektuelle Anstrengung ist vonnöten, um zu verstehen, dass eine solch massive territoriale Expansion, die zum Teil über Jahrhunderte gewaltvoll erhalten wurde, erstens nicht nur durch militärische Präsenz möglich war, zweitens nicht mit der bloßen formalen Unabhängigkeit der kolonisierten Staaten zu einem Ende kommen konnte und schließlich kaum nur Spuren in den kolonisierten Ländern hinterlassen haben kann, sondern auch den globalen Norden prägten. Postkoloniale Studien nähern sich dieser Komplexität und irritieren dabei die Vorstellung einer zwangsläufigen, geradezu naturwüchsigen, kolonialen Beherrschung durch Europa. Sie werfen einen Blick auf die Mannigfaltigkeit kolonialer Interventionen und deren Wirkmächtigkeit bis in die heutigen Tage (etwa Randeria/Eckert 2009).
In ihrem Buch "Cultivating Humanity" formuliert Martha Nussbaum folgenden Appell: "(…) die Welt um uns herum ist unausweichlich international. Themen vom Handel bis zur Landwirtschaft – über die Menschenrechte bis hin zu der Linderung von Hungersnöten – fordern uns dazu heraus, den Blick über eng gefasste Gruppenloyalitäten hinaus zu wagen und weit entfernte Lebenswirklichkeiten zu berücksichtigen. (…) Die Kultivierung unserer Menschlichkeit in einer komplexen und ineinander verflochtenen Welt, bedarf eines Verständnisses über die Art und Weise in der gemeinsame Bedürfnisse und Ziele in unterschiedlichen Lebensverhältnissen je verschieden identifiziert und verfolgt werden" (1997, 10). Diese Forderung, die das liberale westliche Individuum dazu aufruft, sich angesichts zunehmender globaler Interdependenzen für Belange verantwortlich zu zeigen, die über das jeweilige Eigeninteresse hinausgehen, erscheint auf den ersten Blick als ein überaus lobenswertes Unternehmen.
Background: Although literature provides support for cognitive behavioral therapy (CBT) as an efficacious intervention for social phobia, more research is needed to improve treatments for children. Methods: Forty four Caucasian children (ages 8-14) meeting diagnostic criteria of social phobia according to the Diagnostic and Statistical Manual of Mental Disorders (4th ed.; APA, 1994) were randomly allocated to either a newly developed CBT program focusing on cognition according to the model of Clark and Wells (n = 21) or a wait-list control group (n = 23). The primary outcome measure was clinical improvement. Secondary outcomes included improvements in anxiety coping, dysfunctional cognitions, interaction frequency and comorbid symptoms. Outcome measures included child report and clinican completed measures as well as a diagnostic interview. Results: Significant differences between treatment participants (4 dropouts) and controls (2 dropouts) were observed at post test on the German version of the Social Phobia and Anxiety Inventory for Children. Furthermore, in the treatment group, significantly more children were free of diagnosis than in wait-list group at post-test. Additional child completed and clinician completed measures support the results. Discussion: The study is a first step towards investigating whether CBT focusing on cognition is efficacious in treating children with social phobia. Future research will need to compare this treatment to an active treatment group. There remain the questions of whether the effect of the treatment is specific to the disorder and whether the underlying theoretical model is adequate. Conclusion: Preliminary support is provided for the efficacy of the cognitive behavioral treatment focusing on cognition in socially phobic children. Active comparators should be established with other evidence-based CBT programs for anxiety disorders, which differ significantly in their dosage and type of cognitive interventions from those of the manual under evaluation (e.g. Coping Cat).
Background: The interferon-inducible immunity-related GTPases (IRG proteins/p47 GTPases) are a distinctive family of GTPases that function as powerful cell-autonomous resistance factors. The IRG protein, Irga6 (IIGP1), participates in the disruption of the vacuolar membrane surrounding the intracellular parasite, Toxoplasma gondii, through which it communicates with its cellular hosts. Some aspects of the protein's behaviour have suggested a dynamin-like molecular mode of action, in that the energy released by GTP hydrolysis is transduced into mechanical work that results in deformation and ultimately rupture of the vacuolar membrane. Results: Irga6 forms GTP-dependent oligomers in vitro and thereby activates hydrolysis of the GTP substrate. In this study we define the catalytic G-domain interface by mutagenesis and present a structural model, of how GTP hydrolysis is activated in Irga6 complexes, based on the substrate-twinning reaction mechanism of the signal recognition particle (SRP) and its receptor (SRalpha). In conformity with this model, we show that the bound nucleotide is part of the catalytic interface and that the 3'hydroxyl of the GTP ribose bound to each subunit is essential for trans-activation of hydrolysis of the GTP bound to the other subunit. We show that both positive and negative regulatory interactions between IRG proteins occur via the catalytic interface. Furthermore, mutations that disrupt the catalytic interface also prevent Irga6 from accumulating on the parasitophorous vacuole membrane of T. gondii, showing that GTP-dependent Irga6 activation is an essential component of the resistance mechanism. Conclusions: The catalytic interface of Irga6 defined in the present experiments can probably be used as a paradigm for the nucleotide-dependent interactions of all members of the large family of IRG GTPases, both activating and regulatory. Understanding the activation mechanism of Irga6 will help to explain the mechanism by which IRG proteins exercise their resistance function. We find no support from sequence or G-domain structure for the idea that IRG proteins and the SRP GTPases have a common phylogenetic origin. It therefore seems probable, if surprising, that the substrate-assisted catalytic mechanism has been independently evolved in the two protein families.
Background: Although being considered as a rarely observed HIV-1 protease mutation in clinical isolates, the L76V-prevalence increased 1998-2008 in some European countries most likely due to the approval of Lopinavir, Amprenavir and Darunavir which can select L76V. Beside an enhancement of resistance, L76V is also discussed to confer hypersusceptibility to the drugs Atazanavir and Saquinavir which might enable new treatment strategies by trying to take advantage of particular mutations. Results: Based on a cohort of 47 L76V-positive patients, we examined if there might exist a clinical advantage for L76V-positive patients concerning long-term success of PI-containing regimens in patients with limited therapy options. Genotypic- and phenotypic HIV-resistance tests from 47 mostly multi-resistant, L76V-positive patients throughout Germany were accomplished retrospectively 1999-2009. Five genotype-based drug-susceptibility predictions received from online interpretation-tools for Atazanavir, Saquinavir, Amprenavir and Lopinavir, were compared to phenotype-based predictions that were determined by using a recombinant virus assay along with a Virtual Phenotype™(Virco). The clinical outcome of the L76V-adapted follow-up therapy was determined by monitoring viral load for 96 weeks. Conclusions: In this analysis, the mostly used interpretation systems overestimated the L76V-mutation concerning Atazanavir- and SQV resistance. In fact, a clear benefit in drug susceptibility for these drugs was observed in phenotype analysis after establishment of L76V. More importantly, long-term therapy success was significantly higher in patients receiving Atazanavir and/or Saquinavir plus one L76V-selecting drug compared to patients without L76V-selecting agents (p = 0.002). In case of L76V-occurrence ATV and/or SQV may represent encouraging options for patients in deep salvage situations.
Background: The combination of high-throughput transcript profiling and next-generation sequencing technologies is a prerequisite for genome-wide comprehensive transcriptome analysis. Our recent innovation of deepSuperSAGE is based on an advanced SuperSAGE protocol and its combination with massively parallel pyrosequencing on Roche's 454 sequencing platform. As a demonstration of the power of this combination, we have chosen the salt stress transcriptomes of roots and nodules of the third most important legume crop chickpea (Cicer arietinum L.). While our report is more technology-oriented, it nevertheless addresses a major world-wide problem for crops generally: high salinity. Together with low temperatures and water stress, high salinity is responsible for crop losses of millions of tons of various legume (and other) crops. Continuously deteriorating environmental conditions will combine with salinity stress to further compromise crop yields. As a good example for such stress-exposed crop plants, we started to characterize salt stress responses of chickpeas on the transcriptome level. Results: We used deepSuperSAGE to detect early global transcriptome changes in salt-stressed chickpea. The salt stress responses of 86,919 transcripts representing 17,918 unique 26bp deepSuperSAGE tags (UniTags) from roots of the salt-tolerant variety INRAT-93 two hours after treatment with 25 mM NaCl were characterized. Additionally, the expression of 57,281 transcripts representing 13,115 UniTags was monitored in nodules of the same plants. From a total of 144,200 analyzed 26bp tags in roots and nodules together, 21,401 unique transcripts were identified. Of these, only 363 and 106 specific transcripts, respectively, were commonly up- or down-regulated (>3.0-fold) under salt stress in both organs, witnessing a differential organ-specific response to stress. Profiting from recent pioneer works on massive cDNA sequencing in chickpea, more than 9,400 UniTags were able to be linked to UniProt entries. Additionally, gene ontology (GO) categories over-representation analysis enabled to filter out enriched biological processes among the differentially expressed UniTags. Subsequently, the gathered information was further cross-checked with stress-related pathways. From several filtered pathways, here we focus exemplarily on transcripts associated with the generation and scavenging of reactive oxygen species (ROS), as well as on transcripts involved in Na+ homeostasis. Although both processes are already very well characterized in other plants, the information generated in the present work is of high value. Information on expression profiles and sequence similarity for several hundreds of transcripts of potential interest is now available. Conclusions: This report demonstrates, that the combination of the high-throughput transcriptome profiling technology SuperSAGE with one of the next-generation sequencing platforms allows deep insights into the first molecular reactions of a plant exposed to salinity. Cross validation with recent reports enriched the information about the salt stress dynamics of more than 9,000 chickpea ESTs, and enlarged their pool of alternative transcripts isoforms. As an example for the high resolution of the employed technology that we coin deepSuperSAGE, we demonstrate that ROS-scavenging and -generating pathways undergo strong global transcriptome changes in chickpea roots and nodules already 2 hours after onset of moderate salt stress (25mM NaCl). Additionally, a set of more than 15 candidate transcripts are proposed to be potential components of the salt overly sensitive (SOS) pathway in chickpea. Newly identified transcript isoforms are potential targets for breeding novel cultivars with high salinity tolerance. We demonstrate that these targets can be integrated into breeding schemes by micro-arrays and RT-PCR assays downstream of the generation of 26bp tags by SuperSAGE.
Vacuum-assisted closure (VAC) of complex infected wounds has recently gained popularity among various surgical specialties. The system is based on the application of negative pressure by controlled suction to the wound surface. The effectiveness of the VAC System on microcirculation and the promotion of granulation tissue proliferation are proved. In our case report we illustrate a scenario were a patient developed severe bleeding from the ascending aorta by penetration of wire fragments in the vessel. We conclude that all free particles in the sternum have to be removed completely before negative pressure is used.
Im ersten Teil des Interviews mit dem italienischen Politologen Marco Revelli (Arranca! Nr. 12) ging es um die Funktionsweise der fordistischen und postfordistischen Fabrik, um die neuentstehenden und in die Gesellschaft verlagerten Konflikte sowie um die tendenziell überflüssige Rolle der Gewerkschaften. Im vorliegenden zweiten Teil nimmt Revelli zu der Frage nach Widerstandsmöglichkeiten Stellung.
Marco Revelli, eine der zentralen Figuren der Unibesetzung in Turin 1967, lehrt heute an der Fakultät für politische Wissenschaften der selben Uni. Revelli forschte und publizierte verschiedene Bücher zu Faschismus sowie zum Postfordismus, unter anderem auch einen Beitrag über Fordismus und Toyotismus für das Buch "Verabredungen zum Jahrhundertende" von Rossana Rossanda und Pietro Ingrao. In der deutschen Ausgabe wurde der Beitrag Revellis allerdings nicht veröffentlicht, er erschien im April ‘97 als Beilage zu der Zeitschrift "Sozialismus".
In Genua war bereits im Oktober 1969 die bewaffnete Gruppe "22. Oktober" als Reaktion auf die Kämpfe 1968/69 entstanden. Sie hatte die "Auslösung eines revolutionären Partisanenkrieges" zum Ziel. Um den Großverleger Feltrinelli bilden sich, kurz nach dem Mailänder Anschlag, die Partisanenaktionsgruppen GAP. Sie versuchen an die Tradition der Partisanen anzuknüpfen und vertreten ein klassisches Guerillakonzept. Im November 1970 entstehen die Roten Brigaden (BR) aus Teilen der Organisation "Proletarische Linke" und der Einheitsbasiskomitees (CUB) der Mailänder Fabriken. Sie sind vor allem bei Pirelli aktiv, greifen mit Anschlägen auf Autos von Abteilungscapos unterstützend in die Arbeiterkämpfe ein und agieren zunächst relativ offen. ...
In den Kämpfen vom Juni/Juli 1960 in Genua war der Dissens zur klassenversöhnlerischen und abwiegelnden Haltung der Italienischen Kommunistischen Partei (PCI) deutlich zu Tage getreten. Dabei waren Jugendliche als besonders militant aufgefallen. Die Medien nannten sie die "Jugendlichen mit den gestreiften Trikots". Die Bezeichnung machte deutlich, daß sie keine Arbeiter waren, zeugte aber auch von der Unfähigkeit, sie einzuordnen. Diese Generation von Jugendlichen war geprägt von einer Unzufriedenheit mit den rigiden Normender italienischen Nachkriegsgesellschaft. Die PCI war zwar auch für sie, in Ermangelung von Alternativen, der einzige Bezugspunkt. Doch ihre Vorstellungen standen im krassen Widerspruch zu der von der PCI propagierten Arbeitsmoral. Sie fanden sich ehr in der Haltung der aus Süditalien stammenden Arbeiter wieder. Allein von Ende der fünfziger bis Mitte der sechziger Jahre waren es nahezu anderthalb Millionen, die aus dem bäuerlichen Süden in den Norden auswanderten und ihren Platz am Band einnahmen. Für diese Massenarbeiter stellte sich Arbeit nicht wie von der PCI propagiert als "Befreiung" dar, sondern als Plackerei. ...
Im Westen nichts Neues : zur Tauglichkeit des Imperialismus-Begriffes für die aktuelle Analyse

Rund um den Krieg gegen Jugoslawien erlebte der bereits etwas angestaubte Begriff "Imperialismus" eine erneute Renaissance. Der NATO-Angriff wurde als "imperialistischer Eingriff" tituliert und ganz Hartgesottenen hielten die Lektüre Lenins für unausweichlich, um den Werdegang der Welt zu verstehen. Wir gehen allerdings davon aus, dass der Begriff "Imperialismus", die Realität nicht mehr zu erfassen vermag und folglich auch kein Analyseinstrument mehr darstellt, das Handlungsoptionen aufzeigen kann. Angesichts der weitreichenden Veränderungen der Produktionsformen und politisch-ökonomischen Regulationsmechanismen im Verlauf der letzten zwei Jahrzehnte haben sich die militärischen Interventionen in ihrer Qualität grundlegend verändert. Dies macht sie weder besser noch schlechter als den altbekannten Imperialismus, doch ihre Andersartigkeit verlangt zur Kenntnis genommen zu werden, da es sonst schwer fällt politische Gegenstrategien zu entwickeln.
Marco Revelli, un des acteurs de l’occupation de l’université de Turin en 1967, est aujourd’hui professeur de sciences politiques de cette même université. Il a publié de nombreux livres et textes sur le fordisme et le post-fordisme. "Nation", "Nationalismus", "ethnischer Konflikt" usw. sind häufig verwendete Formulierungen. Hier soll ein Überblick über verschiedene Definitionen und Analysen erfolgen. Die Darstellung bleibt auf Ansätze beschränkt, die Ethnizität als Form sozialen Handelns analysieren, da Ansätze, die sich auf biologische Erbfaktoren, "Charaktereigenschaften" etc. berufen, für Linke sicher keinen Wert darstellen. Die beschriebenen Ansätze können zwei Funktionen erfüllen: Zum einen als analytisches Handwerkszeug dienen, zum anderen die Handlungsgrundlage, d.h. das bewußte theoretische Fundament, von Akteuren bilden. Bei der Anwendung auf politische Bewegungen und historische Situationen muß jedoch immer bedacht werden, daß politische Kultur und Verwendung bestimmter Begriffe nur vor dem konkreten historischen Hintergrund und der spezifischen Erfahrungswelt nachzuvollziehen sind. Der eigene Maßstab darf nicht zum einzig verbindlichen erklärt werden. ...
Im öffentlichen Diskurs wird Kolumbien vor allem mit Drogenhandel assoziiert. Der über Jahre angehäufte Reichtum der Rauschgiftkartelle beläuft sich mittlerweile zwar auf rund 40% des Gesamtbesitzes im Land, das Drogengeschäft macht jedoch "lediglich" 6% des BIP aus; das ist wesentlich weniger als in Bolivien und Peru. Anbau und Handel mit Koka/Kokain und in geringerem Maße auch mit Opium/Heroin haben keines der Bürgerkriegs-Probleme geschaffen, sondern bestehende nur verschärft. Wo also liegen die eigentlichen Ursachen für die brutalen Menschenrechtsverletzungen im jahrzehntealten Bürgerkrieg, in dem seit 1964 über 200000 Menschen ums Leben kamen? ...
Dass Arbeit und Produktion seit einigen Jahrzehnten einem rasanten Wandel in Form und Inhalt unterliegen, ist mittlerweile schon ein Allgemeinplatz. Doch die Reaktionen der Linken auf diese Veränderungen sind nach wie vor erschreckend. Die alte staatsfixierte Linke, also alle Kinder der Sozialdemokratie, von den Gewerkschaften bis hin zu den Parteikommunisten jeglicher Couleur, hat sich im Groben auf zwei Positionen verteilt: auf der einen Seite die "besseren Modernisierer", die Sozialdemokraten, mit denen wir es in fast allen Staaten der EU zu tun haben und deren "Neoliberalismus" sich vom us-amerikanischen Modell nur darin unterscheidet, dass diese den Sozialstaat (noch stärker als bisher) in eine Zwangsgemeinschaft verwandeln, um so das wegfallende Disziplinierungsmoment "Lohnarbeit" durch andere zu ersetzen und gleichzeitig Arbeits- und Beschäftigungsverhältnisse zu deregulieren. So wird das Bild des "aktivierenden Staates" propagiert, d.h. eine Abkehr vom früheren Wohlfahrtsstaat im Rahmen dessen - mal ganz abgesehen von seiner Disziplinierungs- und Kontrollfunktion - die Unterstützung ein Recht darstellte und die EmpfängerInnen in erster Linie alimentiert wurden. Das führt zu einem Modell staatlicher Förderung, in dem für Unterstützung eine Gegenleistung erwartet wird und die EmpfängerInnen von Unterstützung "animiert" (besser gesagt: gezwungen) werden sollen, die Leistungen nicht mehr in Anspruch zu nehmen und sich "Arbeit zu suchen". Da dies auf dem regulären Arbeitsmarkt kaum noch möglich ist, wird die "Selbstständigkeit" als Allheilmittel und Chance propagiert. So zahlt etwa das Arbeitsamt an "Arbeitslose", die eine selbstständige Aktivität in die Wege leiten, die Unterstützung sechs Monate weiter - danach erlöschen aber jegliche Ansprüche. Dass es sich hierbei um die Durchsetzung von Maßnahmen handelt, die darauf abzielen, Arbeitsverhältnissse weiter zu deregulieren und die in einen ideolgogischen Rahmen eingebettet sind, ist offensichtlich. Die hierin propagierten Anforderungen stehen aber im Widerspruch zur Selbstwahrnehmung vieler selbstständig Beschäftigter. ...
"Genua bedeutet zunächst eine Wende. Es war die gräßte Mobilisierung, die diese Bewegung bislang weltweit auf die Beine gestellt hat. Allerdings ist es nicht einfach für eine gerade entstandene Bewegung, einen solchen Schlag zu Überleben. Deshalb würde ich in Bezug auf Genua auch nicht von einem Erfolg sprechen, sondern eher von einer Etappe: Wir haben unsere Unschuld verloren, weil wir uns unserer Rechte zu sicher waren. (...) Genua zeugt von einer militärischen Logik der Regierung bezüglich der Mobilisierungen gegen den G8. Wir haben Widerstand geleistet und auch der Widerstand war eine politische Angelegenheit. Doch die militärische Logik des Konflikts zu akzeptieren, wäre Wahnsinn und politischer Selbstmord. In Genua waren alle Ordnungskräfte, die Armee, die Geheimdienste der acht - äkonomisch und militärisch - mächtigsten Länder des Planeten. Unsere Bewegung kann sich nicht mit dieser Militärmacht messen. Wir würden im Laufe von drei Monaten zerdrückt werden. Also müssen wir einen dritten Weg finden zwischen denen, die ihre Ablehnung der ökonomischen Globalisierung nur verbal äußern und jenen, die sich für die symbolische Geste entscheiden wie es z.B. das Demolieren einer Bank sein kann. Selbstständig Beschäftigten wird in der Öffentlichkeit ein erhöhter Individualismus und Egoismus nachgesagt. Gleich, ob die ihnen zugeordneten Eigenschaften positiv oder negativ bewertet werden, scheint Einigkeit zu bestehen, dass sie kein Interesse an einer organisierten Interessensvertretung haben. Sei es weil sie in Gewerkschaftsform als anachronistisch und fortschrittshemmend angesehen wird oder weil die vermeintlich privilegierte Situation der Selbstständigen eine Interessensvertretung überflüssig oder unerwünscht macht. Im folgenden Beitrag sollen einerseits die Relevanz der Frage der Organisierung und Interessensvertretung im Bereich der selbstständig Beschäftigten dargelegt werden und andererseits ihr Verhältnis zu traditionellen Interessensvertretungen (Gewerkschaften, Kammern usw.) sowie ihre Vorstellungen von einer Interessensvertretung dargestellt werden. Grundlage für den zweiten Teil sind die Ergebnisse einer Untersuchung in Berlin im Rahmen der zweijährigen Studie "Moriana" (1999/2000) des Mailänder Instituts A.A.Ster in sieben europäischen Städten. Diese hatte die Erforschung der neuen sozialen Zusammensetzung und der sich ausbreitenden neuen Arbeitsformen in metropolitanen Zentren zum Ziel, die während des Fordismus durch das Modell des urbanen Industriekapitalismus gekennzeichnet waren, in der heutigen Transitionsphase durch Formen informeller Ökonomie, gesellschaftlicher Ökonomie und Dienstleistungsökonomie. ...
Hugo Chávez je leta 1989 s 56, 4% dobil predsedniške volitve. Uporabljal je protineoliberalni diskurz, še posebej proti privatizaciji državnega naftnega koncerna Petroleos de Venezuela S. A. (PDVSA) in obljubil volitve za ustavodajno skupšcino. Drugega februarja 1999 je prevzel mandat, 25. julija je bila izvoljena ustavodajna skupšcina, 15. decembra je bila preko referenduma z 80% glasov izvoljena nova ustava, "Bolivarijanska ustava". Leta 2000 je bil Chávez v skladu z novo ustavo in z 59,7% glasov ponovno izvoljen. Njegovo kampanjo so podpirale skoraj vse leve stranke, bazicni sindikati, staroselske kmetice in kmetje ter bazicne organizacije. Trenutno je vlada sestavljena iz petih strank in sloni na široki osnovi. ...
Wenn sich am 20. März der Beginn des Irakkrieges zum zweiten Mal jährt, werden über 30000 Iraker im Dienste privater Militärfirmen (PMF) stehen. Hinzu kommen 6000 bis 20000 Ausländer. PMF stellen damit die – nach den US-Streitkräften – zweitstärkste "Armee" im Irak. Die coalition of the willing (Koalition der Willigen) wird immer mehr zu einer coalition of the billing (Koalition der Rechnungsteller). In den letzten zehn Jahren unterzeichnete allein die US-Regierung über 3000 Verträge mit Privatunternehmen, um sich deren Dienstleistungen für ihre im Ausland stationierten Truppen zu sichern. Im Irak vergab die Bush-Regierung, einem Report des Centre for Public Integrity in Washington zufolge, Aufträge mit einem Gesamtvolumen von 48,7 Mrd. US-Dollar an 150 solcher privater Militärdienstleistungsunternehmen. Schon bei der Invasion wurden viele der hoch entwickelten Waffensysteme von Spezialisten bedient (so zum Beispiel Predator-Drohnen, Global Hawks, B-2 Stealth-Bomber). Im Irak obliegt es Mitarbeitern von PMF, Patrouille zu laufen, Gebäude und Infrastruktur zu bewachen und den Personenschutz zu stellen. Selbst das Personal in den irakischen Militärgefängnissen stammt von privaten Sicherheitsdiensten. So waren Mitarbeiter der PMF Caci und Titan in die Foltervorfälle im Abu-Ghraib-Gefängnis verstrickt. Aber auch Privatunternehmen greifen mittlerweile gerne auf die Dienste von PMF zurück. ...

El proceso venezolano de transformación social es ampliamente desconocido en el exterior. Los medios se concentran principalmente en la persona de Chávez ignorando los amplios procesos de cambio y los movimientos emprendidos desde abajo. Las interrogantes que provocan dudas son muchas: ¿Hay realmente un proceso de transformación social en Venezuela? ¿Se está copiando el modelo cubano? ¿Como se puede confiar en Chávez que es un militar? ¿No es Chávez un autoritario? Der vorliegende Aufsatz analysiert die Bildungsreformen, die in Venezuela seit Antritt der Regierung unter Hugo Chávez im Februar 1999 in die Wege geleitet wurden. Nach einem kurzen historischen Überblick bis 1999 folgt eine Erörterung der grundlegenden Linien der Bildungspolitik. Daran schließt die Darstellung des neuen Modells der Escuelas Bolivarianas (Bolivarianische Schulen) an sowie der Misiones (Missionen) genannten Sonderprogramme im Bildungssektor, welche einen Schwerpunkt der Umsetzung des Anspruchs „Bildung für alle“ darstellen: Die Misión Robinson I, II und III für die Alphabetisierung und den Grundschulabschluss und die Misión Ribas für die weiterführende Schule. In den vergangenen Jahren sind in Venezuela, Bolivien und Ecuador Linksregierungen gewählt worden, die eine Wiederverstaatlichung der Erdöl- und Erdgasressourcen ihrer Länder zu ihrem wichtigsten Projekt erklärt haben. Mit dieser Politik verfolgen sie das Ziel, die Kontrolle über die Rohstoffe zurückzugewinnen und die daraus resultierenden Mehreinnahmen für die eigene Entwicklung zu nutzen. Darüber hinaus setzt Venezuela seinen Ölreichtum auch zur lateinamerikanischen Integration ein. Dieses Ansinnen weist durchaus Ähnlichkeiten mit der europäischen Montanunion der 50er Jahre auf. Der Militär Chávez ist der Hoffnungsträger der sozialen Bewegung in Venezuela. Seine Rolle ist aber ambivalent. Einerseits stützt er die zunehmende Macht der Basisarbeit. Anderseits scheint Chávez ihr alleiniger Garant der Beteiligung und schwächt so die Selbstorganisation wieder. Klientelistische Strukturen, wachsende Korruption und das gescheiterte Verfassungsreferendum zeigen die Grenzen des jetzigen Transformationsprozesses auf.
Die neuen Söldner

Bezahlte Erbringer von militärischen Dienstleistungen, Söldner genannt, sind in der Kriegsgeschichte nicht unbekannt. Mit der Erfindung der Privaten Militärunternehmen, der Private Military Contractors (PMC) und Private Security Contractors (PSC) Anfang der 1990er Jahre wurde das Geschäft allerdings in den vermeintlich »sauberen« Wirtschaftsbereich überführt. Somit können die Unternehmen offen rekrutieren und ihre Dienstleistungen auf dem Weltmarkt anbieten. Die Industrie der neuen Söldner hat sich in den vergangenen 20 Jahren weltweit entwickelt und PMC und PSC aus den USA, Großbritannien, Israel, Frankreich, Deutschland, Australien, Russland, Rumänien, Bulgarien und anderen Länder bieten Dienstleistungen jeder Art an. Hoch professionelle Unternehmen aus den USA und Großbritannien stellen jedoch mehr als 70 Prozent der Militärdienstleistungen rund um den Globus zur Verfügung. Sie kommen vor allem im Irak und Afghanistan, aber auch in Kolumbien, Somalia, Sudan und anderen Ländern zum Einsatz. ...
Nach ihrer angeblichen Demobilisierung in Kolumbien im Jahr 2003 begannen die Paramilitärs, massiv nach Venezuela einzusickern. Von den Grenzgebieten aus hatten sie sich zunächst im Gebiet des Andenkorridors im Nordwesten Venezuelas ausgebreitet. Sogar logistische Zentren zur Unterstützung der konterrevolutionären Aktivitäten einschließlich der Anwesenheit ausländischer Spezialisten soll es in der Region geben. El presente ensayo resume y analiza los diferentes esfuerzos organizacionales del gobierno bolivariano de Venezuela de alcanzar una democratización de la propiedad y administración de los medios de producción. Las diferentes iniciativas y medidas adoptadas desde el 2000 – primero reforzar el cooperativismos, luego la creación de Empresas de Producción Social (EPS) y finalmente las Empresas Socialistas, como tambien el conccepto del desarrollo endógeno, orientación normativa de la organización productiva y de los programas estadales de formación laboral, son presentados y analizados con visión crítica. En respecto a la organización interna de lugares de producción se hace un análisis crítico de los modelos de cogestión, autogestión y control obrero mirando los logros y las fallas recurriendo a trabajo de campo hecho en tres fábricas: La fábrica de aluminio Alcasa; la papelera Invepal y la fábrica de válvulas Inveval. Kolumbien ist seit mehr als zwanzig Jahren ein Experimentierfeld für die privatisierte Kriegsführung. Polizei, Armee, Politik, Drogenhandel, Großgrundbesitzer, Paramilitärs, US-Armee, die nordamerikanische Antidrogenagentur DEA, transnationale Konzerne und private militärische Einheiten (PME) kooperieren in wechselnder Besetzung, um ihre jeweiligen Interessen gegen Bauernorganisationen, Gewerkschaften, sozialen Bewegungen und bewaffnete Aufstandsbewegungen durchzusetzen. In Kolumbien sind um die zwanzig PME mit etwa 2000 Angestellten tätig. Die größte PME ist DynCorp, die auch im Irak arbeitet. In Kolumbien ist eine ihrer Dienstleistungen die Besprühung von Coca- und Mohnfeldern. Für die Besprühungen in Kolumbien stehen DynCorp 88 Helikopter und Kleinflugzeuge des Pentagon zur Verfügung. Die Firma gibt die Anzahl ihrer Beschäftigten in Kolumbien nicht bekannt. Geschätzt werden zwischen 100 und 355 Mitarbeiter, von denen weniger als ein Drittel US-Amerikaner sind. Die Einnahmen von DynCorp stiegen von 6,6 Millionen US-Dollar 1996 auf 30 bis 40 Millionen Dollar jährlich seit 1999. ...
In den letzten Jahrzehnten hat vor allem die Frage nach der Übernahme der (Staats-)Macht für Kontroversen innerhalb der Linken gesorgt. Ob der Staat übernommen wird, bis zu einem bestimmten Punkt mit staatlichen Institutionen zusammengearbeitet werden solle oder doch lieber jede Kooperation vermieden werden müsse, war ein zentraler Streitpunkt. Die Wahl verschiedener linker Regierungen in Lateinamerika, vor allem die Fälle Venezuelas und Boliviens, spielen eine zentrale Rolle. Mit der Wahl von Hugo Chávez zum Präsidenten Venezuelas und seiner Amtsübernahme Anfang 1999 begann ein Prozess wirksamer und auf eine sehr breite linke Bewegung gründender sozialer Transformationen, der die Linke zwingt, bestimmte tradierte Konzepte neu zu denken.
Background: The treatment of high-risk neuroblastoma patients consists of multimodal induction therapy to achieve remission followed by consolidation therapy to prevent relapses. However, the type of consolidation therapy is still discussed controversial. We applied metronomic chemotherapy in the prospective NB90 trial and monoclonal anti-GD2-antibody (MAB) ch14.18 in the NB97 trial. Here, we present the long term outcome data of the patient cohort. Methods: A total of 334 stage 4 neuroblastoma patients one year or older were included. All patients successfully completed the induction therapy. In the NB90 trial, 99 patients received at least one cycle of the oral maintenance chemotherapy (NB90 MT, 12 alternating cycles of oral melphalan/etoposide and vincristine/cyclophosphamide). In the NB97 trial, 166 patients commenced the MAB ch14.18 consolidation therapy (six cycles over 12 months). Patients who received no maintenance therapy according to the NB90 protocol or by refusal in NB97 (n = 69) served as controls. Results: The median observation time was 11.11 years. The nine-year event-free survival rates were 41 ± 4%, 31 ± 5%, and 32 ± 6% for MAB ch14.18, NB90 MT, and no consolidation, respectively (p = 0.098). In contrast to earlier reports, MAB ch14.18 treatment improved the long-term outcome compared to no additional therapy (p = 0.038). The overall survival was better in the MAB ch14.18-treated group (9-y-OS 46 ± 4%) compared to NB90 MT (34 ± 5%, p = 0.026) and to no consolidation (35 ± 6%, p = 0.019). Multivariable Cox regression analysis revealed ch14.18 consolidation to improve outcome compared to no consolidation, however, no difference between NB90 MT and MAB ch14.18-treated patients was found. Conclusions: Follow-up analysis of the patient cohort indicated that immunotherapy with MAB ch14.18 may prevent late relapses. Finally, metronomic oral maintenance chemotherapy also appeared effective.
Background: Many cancer patients seek homeopathy as a complementary therapy. It has rarely been studied systematically, whether homeopathic care is of benefit for cancer patients. Methods: We conducted a prospective observational study with cancer patients in two differently treated cohorts: one cohort with patients under complementary homeopathic treatment (HG; n=259), and one cohort with conventionally treated cancer patients (CG; n=380). For a direct comparison, matched pairs with patients of the same tumour entity and comparable prognosis were to be formed. Main outcome parameter: change of quality of life (FACT-G, FACIT-Sp) after 3 months. Secondary outcome parameters: change of quality of life (FACT-G, FACIT-Sp) after a year, as well as impairment by fatigue (MFI) and by anxiety and depression (HADS). Results: HG: FACT-G, or FACIT-Sp, respectively improved statistically significantly in the first three months, from 75.6 (SD 14.6) to 81.1 (SD 16.9), or from 32.1 (SD 8.2) to 34.9 (SD 8.32), respectively. After 12 months, a further increase to 84.1 (SD 15.5) or 35.2 (SD 8.6) was found. Fatigue (MFI) decreased; anxiety and depression (HADS) did not change. CG: FACT-G remained constant in the first three months: 75.3 (SD 17.3) at t0, and 76.6 (SD 16.6) at t1. After 12 months, there was a slight increase to 78.9 (SD 18.1). FACIT-Sp scores improved significantly from t0 (31.0 - SD 8.9) to t1 (32.1 - SD 8.9) and declined again after a year (31.6 - SD 9.4). For fatigue, anxiety, and depression, no relevant changes were found. 120 patients of HG and 206 patients of CG met our criteria for matched-pairs selection. Due to large differences between the two patient populations, however, only 11 matched pairs could be formed. This is not sufficient for a comparative study. Conclusion: In our prospective study, we observed an improvement of quality of life as well as a tendency of fatigue symptoms to decrease in cancer patients under complementary homeopathic treatment. It would take considerably larger samples to find matched pairs suitable for comparison in order to establish a definite causal relation between these effects and homeopathic treatment.
Background: To evaluate the effectivity of fractionated radiotherapy in adolescent and adult patients with pineal parenchymal tumors (PPT). Methods: Between 1982 and 2003, 14 patients with PPTs were treated with fractionated radiotherapy. 4 patients had a pineocytoma (PC), one a PPT with intermediate differentiation (PPTID) and 9 patients a pineoblastoma (PB), 2 of which were recurrences. All patients underwent radiotherapy to the primary tumor site with a median total dose of 54 Gy. In 9 patients with primary PB treatment included whole brain irradiation (3 patients) or irradiation of the craniospinal axis (6 patients) with a median total dose of 35 Gy. Results: Median follow-up was 123 months in the PC patients and 109 months in the patients with primary PB. 7 patients were free from relapse at the end of follow-up. One PC patient died from spinal seeding. Among 5 PB patients treated with radiotherapy without chemotherapy, 3 developed local or spinal tumor recurrence. Both patients treated for PB recurrences died. The patient with PPTID is free of disease 7 years after radiotherapy. Conclusion: Local radiotherapy seems to be effective in patients with PC and some PPTIDs. Diagnosis and treatment of patients with more aggressive variants of PPTIDs as well as treatment of PB need to be further improved, since local and spinal failure even despite craniospinal irradiation (CSI) is common. As PPT are very rare tumors, treatment within multi-institutional trials remains necessary.
Background: It has been demonstrated that cognitive behavioural therapy (CBT) has a moderate effect on symptom reduction and on general well being of patients suffering from psychosis. However, questions regarding the specific efficacy of CBT, the treatment safety, the cost-effectiveness, and the moderators and mediators of treatment effects are still a major issue. The major objective of this trial is to investigate whether CBT is specifically efficacious in reducing positive symptoms when compared with non-specific supportive therapy (ST) which does not implement CBT-techniques but provides comparable therapeutic attention. Methods: The POSITIVE study is a multicenter, prospective, single-blind, parallel group, randomised clinical trial, comparing CBT and ST with respect to the efficacy in reducing positive symptoms in psychotic disorders. CBT as well as ST consist of 20 sessions altogether, 165 participants receiving CBT and 165 participants receiving ST. Major methodological aspects of the study are systematic recruitment, explicit inclusion criteria, reliability checks of assessments with control for rater shift, analysis by intention to treat, data management using remote data entry, measures of quality assurance (e.g. on-site monitoring with source data verification, regular query process), advanced statistical analysis, manualized treatment, checks of adherence and competence of therapists. Research relating the psychotherapy process with outcome, neurobiological research addressing basic questions of delusion formation using fMRI and neuropsychological assessment and treatment research investigating adaptations of CBT for adolescents is combined in this network. Problems of transfer into routine clinical care will be identified and addressed by a project focusing on cost efficiency. Discussion: This clinical trial is part of efforts to intensify psychotherapy research in the field of psychosis in Germany, to contribute to the international discussion on psychotherapy in psychotic disorders, and to help implement psychotherapy in routine care. Hepatitis C virus (HCV) naturally infects only humans and chimpanzees. The determinants responsible for this narrow species tropism are not well defined. Virus cell entry involves human scavenger receptor class B type I (SR-BI), CD81, claudin-1 and occludin. Among these, at least CD81 and occludin are utilized in a highly species-specific fashion, thus contributing to the narrow host range of HCV. We adapted HCV to mouse CD81 and identified three envelope glycoprotein mutations which together enhance infection of cells with mouse or other rodent receptors approximately 100-fold. These mutations enhanced interaction with human CD81 and increased exposure of the binding site for CD81 on the surface of virus particles. These changes were accompanied by augmented susceptibility of adapted HCV to neutralization by E2-specific antibodies indicative of major conformational changes of virus-resident E1/E2-complexes. Neutralization with CD81, SR-BI- and claudin-1-specific antibodies and knock down of occludin expression by siRNAs indicate that the adapted virus remains dependent on these host factors but apparently utilizes CD81, SR-BI and occludin with increased efficiency. Importantly, adapted E1/E2 complexes mediate HCV cell entry into mouse cells in the absence of human entry factors. These results further our knowledge of HCV receptor interactions and indicate that three glycoprotein mutations are sufficient to overcome the species-specific restriction of HCV cell entry into mouse cells. Moreover, these findings should contribute to the development of an immunocompetent small animal model fully permissive to HCV.
HDL, through sphingosine-1-phosphate (S1P), exerts direct cardioprotective effects on ischemic myocardium. It remains unclear whether other HDL-associated sphingophospholipids have similar effects. We therefore examined if HDL-associated sphingosylphosphorylcholine (SPC) reduces infarct size in a mouse model of transient myocardial ischemia/reperfusion. Intravenously administered SPC dose-dependently reduced infarct size after 30 minutes of myocardial ischemia and 24 hours reperfusion compared to controls. Infarct size was also reduced by postischemic, therapeutical administration of SPC. Immunohistochemistry revealed reduced polymorphonuclear neutrophil recruitment to the infarcted area after SPC treatment, and apoptosis was attenuated as measured by TUNEL. In vitro, SPC inhibited leukocyte adhesion to TNFα-activated endothelial cells and protected rat neonatal cardiomyocytes from apoptosis. S1P3 was identified as the lysophospholipid receptor mediating the cardioprotection by SPC, since its effect was completely absent in S1P3-deficient mice. We conclude that HDL-associated SPC directly protects against myocardial reperfusion injury in vivo via the S1P3 receptor.
Snake bite is one of the most neglected public health issues in poor rural communities living in the tropics. Because of serious misreporting, the true worldwide burden of snake bite is not known. South Asia is the world's most heavily affected region, due to its high population density, widespread agricultural activities, numerous venomous snake species and lack of functional snake bite control programs. Despite increasing knowledge of snake venoms' composition and mode of action, good understanding of clinical features of envenoming and sufficient production of antivenom by Indian manufacturers, snake bite management remains unsatisfactory in this region. Field diagnostic tests for snake species identification do not exist and treatment mainly relies on the administration of antivenoms that do not cover all of the important venomous snakes of the region. Care-givers need better training and supervision, and national guidelines should be fed by evidence-based data generated by well-designed research studies. Poorly informed rural populations often apply inappropriate first-aid measures and vital time is lost before the victim is transported to a treatment centre, where cost of treatment can constitute an additional hurdle. The deficiency of snake bite management in South Asia is multi-causal and requires joint collaborative efforts from researchers, antivenom manufacturers, policy makers, public health authorities and international funders.
Piracetam, the prototype of the so-called nootropic drugs’ is used since many years in different countries to treat cognitive impairment in aging and dementia. Findings that piracetam enhances fluidity of brain mitochondrial membranes led to the hypothesis that piracetam might improve mitochondrial function, e.g., might enhance ATP synthesis. This assumption has recently been supported by a number of observations showing enhanced mitochondrial membrane potential, enhanced ATP production, and reduced sensitivity for apoptosis in a variety of cell and animal models for aging and Alzheimer disease. As a specific consequence, substantial evidence for elevated neuronal plasticity as a specific effect of piracetam has emerged. Taken together, this new findings can explain many of the therapeutic effects of piracetam on cognition in aging and dementia as well as different situations of brain dysfunctions. Leukotrienes constitute a group of bioactive lipids generated by the 5-lipoxygenase (5-LO) pathway. An increasing body of evidence supports an acute role for 5-LO products already during the earliest stages of pancreatic, prostate, and colorectal carcinogenesis. Several pieces of experimental data form the basis for this hypothesis and suggest a correlation between 5-LO expression and tumor cell viability. First, several independent studies documented an overexpression of 5-LO in primary tumor cells as well as in established cancer cell lines. Second, addition of 5-LO products to cultured tumor cells also led to increased cell proliferation and activation of anti-apoptotic signaling pathways. 5-LO antisense technology approaches demonstrated impaired tumor cell growth due to reduction of 5-LO expression. Lastly, pharmacological inhibition of 5-LO potently suppressed tumor cell growth by inducing cell cycle arrest and triggering cell death via the intrinsic apoptotic pathway. However, the documented strong cytotoxic off-target effects of 5-LO inhibitors, in combination with the relatively high concentrations of 5-LO products needed to achieve mitogenic effects in cell culture assays, raise concern over the assignment of the cause, and question the relationship between 5-LO products and tumorigenesis. Introduction: The Vbeta12-transgenic mouse was previously generated to investigate the role of antigen-specific T cells in collagen-induced arthritis (CIA), an animal model for rheumatoid arthritis. This mouse expresses a transgenic collagen type II (CII)-specific T-cell receptor (TCR) beta-chain and consequently displays an increased immunity to CII and increased susceptibility to CIA. However, while the transgenic Vbeta12 chain recombines with endogenous alpha-chains, the frequency and distribution of CII-specific T cells in the Vbeta12-transgenic mouse has not been determined. The aim of the present report was to establish a system enabling identification of CII-specific T cells in the Vbeta12-transgenic mouse in order to determine to what extent the transgenic expression of the CII-specific beta-chain would skew the response towards the immunodominant galactosylated T-cell epitope and to use this system to monitor these cells throughout development of CIA. Methods: We have generated and thoroughly characterized a clonotypic antibody, which recognizes a TCR specific for the galactosylated CII(260-270) peptide in the Vbeta12-transgenic mouse. Hereby, CII-specific T cells could be quantified and followed throughout development of CIA, and their phenotype was determined by combinatorial analysis with the early activation marker CD154 (CD40L) and production of cytokines. Results: The Vbeta12-transgenic mouse expresses several related but distinct T-cell clones specific for the galactosylated CII peptide. The clonotypic antibody could specifically recognize the majority (80%) of these. Clonotypic T cells occurred at low levels in the naïve mouse, but rapidly expanded to around 4% of the CD4+ T cells, whereupon the frequency declined with developing disease. Analysis of the cytokine profile revealed an early Th1-biased response in the draining lymph nodes that would shift to also include Th17 around the onset of arthritis. Data showed that Th1 and Th17 constitute a minority among the CII-specific population, however, indicating that additional subpopulations of antigen-specific T cells regulate the development of CIA. Conclusions: The established system enables the detection and detailed phenotyping of T cells specific for the galactosylated CII peptide and constitutes a powerful tool for analysis of the importance of these cells and their effector functions throughout the different phases of arthritis.

Background: The immune system is a complex adaptive system of cells and molecules that are interwoven in a highly organized communication network. Primary immune deficiencies are disorders in which essential parts of the immune system are absent or do not function according to plan. X-linked agammaglobulinemia is a B-lymphocyte maturation disorder in which the production of immunoglobulin is prohibited by a genetic defect. Patients have to be put on life-long immunoglobulin substitution therapy in order to prevent recurrent and persistent opportunistic infections. Methodology: We formulate an immune response model in terms of stochastic differential equations and perform a systematic analysis of empirical therapy protocols that differ in the treatment frequency. The model accounts for the immunoglobulin reduction by natural degradation and by antigenic consumption, as well as for the periodic immunoglobulin replenishment that gives rise to an inhomogeneous distribution of immunoglobulin specificities in the shape space. Results are obtained from computer simulations and from analytical calculations within the framework of the Fokker-Planck formalism, which enables us to derive closed expressions for undetermined model parameters such as the infection clearance rate. Conclusions: We find that the critical value of the clearance rate, below which a chronic infection develops, is strongly dependent on the strength of fluctuations in the administered immunoglobulin dose per treatment and is an increasing function of the treatment frequency. The comparative analysis of therapy protocols with regard to the treatment frequency yields quantitative predictions of therapeutic relevance, where the choice of the optimal treatment frequency reveals a conflict of competing interests: In order to diminish immunomodulatory effects and to make good economic sense, therapeutic immunoglobulin levels should be kept close to physiological levels, implying high treatment frequencies. However, clearing infections without additional medication is more reliably achieved by substitution therapies with low treatment frequencies. Our immune response model predicts that the compromise solution of immunoglobulin substitution therapy has a treatment frequency in the range from one infusion per week to one infusion per two weeks.
Vielleicht gibt es irgendwann Wanderungsbewegungen in den Weltraum. Je später die zukünftigen Generationen leben, umso geringer ist unser Wissen um ihre Lebenssituation, ihre Möglichkeiten und ihre daran wohl angepassten Interessen. Dass unser Lebensstil und Konsum auf Dauer das Klima bedrohen, die Vielfalt der Arten reduzieren, das Trinkwasser immer knapper werden lassen, ist der Bevölkerung durchaus bewusst. Doch nur eine Minderheit ist bereit, im Alltag auf das Auto zu verzichten, für Urlaubsreisen nicht das Flugzeug zu benutzen, ökologische Lebensmittel zu kaufen oder beim Kauf von Möbeln auf das Zertifikat »nachhaltige Holzwirtschaft« zu setzen. Wie lässt sich der Widerspruch zwischenWissen um den eigenen Ressourcenverbrauch und Sorge um die Umwelt einerseits und wenig nachhaltigem Konsumverhalten andererseits erklären?
Dass Ludwig Börne seine langjährige vertraute Freundin Jeanette Wohl in einem seiner Briefe scherzhaft mit »Moppel« ansprach, brachte erst eine Spezialaufnahme ans Licht, denn die Stelle war eigentlich für die Nachwelt geschwärzt geworden. Immer mehr Dokumente stehen auch in mehrschichtigen Aufnahmen für die wissenschaftliche Analyse zur Verfügung. Bald werden die bekanntesten schriftlichen Quellen vom Altertum bis zur Gegenwart nachhaltig erfasst und gespeichert und somit digital und online aufrufbar sein. Wie nutzen die Frankfurter Geisteswissenschaftler diese neuen fast grenzenlosen Chancen? Im LOEWE-Schwerpunkt »Digital Humanities« geht es darum, Textcorpora mit digitalen Methoden auszuwerten, zu vergleichen und mit Bilddaten zu verknüpfen.
Die Sonne strahlt weltweit pro Tag genügend Licht ein, um den Weltenergiebedarf für ein ganzes Jahr abzudecken. Somit ist sie die Quelle aller erneuerbarer Energien, denn neben der Erzeugung von Elektrizität aus Licht (Photovoltaik) regelt sie die Gezeiten und damit auch Wind und Wellen, die bei der Windkraft und in Gezeitenkraftwerken genutzt werden. Außerdem liefert sie die Energie für die Photosynthese in nachwachsenden Rohstoffen. Es gibt diesbezüglich nur ein grundlegendes Problem: Erneuerbare Energien fi nden wir in ausreichender Menge vor allem an Stellen mit mangelnder Infrastruktur. Sonnenenergie gibt es am meisten in der Wüste, Wind auf dem Meer und Biomasse im Dschungel. An Orten hoher Industrialisierung und damit auch hoher Bevölkerungsdichte ist für die »Erneuerbaren « so gut wie kein Platz. Es gibt demnach kein Energieproblem, aber ein Problem der Energiespeicherung und des Energietransportes.
